    Redox modulation of plant developmental regulators from the class I TCP transcription factor family

    TEOSINTE BRANCHED1-CYCLOIDEA-PROLIFERATING CELL FACTOR1 (TCP) transcription factors participate in plant developmental processes associated with cell proliferation and growth. Most members of class I, one of the two classes that compose the family, have a conserved cysteine at position 20 (Cys-20) of the TCP DNA-binding and dimerization domain. We show that Arabidopsis (Arabidopsis thaliana) class I proteins with Cys-20 are sensitive to redox conditions, since their DNAbinding activity is inhibited after incubation with the oxidants diamide, oxidized glutathione, or hydrogen peroxide or with nitric oxide-producing agents. Inhibition can be reversed by treatment with the reductants dithiothreitol or reduced glutathione or by incubation with the thioredoxin/thioredoxin reductase system. Mutation of Cys-20 in the class I protein TCP15 abolished its redox sensitivity. Under oxidizing conditions, covalently linked dimers were formed, suggesting that inactivation is associated with the formation of intermolecular disulfide bonds. Inhibition of class I TCP protein activity was also observed in vivo, in yeast (Saccharomyces cerevisiae) cells expressing TCP proteins and in plants after treatment with redox agents. This inhibition was correlated with modifications in the expression of the downstream CUC1 gene in plants. Modeling studies indicated that Cys-20 is located at the dimer interface near the DNA-binding surface. This places this residue in the correct orientation for intermolecular disulfide bond formation and explains the sensitivity of DNA binding to the oxidation of Cys-20. The redox properties of Cys-20 and the observed effects of cellular redox agents both in vitro and in vivo suggest that class I TCP protein action is under redox control in plants.Fil: Viola, Ivana Lorena.     The Impact of the Long-Distance Transport of a BEL1-Like Messenger RNA on Development

    BEL1- and KNOTTED1-type proteins are transcription factors from the three-amino-loop-extension superclass that interact in a tandem complex to regulate the expression of target genes. In potato (Solanum tuberosum), StBEL5 and its Knox protein partner regulate tuberization by targeting genes that control growth. RNA movement assays demonstrated that StBEL5 transcripts move through the phloem to stolon tips, the site of tuber induction. StBEL5 messenger RNA originates in the leaf, and its movement to stolons is induced by a short-day photoperiod. Here, we report the movement of StBEL5 RNA to roots correlated with increased growth, changes in morphology, and accumulation of GA2-oxidase1, YUCCA1a, and ISOPENTENYL TRANSFERASE transcripts. Transcription of StBEL5 in leaves is induced by light but insensitive to photoperiod, whereas in stolon tips growing in the dark, promoter activity is enhanced by short days. The heterodimer of StBEL5 and POTH1, a KNOTTED1-type transcription factor, binds to a tandem TTGAC-TTGAC motif that is essential for regulating transcription. The discovery of an inverted tandem motif in the StBEL5 promoter with TTGAC motifs on opposite strands may explain the induction of StBEL5 promoter activity in stolon tips under short days. Using transgenic potato lines, deletion of one of the TTGAC motifs from the StBEL5 promoter results in the reduction of GUS activity in new tubers and roots. Gel-shift assays demonstrate BEL5/POTH1 binding specificity to the motifs present in the StBEL5 promoter and a double tandem motif present in the StGA2-oxidase1 promoter. These results suggest that, in addition to tuberization, the movement of StBEL5 messenger RNA regulates other aspects of vegetative development.Fil: Lin, Tian.     Nutritional habits in recipients of cash-transfer plan in the Municipality of Berisso

    En este artículo se describen algunos de los resultados de un proyecto de interdisciplinario de investigación/ intervención en población beneficiaria de planes sociales del Municipio de Berisso (Buenos Aires, Argentina). El objetivo del proyecto fue analizar qué sucede con la alimentación, en esta nueva modalidad de entrega de dinero, en la que los beneficiarios tienen la posibilidad de elegir su abastecimiento. Con este fin se llevó adelante un estudio prospectivo de corte trasversal, realizado en tres etapas (diagnóstico, intervención, impacto) con combinación y uso alternativo de técnicas cuantitativas y cualitativas. Se observó una mejora en los ingresos y un aporte significativo a la educación en salud alimentaria, pero la misma no se tradujo en una modificación de los alimentos consumidos, ni del estado nutricional de la población.This paper presents the main findings of an interdisciplinary research/intervention project conducted in the Municipality of Berisso (Buenos Aires Province, Argentina), on people receiving social welfare cash transfer plans from the Municipal government. The aim of the project was to assess the impact of cash transfer plans on feeding habits. The study was based on a cross-sectional prospective cohort in three analytical stages (diagnosis, intervention, impact), through a combination and alternative use of quantitative and qualitative techniques. The findings gathered showed an income upgrading of recipients of cash transfers as well a meaningful contribution to dietary education.     Relation between cost of drug treatment and body mass index in people with type 2 diabetes in Latin America

    Aims Despite the frequent association of obesity with type 2 diabetes (T2D), the effect of the former on the cost of drug treatment of the latest has not been specifically addressed. We studied the association of overweight/obesity on the cost of drug treatment of hyperglycemia, hypertension and dyslipidemia in a population with T2D. Methods This observational study utilized data from the QUALIDIAB database on 3,099 T2D patients seen in Diabetes Centers in Argentina, Chile, Colombia, Peru, and Venezuela. Data were grouped according to body mass index (BMI) as Normal (18.5BMI<25), Overweight (25BMI<30), and Obese (BMI30). Thereafter, we assessed clinical and metabolic data and cost of drug treatment in each category. Statistical analyses included group comparisons for continuous variables (parametric or non-parametric tests), Chi-square tests for differences between proportions, and multivariable regression analysis to assess the association between BMI and monthly cost of drug treatment. Results Although all groups showed comparable degree of glycometabolic control (FBG, HbA1c), we found significant differences in other metabolic control indicators. Total cost of drug treatment of hyperglycemia and associated cardiovascular risk factors (CVRF) increased significantly (p<0.001) with increment of BMI. Hyperglycemia treatment cost showed a significant increase concordant with BMI whereas hypertension and dyslipidemia did not. Despite different values and percentages of increase, this growing cost profile was reproduced in every participating country. BMI significantly and independently affected hyperglycemia treatment cost. Conclusions Our study shows for the first time that BMI significantly increases total expenditure on drugs for T2D and its associated CVRF treatment in Latin America.Fil: Elgart, Jorge Federico.     The X-ray variability of Seyfert 1.8/1.9 galaxies

    Seyfert 1.8/1.9 are sources showing weak broad H-alpha components in their optical spectra. We aim at testing whether Seyfert 1.8/1.9 have similar properties at UV and X-ray wavelengths to Seyfert 2. We use the 15 Seyfert 1.8/1.9 in the Veron Cetty and Veron catalogue with public data available from the Chandra and/or XMM-Newton archives at different dates, with timescales between observations ranging from days to years. Our results are homogeneously compared with a previous work using the same methodology applied to a sample of Seyfert 2 (Hernandez-Garcia et al. 2015). X-ray variability is found in all 15 nuclei over the aforementioned ranges of timescales. The main variability pattern is related to intrinsic changes in the sources, which are observed in ten nuclei. Changes in the column density are also frequent, as they are observed in six nuclei, and variations at soft energies, possibly related to scattered nuclear emission, are detected in six sources. X-ray intraday variations are detected in six out of the eight studied sources. Variations at UV frequencies are detected in seven out of nine sources. A comparison between the samples of Seyfert 1.8/1.9 and 2 shows that, even if the main variability pattern is due to intrinsic changes of the sources in the two families, these nuclei exhibit different variability properties in the UV and X-ray domains. In particular, variations in the broad X-ray band on short time-scales (days/weeks), and variations in the soft X-rays and UV on long time-scales (months/years) are detected in Seyfert 1.8/1.9 but not in Seyfert 2. Overall, we suggest that optically classified Seyfert 1.8/1.9 should be kept separated from Seyfert 2 galaxies in UV/X-ray studies of the obscured AGN population because their intrinsic properties might be different.Comment: Accepted for publication in A&A. arXiv admin note: text overlap with arXiv:1505.0116

    Seasonal freshening of NW Mediterranean surface water impacts microbial heterotrophic activity and dissolved organic matter

    International audienceThe Rhone river represents the most important source of freshwater, nutrients and organic matter to the northwestern (NW) Mediterranean Sea and riverine input markedly affects biogeochemistry and ecosystem functioning in the estuarine and coastal zone. Structures of low salinity waters (LSW) originating near the river plume can also be transported along the continental shelf and offshore. The objective of the present study was to investigate the influence of LSW distant from their source, focusing on dissolved organic matter (DOM) and related microbial processes during two annual cycles (2007 and 2008) at a time series site characterized by the regular occurrence of LSW in spring (Microbial Observatory Laboratoire Arago). We observed enhanced bacterial heterotrophic production and community respiration and specific DOM features within these LSW, concurrently with low net community production. Our results suggest that LSW represent a mechanism of labile DOM supply, thereby sustaining enhanced heterotrophic microbial metabolism
