    Análise quantitativa dos tecidos gengivais de ratos tratados com fenitoína e ciclosporina

    Os aumentos gengivais podem ser decorrentes de reações teciduais a estímulos idiopáticos, patológicos e farmacológicos. O objetivo desse trabalho foi avaliar morfometricamente e estereologicamente a ação da fenitoína (Fen) e ciclosporina (CsA) sobre os tecidos gengivais de ratos. Dez ratos receberam, por via intraperitonial, Fen na dose inicial de 2 mg/kg de peso corporal/dia, aumentando 2 mg a cada duas semanas, durante 60 dias. Em outros 10 ratos, administraram-se, por via subcutânea, 10mg/kg de peso corporal/dia de CsA, durante o mesmo período do grupo anterior. Os valores morfométricos e estereométricos dos tecidos gengivais dos ratos tratados com CsA foram significativamente maiores quando comparado com os valores dos tecidos gengivais do grupo tratado com Fen. Esses resultados sugerem que a CsA na dose utilizada é mais eficaz no desenvolvimento do aumento gengival em ratos, podendo estar atuando na proliferação de fibroblastos e no desequilíbrio fisiológico da síntese de fibras colágenas.Reactions to a number of idiopathic, pathological and pharmacological stimuli may result in overgrowth of gingiva, whose mechanisms have not been established yet. The aim of the present work was to evaluate morphometrically and stereologically the effect of phenytoin (PHT) and cyclosporine (Cs) on the gingival tissues of rats. Ten rats received daily IP injection of PHT, 2 mg/kg of body weight/day, which was increased in 2 mg at each two weeks, during 60 days. Ten rats received 10 mg/kg of body weight/day of Cs, subcutaneously, during the same period of the previous group. All rats treated with Cs developed gingival overgrowth, with increased thickness of the epithelium, height and wideness of connective tissues, as well as, increased density of fibroblasts and collagen fibers, when compared with the rats treated with PHT and with the control. These results suggest that Cs, in the used dose, is more efficient in the development of gingival overgrowth in rats. Cs-induced gingival overgrowth is a tissue with an altered composition, characterized by increasing of the density of fibroblasts and by physiologic unbalance of collagen synthesis

    Freqüência de Candida sp. em biópsias de lesões da mucosa bucal

    Candidosis is the most common fungal infection in the oral cavity, and is usually associated with local and systemic predisposing factors. The ocurrence and relevance of Candidal infection in oral lesions such as liquen planus, leukoplakias and carcinomas are still to be understood. The aim of the present study was to define the frequency of infection by Candida sp. on biopsies of oral mucosal lesions and associate its presence with malignant and dysplastic lesions. Histopathology reports issued between 1990 and 2001 inclusive were reviewed. Three sections of each mucosal biopsy were stained using the periodic acid-Schiff (PAS) technique. From the 832 biopsies 27.2% were PAS positive, of which 83.25% were obtained from male patients. There was positive association between fungic infection and mild, moderate and severe epithelial dysplasia, squamous cell carcinoma and hiperqueratosis (p < 0.05). There was no association between fungic infection and inflammatory fibrous hyperplasia, hyperkeratosis, lichen planus and pyogenic granuloma (p < 0.05). The frequency of infection in the tongue was significantly higher (p < 0.05) than in the other sites. Our results do not show a causal relation between Candida sp. and dysplastic lesions and carcinomas, but do confirm the higher presence of that microrganism in those lesions.O objetivo desse trabalho foi determinar a freqüência da infecção por Candida sp. em biópsias de lesões da mucosa bucal, assim como associar a presença de Candida sp. com lesões malignas e lesões com vários graus de displasia. Foram utilizadas 832 biópsias da mucosa bucal, previamente incluídas em parafinas, cujos blocos foram obtidos dos arquivos da Disciplina de Patologia da Faculdade de Odontologia de Araraquara da UNESP, no período entre 1990-2001. Três cortes seqüenciais foram corados pelo ácido periódico de Schiff (PAS). Do total de biópsias 27,2% foram PAS positivas, dessas 83,25% eram provenientes de pacientes do sexo masculino. Houve associação positiva entre infecção com displasia epitelial leve, moderada, severa, carcinoma espinocelular e hiperqueratose (p < 0,05). Não houve associação entre hiperplasia fibrosa inflamatória, líquen plano, granuloma piogênico (p < 0,05) com infecções fúngicas. A língua foi o sítio mais acometido por infecções em relação a outros sítios (p < 0,05). A partir dos dados quantitativos, concluiu-se que houve correlação positiva de infecção por fungos, lesões displásicas e carcinoma, sendo mais freqüente no sexo masculino. Estes dados não permitem inferir se o fungo causa displasia epitelial e carcinoma, mas confirmam a maior presença de Candida nessas lesões

    Quantitative evaluation of Streptococcus mutans and Candida sp and salivary factors in the oral cavity of patients submitted to radiotherapy

    O objetivo deste trabalho foi analisar quantitativamente os microrganismos, Streptococcus do grupo mutans e Candida sp, da cavidade bucal de pacientes com carcinoma de orofaringe antes, durante e após o tratamento com radioterapia e correlacionar fatores salivares como pH, capacidade tampão (CT) e fluxo salivar (FS). Amostras de saliva foram coletadas, diluídas e inoculadas em ágar SB-20 e ágar Sabouraud, respectivamente para Streptococcus do grupo mutans e Candida sp. Previamente à diluição, a saliva concentrada foi analisada, determinando-se os fatores salivares. Após crescimento das colônias, o número de microrganismos foi determinado em UFC/ml. A análise dos resultados permitiu concluir que houve correlação positiva entre os fatores salivares e a presença de microrganismos ilustrada pelo aumento no número de UFC/ml dos microrganismos analisados concomitantemente com a diminuição do fluxo salivar. Os efeitos da radiação comprometeram a homeostasia salivar e favoreceram o aumento das infecções por leveduras e bactérias durante o tratamento radioterápico.The aim of this study was to quantify the microorganisms Streptococcus mutans and Candida sp in the oral cavity of patients with oropharynx carcinoma, before, during and after radiotherapy, and to correlate the results with salivary factors such as pH, buffer capacity (CT) and flow rate (FS). Saliva samples were collected, diluted and inoculated in SB-20 agar and in Sabouraud agar, for Streptococcus mutans and Candida sp, respectively. Previously to dilution, the concentrated saliva was analyzed, and the salivary factors were determined. After the growth of colonies, the number of microorganisms was determined in CFU/ml. The analysis of the results allowed to conclude that the salivary factors are related to the presence of microorganisms, and that the number of CFU/ml increased as salivary flow rate decreased.     Systemic Dietary Hesperidin Modulation of Osteoclastogenesis, Bone Homeostasis and Periodontal Disease in Mice

    This study aimed to evaluate the effects of hesperidin (HE) on in vitro osteoclastogenesis and dietary supplem entation on mouse periodontal disease and femoral bone phenotype. RAW 264.7 cells were stimulated with RANKL in the presence or absence of HE (1, 100 or 500 µM) for 5 days, and evaluated by TRAP, TUNEL and Western Blot (WB) analyses. In vivo, C57BL/6 mice were given HE via oral gavage (125, 250 and 500 mg/kg) for 4 weeks. A sterile silk ligature was placed between the first and second right maxillary molars for 10 days and microcomputed tomography (μCT), histopathological and immunohistochemical evaluation were performed. Femoral bones subjected or not to dietary HE (500 mg/kg) for 6 and 12 weeks were evaluated using μCT. In vitro, HE 500 µM reduced formation of RANKL-stimulated TRAP-positive(+) multinucleated cells (500 µM) as well as c-Fos and NFATc1 protein expression (p < 0.05), markers of osteoclasts. In vivo, dietary HE 500 mg/kg increased the alveolar bone resorption in ligated teeth (p < 0.05) and resulted in a significant increase in TRAP+ cells (p < 0.05). Gingival inflammatory infiltrate was greater in the HE 500 mg/kg group even in the absence of ligature. In femurs, HE 500 mg/kg protected trabecular and cortical bone mass at 6 weeks of treatment. In conclusion, HE impaired in vitro osteoclastogenesis, but on the contrary, oral administration of a high concentration of dietary HE increased osteoclast numbers and promoted inflammation-induced alveolar bone loss. However, HE at 500 mg/kg can promote a bone-sparing effect on skeletal bone under physiological conditions

    Genetic polymorphism of Streptococcus mutans in Brazilian family members Polimorfismo genético de Streptococcus mutans em membros de famílias brasileiras

    The aim of this study was to determine whether random amplified polymorphic DNA (AP-PCR) analysis is able to differentiate genetically different clones of mutans streptococci, in 22 Brazilian family members. Stimulated saliva samples were collected from fathers, mothers and infants. For 5-18 months babies with erupting primary dentition, plaque samples were collected using sterile tooth pick tips. From these samples, mutans streptococci were isolated on SB-20 agar plates. After growth, representative colonies were identified by biochemical methods on the basis of carbohydrate fermentation. Streptococcus mutans isolates were obtained from all family members and AP-PCR typed separately with a random primer (OPA-13). Bacterial cell lysates were used as template in PCR reactions and the amplified DNA fragments obtained were compared by agarose gel electrophoresis. Results demonstrated that the father shared the baby's genotype in three families and the mother shared the baby's genotype in 12 families seven babies harbored Streptococcus mutans strains similar to those of their siblings. The technique was able to demonstrate the genetic Streptococcus mutans in Brazilian family members.<br>O objetivo deste estudo foi determinar, através da técnica de reação em cadeia da polimerase com primers arbitrários (AP-PCR) a capacidade de diferenciar clones geneticamente distintos de Streptococcus mutans e estabelecer o grau de similaridade intrafamilial para os isolados. Para o presente estudo, foram selecionadas 22 famílias brasileiras. Amostras de saliva foram coletadas de todos os membros das famílias. Das crianças com idade entre 5-18 meses obteve-se amostras de placa dental. Após o isolamento das colônias com características morfológicas, realizou-se a identificação bioquímica com base na fermentação de carboidratos. O polimorfismo genético de Streptococcus mutans foi pesquisado através da técnica de AP-PCR utilizando-se o primer OPA-13. Os fragmentos de DNA obtidos foram amplificados e comparados através de eletroforese em gel de agarose. Dentre as espécies identificadas nas 22 famílias analisadas, o pai apresentou cepas com similaridade genética aos dos bebês em três das famílias analisadas; em 12 famílias a mãe apresentou cepas com similaridade com as cepas de Streptococcus mutans do bebê e 7 bebês apresentaram cepas de S. mutans com similaridade genética das cepas do irmão mais velho. A técnica de AP-PCR foi eficaz em demonstrar a heterogeneidade genética de Streptococcus mutans entre os membros das famílias brasileiras analisadas

De fato, informações equilibradas, acuradas e atuais estão colaborando de forma estimulante no estudo da relação entre doença periodontal e periapical crônica com o desenvolvimento ou estabelecimento de algumas doen- ças cardiovasculares. Dessa forma, o objetivo da presente trabalho foi descrever de forma sucinta e objetiva a relação entre doença periodontal crônica e doença periapical como fatores de risco para o desenvolvimento ou estabeleciomento da aterosclerose.     Comparison of two different methods for detecting periodontal pathogenic bacteria

    perform a comparative analysis between two methods for detecting Porphyromonas gingivalis, Tannerella forsythia and Porphyromonas endodontalis in periodontal plaque samples. Methods: The study sample consisted of twenty systemically healthy patients showing generalized chronic periodontitis. The subgingival samples for microbiological analysis were collected before (baseline) and 60 days after a basic periodontal therapy from 30 non-adjacent affected sites (Probing Depth (PD): 5-7 mm, Clinical Attachment Loss (CAL) &#8805; 5 mm, positive for Bleeding on Probing (BOP)). Microbiological analysis was performed by PCR and qPCR. To allow a comparative analysis between both methods, qPCR was divided in three different scores (score 2: presence of more than 100 bacteria; score 1: presence of 10-100 bacteria, and score 0: absence of bacteria), in accordance to DNA quantity, while for PCR two scores were assigned: presence or absence of bacteria. Results: qPCR demonstrated higher sensitivity in the detection of these pathogens compared with PCR when scores 1 and 2 were considered positive. However, when only score 2 was considered positive, PCR and qPCR showed better agreement. Conclusions: qPCR demonstrated higher sensitivity than conventional PCR for detection of low numbers of microorganisms and can be useful for the quantification of periodontopathogens

    El &oacute;xido n&iacute;trico (NO) es un radical libre que participa de pr&aacute;cticamente todos los procesos fisiol&oacute;gicos del organismo; sin embargo, en grandes concentraciones puede actuar de forma perjudicial en los tejidos. Su acci&oacute;n inmuno-regulatoria est&aacute; presente en la inflamaci&oacute;n y en los mecanismos de auto inmunidad, siendo intensamente estudiada en el &aacute;rea m&eacute;dica. Recientemente, algunos estudios tambi&eacute;n han citado al NO como factor etiopatog&eacute;nico de la enfermedad periodontal, el cual posee naturaleza inflamatoria y multifactorial. La acci&oacute;n del NO depende de su concentraci&oacute;n en el organismo, pudiendo ser ben&eacute;fica o perjudicial y, basada en eso, investigaciones han enfocado el efecto de algunos inhibidores de NO como supuestos agentes terap&eacute;uticos en los procesos inflamatorios. A partir de ese contexto, este trabajo tuvo como objetivo relatar el papel del NO y de la inhibici&oacute;n de la &oacute;xido n&iacute;trico sintasa (NOS) en el proceso de la modulaci&oacute;n de la enfermedad periodontal. En conclusi&oacute;n, se puede sugerir que el NO parece ejercer un papel fundamental en la evoluci&oacute;n de la enfermedad periodontal inflamatoria y, que la inhibici&oacute;n de la NOS puede ser considerada una terap&eacute;utica promisora en la modulaci&oacute;n del proceso inflamatorio.AbstractNitric oxide (NO) is a free radical with participation in almost all physiologic host processes; however, in high concentrations it may damage the tissues. Its immunoregulatory action is present in the inflammation and in auto-immune mechanisms, being intensively studied in the medical area. Recently, some studies have also reported that NO could play a role as etiopathogenic factor of the periodontal disease, which shows inflammatory and multifactorial course. Due to beneficial or damage effects of NO, according to its concentrations, some studies have been focused in the evaluation of inhibitor of NO-Synthase (NOS) as therapeutic agents in inflammatory processes. In this context, the aim of this study was to report the role of NO and NOS inhibition in the periodontal disease modulation process. In conclusion, it could be suggested that NO seems to play an essential role in evolution of inflammatory periodontal disease, and that the NOS inhibition may be considered as a promising therapeutic in modulation of inflammatory process
