    Ações da guanosina em ratos wistar fêmeas e machos induzidos a isquemia cerebral

    A isquemia cerebral é uma enfermidade grave, de incidência e prevalência global, considerada uma das principais causas de morte e incapacidade no mundo. Ela é decorrente da hipoperfusão ou interrupção sanguínea em uma determinada região encefálica e a falta de oxigênio e glicose leva a falha no metabolismo energético de neurônios e células gliais. A morte destas células libera grande quantidade de glutamato, o principal neurotransmissor excitatório, o que desencadeia a cascata isquêmica, formada por estresse oxidativo, excitotoxicidade e processo inflamatório. Estes insultos causam danos teciduais graves que comprometem, principalmente, o sistema sensorimotor, memória, cognição e as emoções. A guanosina, um derivado da guanina, tem mostrado papel neuroprotetor ao sistema nervoso central, agindo no sistema glutamatérgico. Esses efeitos têm sido demonstrados em ratos machos, mas não em fêmeas, por isso, o objetivo desta tese foi investigar através de testes comportamentais, imunoistoquímico e histológico, a ação da guanosina em ratos Wistar machos e fêmeas induzidos cirurgicamente a um modelo de isquemia focal permanente do córtex cerebral parietal, levando em consideração o ciclo estral das fêmeas imediatamente antes da indução isquêmica. Este trabalho foi aprovado pela Comissão de Ética no Uso de Animais da UFRGS (Nº 29396). Após a verificação do ciclo estral, fêmeas e machos foram anestesiados, posicionados em aparelho estereotáxico e a isquemia focal cortical foi induzida por termocoagulação dos vasos piais. Ao término da cirurgia, os animais foram tratados com solução salina 0,9% ou guanosina 60 mg/kg, ambos por via intraperitoneal, em 4 doses (0, 1, 3 e 6 horas após a indução isquêmica). No teste do cilindro, as fêmeas apresentaram significativamente melhor recuperação sensorimotora a longo prazo comparadas aos machos. No teste do campo aberto, memória de habituação a longo prazo e locomoção em fêmeas foram prejudicadas pela isquemia e os machos não apresentaram prejuízo. No teste do labirinto em cruz elevado não houve diferença significativa entre os grupos. No teste claro/escuro, as fêmeas isquêmicas demonstraram comportamento ansiolítico comparadas aos machos e às fêmeas naïves. A imunoistoquímica de astrócitos e a mensuração do volume de lesão não apresentaram diferença estatística entre os grupos. Com este trabalho demonstrou-se a importância da realização de pesquisas em ambos os gêneros, considerando o ciclo estral, principalmente para enfermidades complexas como a isquemia cerebral. O tratamento com a guanosina é promissor, principalmente na recuperação motora em fêmeas, entretanto, estudos futuros são necessários para melhor compreendimento dos mecanismos envolvidos.Ischemic stroke is a serious disease of global incidence and prevalence, considered a leading cause of death and disability worldwide. It is due to hypoperfusion or blood disruption in a brain region and the lack of oxygen and glucose leads to failure in energetic metabolism of neurons and glial cells. The death of these cells releases large amount of glutamate, the main excitatory neurotransmitter, which triggers the ischemic cascade, formed by oxidative stress, excitotoxicity and inflammatory processes. These insults cause severe tissue damage that mainly compromise the sensorimotor system, memory, cognition and emotions. Guanosine, a guanine derivative, has shown a neuroprotective role in the central nervous system, acting on the glutamatergic system. These effects have been demonstrated in male rats, but not in females, so the aim of this thesis was to investigate through behavioral, immunohistochemical and histological tests, the action of guanosine in male and female Wistar rats surgically induced to a model of focal permanent cerebral ischemia in the motor cortex, considering the female estrous cycle immediately before the ischemic induction. This work was approved by the UFRGS Ethical Committee on the Use of Animals (No. 29396). After estrous cycle verification, females and males were anesthetized, positioned in a stereotaxic apparatus, and cortical focal ischemia was induced by thermocoagulation of the pial vessels. At the end of the surgery, the animals were treated with 0.9% saline or guanosine 60 mg/kg, both intraperitoneally, in 4 doses (0, 1, 3 and 6 hours after ischemic induction). In the cylinder test, females presented significant long-term sensorimotor recovery compared to males. In the open field test, long-term memory habituation and locomotion/exploratory activity in females were impaired by ischemia and males showed no impairment. In the elevated plus maze test, there was no significant difference between groups. In the light/dark test, the ischemic females showed anxiolytic-like behavior compared to naïve males and females. The immunohistochemistry of astrocytes and the measurement of lesion volume did not show any statistical difference between groups. With this work it was demonstrated the importance of conducting researches in both genders, considering the estrous cycle, mainly for complex diseases such as cerebral ischemia. The treatment with guanosine is promising, especially in motor recovery in females; however, future studies are necessary to better understand the mechanisms involved

    Functional recovery caused by human adipose tissue mesenchymal stem cell-derived extracellular vesicles administered 24 h after stroke in rats

    Ischemic stroke is a major cause of death and disability, intensely demanding innovative and accessible therapeutic strategies. Approaches presenting a prolonged period for therapeutic intervention and new treatment administration routes are promising tools for stroke treatment. Here, we evaluated the potential neuroprotective properties of nasally administered human adipose tissue mesenchymal stem cell (hAT-MSC)-derived extracellular vesicles (EVs) obtained from healthy individuals who underwent liposuction. After a single intranasal EV (200 µg/kg) administered 24 h after a focal permanent ischemic stroke in rats, a higher number of EVs, improvement of the blood–brain barrier, and re-stabilization of vascularization were observed in the recoverable peri-infarct zone, as well as a significant decrease in infarct volume. In addition, EV treatment recovered long-term motor (front paws symmetry) and behavioral impairment (short- and long-term memory and anxiety-like behavior) induced by ischemic stroke. In line with these findings, our work highlights hAT-MSC-derived EVs as a promising therapeutic strategy for stroke

    Biochemical and immunocytochemical tumor markers in canine neoplastic effusions

    As efusões cavitárias são de ocorrência frequente na rotina clínica de cães. Na maior parte dos casos são efusões benignas, causadas por distúrbios hidrostáticos do sistema circulatório. As neoplasias são causas comuns de efusões em cães, contudo, nem sempre as células tumorais são encontradas no exame citopatológico. A dosagem de marcadores tumorais e o exame imunocitoquímico são alternativas para tornar o diagnóstico de neoplasia em efusões mais preciso. Os objetivos deste trabalho foram dosar os seguintes marcadores tumorais bioquímicos: antígeno carcinoembrionário (CEA), antígeno associado a câncer 72-4 (CA 72-4) e fragmento de citoqueratina 21-1 (CYFRA 21-1), que ainda não tiveram seu desempenho avaliado em efusões neoplásicas e não neoplásicas caninas, bem como marcadores imunocitoquímicos que incluem dois novos anticorpos primários (MOC-31 e D2-40) para a diferenciação entre carcinoma e mesotelioma. Trinta e duas amostras de líquidos cavitários abdominais e torácicos, provenientes do atendimento clínico do Hospital de Clínicas Veterinárias da Universidade Federal do Rio Grande do Sul foram analisadas. De acordo com o exame citopatológico e ficha clínica do animal essas efusões foram classificadas em dois grupos: neoplásico e não neoplásico. A dosagem dos marcadores tumorais foi realizada pelo método imunoenzimático sanduíche (ELISA), conforme as instruções dos fabricantes. Para a avaliação imunocitoquímica foram utilizadas 14 amostras de efusões neoplásicas. A técnica foi realizada pelo método estreptavidina-biotina ligada a peroxidase ou a fosfatase alcalina, utilizando-se como cromógeno o DAB. Os marcadores tumorais CEA e CA 72-4 não tiveram resultados significativos na diferenciação entre efusões neoplásicas e não neoplásicas, enquanto que o CYFRA 21-1 obteve sensibilidade de 70%, especificidade de 94% e acurácia de 81% para o diagnóstico neoplásico. Em todas as amostras neoplásicas, imunocitoquímica e citopatologia foram compatíveis, verificando-se como válida a padronização dos novos anticorpos para a espécie canina. Este estudo demonstrou que novos marcadores tumorais, tanto bioquímicos como imunocitoquímicos, podem ser empregados no diagnóstico de neoplasias caninas. O marcador tumoral CYFRA 21-1 deve ser utilizado como auxílio diagnóstico para a espécie canina e os anticorpos MOC-31 e D2-40 devem ser incluídos em painéis imunocitoquímicos de rotina para a diferenciação entre carcinomas e mesoteliomas em efusões neoplásicas.The cavity effusions frequently occur in the clinical routine of dogs. In most cases the effusions are benign caused by circulatory system disorders. Neoplasms are common causes of effusions in dogs, however not always the tumor cells are found in cytopathologycal analysis. The dosage of tumor markers is an alternative to make the neoplastic effusion diagnosis more accurate. The aims of this work were to determine the following biochemical tumor markers: carcinoembryonic antigen (CEA), cancer antigen 72-4 (CA 72-4) and cytokeratin fragment 21-1 (CYFRA 21-1), which have not had their performance evaluated in canine neoplastic and non-neoplastic effusions, as well as immunocytochemical markers including two new primary antibodies (MOC-31 and D2-40) for differentiation between carcinoma and mesothelioma tumors. Thrirtytwo samples of abdominal and thoracic cavity fluids, from the clinical care of the Veterinary Hospital of the Federal University of Rio Grande do Sul were analyzed. According to the cytopathology test and patient’s clinical record the effusions were classified in two groups: neoplastic or non-neoplastic. The tumor markers measurement was performed by sandwich enzyme immunoassay (ELISA) according to the manufacturer instructions. Fourteen neoplastic samples were used for the immunocytochemical tests. The tests were performed by streptavidin-biotin method linked to peroxidase or to alkaline phosphatase using the DAB chromogen. The tumor markers CEA and CA 72-4 had no significant results for differentiating between neoplastic and non neoplastic effusions, whereas the tumor marker CYFRA 21-1 obtained 70% of sensibility, 94% of specificity and 81% of accuracy for the neoplastic diagnosis. In all neoplastic samples the immunocytochemical and cytological tests were compatible, which make valid their use for standardization of those new antibodies for the canine species. This study demonstrated that new tumor markers both biochemical and immunocytochemical could be used in the canine neoplastic diagnosis. The tumor marker CYFRA 21-1 must be used for the canine species, and the antibodies MOC-31 and D2-40 must be included in routine immunocytochemistry panel for the differenciation between carcinoma and mesothelioma in neoplastic effusions

    Estudo clínico de osteossarcoma canino

    Background:  :  :  : The canine osteosarcoma is characterized by being an aggressive bone tumor that affects mainly large dogs, middle-aged and male, accounting for 80-85% of bone tumors and 5-6% of all tumors. The risk of developing this tumor is, on average, 125 times greater in dogs weighing over 36 kg than in dogs weighing less than 9 kg. This cancer develops more frequently in the appendicular skeleton (75%), preferably the bones of the forelimbs, where approximately 60% of body weight are sustained. The canine osteosarcoma is very similar to human, in which the metaphyseal region of long bones is the most affected, causing lameness and swelling of the affected limb. The diagnosis is made from epidemiological data, history of the animal, X-rays and laboratory tests such as cytology fine-needle aspiration (FNAC) and measurement of the enzyme alkaline phosphatase (ALP). The staging of the disease becomes a major method for the treatment, whereas primary bone tumors are highly complex and end up interfering with the animal organism in whole. The objective of this study is to report a clinical study of 12 dogs with osteosarcoma, showing the procedures performed from diagnosis to treatment. Material, Methods and Results: Dogs were assisted at the Clinic of Small Animals VUH - UFSM and after the clinical examination were referred to the Department of Radiology. After the radiographic study, blood samples for biochemical and material for cytological analysis of the tumor were collected. After the confirmation of the neoplasm, the physician who prescribed the treatment, depending on the case, the accession to it was left to the owner. In this study, we observed the development of osteosarcoma in dogs from two to 14 years old. Furthermore, in 10 animals (83.3%) the tumor was located in one of the members and nine of these (75%) affected forelimbs. Still, eight dogs (66.7%) were males. The animals weighed on average, 30 kg and five were crossbreeds. Eight dogs (66.7%) had an elevation in enzyme ALP. In this study, five dogs (41%) received no treatment, four (33.3%) were killed and three (25.7%) had the treatment as prescribed by the clinician. Three of the dogs treated survived for a few weeks and two of those who received appropriate treatment survived more than a year. Discussion: The clinical and epidemiological findings of this study corroborate the literature, making them increasingly used in the auxiliary tests cited. For a proper diagnosis it is necessary for the veterinary practitioner to know which tests to determine use. Approximately 98% of the patients have micrometastases at diagnosis, however, only 5% of dogs with osteosarcoma have radiographic evidence of pulmonary metastasis. These will only cause clinical signs in advanced stages, leading the majority of dogs to euthanasia. Moreover, the appearance of bone thinning and explosion in the limb are strong indications of osteosarcoma. The FNAC is one of the main tests for diagnosing neoplastic, because the laboratory technician can visualize the tumor cells in proliferation and other features visible only under a microscope. The enzyme alkaline phosphatase is of great value in the diagnosis and prognosis of this tumor and may demonstrate metastases when elevated. Epidemiological data along with auxiliary tests make the diagnosis of this neoplasm safer, which assists the owner in the decision for the treatment of the affected animals. As there is still little understanding of the anticancer treatment by the owners, treatment schedules may not be implemented, which prolongs the disease and causes more suffering to the dogs

    Aspectos epidemiológicos, clínicos e imuno-histoquímicos do linfoma canino na região de Porto Alegre

    This paper describes the epidemiological, clinical and immunohistochemical characteristics of canine lymphomas diagnosed in the region of Porto Alegre, Brazil. Thirty dogs were enrolled in the study; most of them were male (60%), mixed-breed (23%) and middle- aged or older. The majority (87%) of affected dogs showed the multicentric form. The B-cell phenotype was most frequently detected (62%); 37% of the animals were in clinical stage IV, and 83% were classified as sub-stage “b”. Lymphadenopathy was observed in 67% of the cases, and dyspnea, prostration, decreased appetite and vomiting were the most common clinical signs encountered. Anemia was a frequently encountered laboratory alteration (57%), as were leukocytosis (40%), thrombocytopenia (33%), lymphopenia (30%), hyperglobulinemia (20%) and hypercalcemia (13%). The results of this study indicate that the clinical features of dogs with lymphoma in the region of Porto Alegre are similar to those observed worldwide.Esse trabalho apresenta os achados epidemiológicos, clínicos e imuno-histoquímicos de linfomas caninos diagnosticados na região de Porto Alegre. Trinta cães foram incluídos no estudo; sendo principalmente machos (60%), sem raça definida (23%) e de meia-idade a idosos. A maioria (87%) dos cães afetados apresentou a forma anatômica multicêntrica. O imunofenótipo B foi detectado com mais frequência (62%); 37% dos cães apresentavam estadiamento clínico IV e 83% encontravam-se no subestádio “b”. Linfadenopatia foi observada em 67% dos casos; outros sinais clínicos comumente detectados foram dispneia, prostração, diminuição do apetite e vômitos. Anemia foi a alteração laboratorial mais frequente (57%), seguida por leucocitose (40%), trombocitopenia (33%), linfopenia (30%), hiperglobulinemia (20%), hiperproteinemia (17%) e hipercalcemia (13%). Os resultados do presente estudo indicam que as características epidemiológicas e clínicas de cães com linfoma na região de Porto Alegre são semelhantes às observadas em todo o mundo
