    A Proposal to Expand the Community of Users Able to Process Historical Rainfall Data by Means of the Today Available Open Source Libraries

    The paper presents a software architecture based on open source technologies, implemented by the authors in an experience of processing spatio-temporal data gathered by rain gauges spread across two regions of central Italy. The interest in the automatic processing of data about precipitation is widespread, however, today only an inner circle of stakeholders can think of taking advantage of the available open source libraries because a strong programming skill is required to use them. It is opinion of the authors that the implemented software architecture is suitable for expanding the community of users able to process “by themselves” historical precipitation data. The centre of the architecture is the technology of the spatial database management systems. They offer full support for the creation and management of a spatial database suitable to store the rainfall data usually spread out in several files. Moreover, they allow adding to such a repository a large set of ad hoc “objects” oriented to carrying out spatio-temporal computations on the precipitation. The stakeholders are only required to familiarize with the database’s objects and invoke their execution. A large part of the paper is devoted to show how the adopted conceptual setting can assist nontechnical users in carrying out personalized computations on rain gauges data. The computing needs posed by the experience described in this paper are common to many other areas of high social impact that involve spatio-temporal data, hence we believe that the implemented framework can be exported to them, keeping unaltered operational effectiveness

    Another point of view about the expression of p16 and Ki67 in melanocytic and non-melanocytic cutaneous lesions

    The new analysis on the expression of cell cycle regulators, (used in various neoplasms) and the nominal immunohistological essays, still  represent valid and feasible diagnostic methods in most pathology practice. We examined 114 paraffin-embedded histological specimens of melanocytic cutaneous lesions. The primary objective of this study was to explore the potential diagnostic of  two important cell cycle regulators (p16 and Ki67) evaluating also the variations of expression using a semi quantitative graded scale. While, another aim  was to study an hypothetical correlation between p16 expression (in melanocytic and non-melanocytic lesions) and two independent variables, such as the age of the patients and the  anatomical sites ( if sun exposed or not sun exposed) of the lesions analyzed. Cell population was considered positive for antibody-specific p16 and Ki67 when at least 33 % of the cells showed well defined nuclear and/or cytoplasmic staining. A special p16 and Ki67 trend was found only in Spitz nevus (SN), atypical Spitz nevus (ASN) and invasive malignant melanoma (MM) . While, regarding the other lesions ( junctional melanocytic nevus, in situ MM, superficial spreading MM, non-melanoma cancers) discriminative values were not found. P16  was over expressed on sun exposed sites and was hypo expressed on non-sun-exposed areas , founding a statistical significance correlation ( p &lt; 0.03); while,  p16 expression was over expressed in patients  ≥ 61 while it was hypo expressed in patients ≤ 60 ( p = 0.09). </p

    Development and In Vivo Evaluation of Multidrug Ultradeformable Vesicles for the Treatment of Skin Inflammation

    The aim of this work was to evaluate the effect of two chemically different edge activators, i.e., Tween® 80 and sodium deoxycholate, on (i) the physical, mechanical, and biological properties of ultradeformable vesicles, and (ii) the administration of naproxen sodium-loaded multidrug ultradeformable vesicles for the transdermal route in order to obtain therapeutically meaningful drug concentrations in the target tissues and to potentiate its anti-inflammatory effect by association with the antioxidant drug idebenone. The results obtained in this investigation highlighted a synergistic action between naproxen and idebenone in the treatment of inflammatory disease with a more pronounced anti-inflammatory effect in multidrug ultradeformable vesicles compared to the commercial formulation of Naprosyn® gel. Systems made up of Tween® 80 appeared to be the most suitable in terms of percutaneous permeation and anti-inflammatory activity due to the greater deformability of these vesicles compared to multidrug ultradeformable vesicles with sodium deoxycholate. Our findings are very encouraging and suggest the use of these carriers in the topical treatment of inflammatory diseases

    Abstract Traumatic soft tissue injuries of the finger represent a frequent hand injury challenging hand surgeons. We report a case involving a 30-year-old man with a dorsal index finger soft tissue wound failing conservative treatment. The novel use of applied fibrin membranes and concentrated growth factors yielded complete resolution of the injury in 6 months without need for skin grafting

    Optimization of Pyrazole Compounds as Antibiotic Adjuvants Active against Colistin- and Carbapenem-Resistant Acinetobacter baumannii

    The diffusion of antibiotic-resistant, Gram-negative, opportunistic pathogens, an increasingly important global public health issue, causes a significant socioeconomic burden. Acinetobacter baumannii isolates, despite causing a lower number of infections than Enterobacterales, often show multidrug-resistant phenotypes. Carbapenem resistance is also rather common, prompting the WHO to include carbapenem-resistant A. baumannii as a "critical priority" for the discovery and development of new antibacterial agents. In a previous work, we identified several series of compounds showing either direct-acting or synergistic activity against relevant Gram-negative species, including A. baumannii. Among these, two pyrazole compounds, despite being devoid of any direct-acting activity, showed remarkable synergistic activity in the presence of a subinhibitory concentration of colistin on K. pneumoniae and A. baumannii and served as a starting point for the synthesis of new analogues. In this work, a new series of 47 pyrazole compounds was synthesized. Some compounds showed significant direct-acting antibacterial activity on Gram-positive organisms. Furthermore, an evaluation of their activity as potential antibiotic adjuvants allowed for the identification of two highly active compounds on MDR Acinetobacter baumannii, including colistin-resistant isolates. This work confirms the interest in pyrazole amides as a starting point for the optimization of synergistic antibacterial compounds active on antibiotic-resistant, Gram-negative pathogens

    Patient-reported impact of spondyloarthritis on work disability and working life: the ATLANTIS survey

    Background: The aim was to establish how patients experience the impact of spondyloarthritis (SpA) on work disability and working life. Methods: The survey was performed in 17/20 regions in Italy (1 January to 31 March 2013). A multiple-choice questionnaire was published on the official website of the sponsor - the National Association of Rheumatic Patients (ANMAR) - and hard-copies were distributed at outpatient clinics for rheumatic patients. Results: Respondents (n = 770) were of both sexes (56 % men), educated (62 % at high school or more), of working age (75 % aged 6460 years), and affected by SpA. The most common types diagnosed were ankylosing spondylitis (AS) (39 %) and psoriatic arthritis (PsA) (36 %). Respondents were working full-time (45 %), part-time (8 %) or had retired (22 %); 15 % were unemployed (for reasons linked to the disease or for other reasons, students or housewives). Patients reported disability (39 %), were receiving disability benefits (34 %), were experiencing important limitations that were hindering their professional development/career (36 %) and some had to change/leave their job or lost it because of SpA (21 %). Employed respondents (n = 383) had worked on average 32.2 h in the last 7 days. More hours of work were lost over the last 7 days due to SpA (2.39 h vs 1.67 h). The indirect costs of the disease amounted to \u20ac106/week for patients reporting well-being/good physical conditions/improvement and \u20ac216/week for those reporting permanent impairment. Conclusions: Most patients were in the midst of their productive years and were experiencing considerable difficulties in carrying out their job because of the disease: half of them reported disability and one third were experiencing important limitations in their career perspective
