    Reduced Fas ligand-expressing splenic CD5+ B lymphocytes in severe collagen-induced arthritis

    Abstract Introduction The objective was to study immune regulation in a mouse model of rheumatoid arthritis that exhibits considerable heterogeneity of disease activity. Methods T-cell receptor transgenic mice, in which nearly all CD4+ T cells recognize a single peptide of type II collagen, were immunized with collagen and observed for development of arthritis for 4 weeks. At 28 days post-immunization, splenocytes were analyzed by flow cytometry and in vitro assays for markers of immune activation and regulation. Results Disease severities ranging from 0 to 12 (on a 12-point scale) were observed. Among splenic lymphocyte populations, only the CD5+ B-cell subset displayed a decrease in relative numbers as arthritis severity increased. Splenic CD5+ B cells expressed higher levels of Fas ligand (FasL) than did CD4+ T cells or CD5- B cells in all mice, and antigen-dependent T-cell death correlated with higher levels of CD5+ B cells in cocultures. Ratios of interleukin (IL)-17 to interferon-gamma production were higher in antigen-driven cultures of splenocytes from severely arthritic mice compared to mildly or nonarthritic mice. A correlation was established between the reduced production of IL-17 in antigen-driven T-cell/B-cell cocultures and FasL, but not IL-10. Confirmation of the direct killing effect of B cells on T cells was demonstrated using an antigen-specific T hybridoma cell line. Conclusions Reduced numbers of splenic FasL+ CD5+ B cells correlated with increasing arthritis severity and decreased T-cell death in a T-cell receptor transgenic mouse model of collagen-induced arthritis.     Framing Biotechnology: A Comparison of U.S. and British National Newspapers

    Despite the potential promise of agricultural biotechnology, consumers’ trust and acceptance varies in some parts of the world. To develop a deeper understanding of the differences in news media coverage of agricultural biotechnology in U.S. and British newspapers, a framing analysis study was conducted, focusing on the news coverage of three major national “newspapers of record” during the calendar year 2002: The New York Times, The Washington Post, and The Guardian. No single frame was dominant in U.S. or British national newspaper coverage of agricultural biotechnology in 2002. Some of the prominent frames were contamination of the food supply, human risk, environmental risk, scientific progress, and world hunger. There were more different frames used in The Guardian (concepts as consumer choice, dependency, and politics) than in the U.S. newspapers. Additionally, this study reiterated findings that British news coverage included more editorial coverage of agricultural biotechnology

    Comparing Linkages Between Descriptive Norms and Current and Intended Outdoor Water Conservation

    The study presented here was used for evaluating how theory of planned behavior variables (attitude, perceived behavioral control, subjective norms) and perceptions of others\u27 outdoor water conservation (descriptive norms) related to individuals\u27 own conservation and intent to conserve. The theory of planned behavior predicted current and intended conservation similarly. Descriptive norms from close-peer and state referent groups improved predictions of current practices but not behavioral intentions. Descriptive norms may be more influential when Extension clients are establishing routines pertaining to outdoor water conservation (e.g., installing a new landscape). Extension professionals should strategically communicate a conservation norm at similar opportune times

    A Thorough Analysis and Categorization of Bacterial Interrupted Adenylation Domains, Including Previously Unidentified Families

    Interrupted adenylation (A) domains are key to the immense structural diversity seen in the nonribosomal peptide (NRP) class of natural products (NPs). Interrupted A domains are A domains that contain within them the catalytic portion of another domain, most commonly a methylation (M) domain. It has been well documented that methylation events occur with extreme specificity on either the backbone (N-) or side chain (O- or S-) of the amino acid (or amino acid-like) building blocks of NRPs. Here, through taxonomic and phylogenetic analyses as well as multiple sequence alignments, we evaluated the similarities and differences between interrupted A domains. We probed their taxonomic distribution amongst bacterial organisms, their evolutionary relatedness, and described conserved motifs of each type of M domain found to be embedded in interrupted A domains. Additionally, we categorized interrupted A domains and the M domains within them into a total of seven distinct families and six different types, respectively. The families of interrupted A domains include two new families, 6 and 7, that possess new architectures. Rather than being interrupted between the previously described a2–a3 or a8–a9 of the ten conserved A domain sequence motifs (a1–a10), family 6 contains an M domain between a6–a7, a previously unknown interruption site. Family 7 demonstrates that di-interrupted A domains exist in Nature, containing an M domain between a2–a3 as well as one between a6–a7, displaying a novel arrangement. These in-depth investigations of amino acid sequences deposited in the NCBI database highlighted the prevalence of interrupted A domains in bacterial organisms, with each family of interrupted A domains having a different taxonomic distribution. They also emphasized the importance of utilizing a broad range of bacteria for NP discovery. Categorization of the families of interrupted A domains and types of M domains allowed for a better understanding of the trends of naturally occurring interrupted A domains, which illuminated patterns and insights on how to harness them for future engineering studies

    Towards low-latency real-time detection of gravitational waves from compact binary coalescences in the era of advanced detectors

    Electromagnetic (EM) follow-up observations of gravitational wave (GW) events will help shed light on the nature of the sources, and more can be learned if the EM follow-ups can start as soon as the GW event becomes observable. In this paper, we propose a computationally efficient time-domain algorithm capable of detecting gravitational waves (GWs) from coalescing binaries of compact objects with nearly zero time delay. In case when the signal is strong enough, our algorithm also has the flexibility to trigger EM observation before the merger. The key to the efficiency of our algorithm arises from the use of chains of so-called Infinite Impulse Response (IIR) filters, which filter time-series data recursively. Computational cost is further reduced by a template interpolation technique that requires filtering to be done only for a much coarser template bank than otherwise required to sufficiently recover optimal signal-to-noise ratio. Towards future detectors with sensitivity extending to lower frequencies, our algorithm's computational cost is shown to increase rather insignificantly compared to the conventional time-domain correlation method. Moreover, at latencies of less than hundreds to thousands of seconds, this method is expected to be computationally more efficient than the straightforward frequency-domain method.Comment: 19 pages, 6 figures, for PR
