Juvenile polyposis of infancy in a child with deletion of BMPR1A and PTEN genes: Surgical approach
Juvenile polyposis of infancy is the most severe and life-threatening form of juvenile polyposis. This disease typically presents in the first two years of life with gastrointestinal bleeding, diarrhea, inanition, and exudative enteropathy. In very few reports concerning this entity, a large deletion in the long arm of chromosome 10 (10q23), encompassing the PTEN and BMPR1A genes, was found. The authors report a case of delayed diagnosis of juvenile polyposis of infancy at 6years of age. A 3.34Mb long de novo deletion was identified at 10q23.1q23.31, encompassing the PTEN and BMPR1A genes. The disease course was severe with diarrhea, abdominal pain, inanition, refractory anemia, rectal bleeding, hypoalbuminemia, and exudative enteropathy. A sub-total colectomy, combined with intraoperative endoscopic removal of ileal and rectal stump polyps, was required for palliative disease control
Guidelines for the recording and evaluation of pharmaco-EEG data in man: the International Pharmaco-EEG Society (IPEG)
The International Pharmaco-EEG Society (IPEG) presents updated guidelines summarising the requirements for the recording and computerised evaluation of pharmaco-EEG data in man. Since the publication of the first pharmaco-EEG guidelines in 1982, technical and data processing methods have advanced steadily, thus enhancing data quality and expanding the palette of tools available to investigate the action of drugs on the central nervous system (CNS), determine the pharmacokinetic and pharmacodynamic properties of novel therapeutics and evaluate the CNS penetration or toxicity of compounds. However, a review of the literature reveals inconsistent operating procedures from one study to another. While this fact does not invalidate results per se, the lack of standardisation constitutes a regrettable shortcoming, especially in the context of drug development programmes. Moreover, this shortcoming hampers reliable comparisons between outcomes of studies from different laboratories and hence also prevents pooling of data which is a requirement for sufficiently powering the validation of novel analytical algorithms and EEG-based biomarkers. The present updated guidelines reflect the consensus of a global panel of EEG experts and are intended to assist investigators using pharmaco-EEG in clinical research, by providing clear and concise recommendations and thereby enabling standardisation of methodology and facilitating comparability of data across laboratories
Charge Ordering and Ferroelectricity in Half-doped Manganites
By means of density-functional simulations for half-doped manganites, such as
pseudocubic Pr0.5Ca0.5MnO3 and bilayer PrCa2Mn2O7, we discuss the occurrence of
ferroelectricity and we explore its crucial relation to the crystal structure
and to peculiar charge/spin/orbital ordering effects. In pseudocubic
Pr0.5Ca0.5MnO3, ferroelectricity is induced in the Zener polaron type
structure, where Mn ions are dimerized. In marked contrast, in bilayer
PrCa2Mn2O7, it is the displacements of apical oxygens bonded to either Mn3+ or
Mn4+ ions that play a key role in the rising of ferroelectricity. Importantly,
local dipoles due to apical oxygens are also intimately linked to charge and
orbital ordering patterns in MnO2 planes, which in turn contribute to
polarization. Finally, an important outcome of our work consists in proposing

Evaluation of neck circumference as a predictor of elevated cardiometabolic risk outcomes in 5–8-year-old Brazilian children
Background: Childhood overweight and obesity is a global health problem that continues to worsen in many low- and middle-income countries. Low-cost measurements for monitoring overweight and relative metabolic risk, such as neck circumference (NC), should be evaluated in different populations and age groups. /
Aim: To test associations of NC and BMI with cardiometabolic parameters in 5-8-year-old Brazilian children. /
Methods: This cross-sectional study carried out from 2004–2006 measured height, weight and NC by anthropometry, and estimated fat and fat-free mass by bioelectrical impedance. Cardiometabolic risk factors assessed were systolic and diastolic blood pressure, high- and low-density lipoprotein cholesterol, triglycerides, and homeostatic model assessment of insulin resistance (HOMA). Associations of NC and BMI with cardiometabolic risk factors were tested using multiple regression and precision-recall plot analysis. /
Results: Analyses included 371 children (52% female). NC associated positively with BMI, fat mass, and fat-free mass, and with systolic blood pressure and HOMA following adjustment for age in sex-stratified multiple regression models. However, the latter relationships largely disappeared following adjustment for BMI. Area under the curve for NC or BMI in association with systolic blood pressure or HOMA >90th percentile was low in the pooled sample, indicating poor classifier performance. /
Conclusions: NC and BMI demonstrated similar associations with cardiometabolic risk factors, although NC mostly did not correlate with risk factors independently of BMI. In contrast to previous studies, NC was a poor classifier of cardiometabolic risk factors in children. The association of NC with both fat and fat-free mass may aid in explaining its poor performance
Rpgrip1 is required for rod outer segment development and ciliary protein trafficking in zebrafish

RPGRIP1 interacts with other retinal disease-causing proteins and has been proposed to have a role in ciliary protein transport; however, its function remains elusive. Here, we describe a new zebrafish model carrying a nonsense mutation in the rpgrip1 gene. Rpgrip1homozygous mutants do not form rod outer segments and display mislocalization of rhodopsin, suggesting a role for RPGRIP1 in rhodopsin-bearing vesicle trafficking. Furthermore, Rab8, the key regulator of rhodopsin ciliary trafficking, was mislocalized in photoreceptor cells of rpgrip1 mutants. The degeneration of rod cells is early onset, followed by the death of cone cells. These phenotypes are similar to that observed in LCA and juvenile RP patients. Our data indicate RPGRIP1 is necessary for rod outer segment development through regulating ciliary protein trafficking. The rpgrip1 mutant zebrafish may provide a platform for developing therapeutic treatments for RP patients.Publisher PDFPeer reviewe
Efficacy and safety of cangrelor in patients with peripheral artery disease undergoing percutaneous coronary intervention – Insights from the CHAMPION program
Abstract Background Peripheral artery disease (PAD) is associated with an increased risk of ischemic events following percutaneous coronary intervention (PCI). More aggressive antiplatelet therapy may mitigate this risk. The present study evaluates the efficacy of cangrelor in patients with PAD undergoing PCI. Methods and results This is a pooled analysis from the CHAMPION PCI, CHAMPION PLATFORM, AND CHAMPION PHOENIX trials, evaluating cangrelor versus either clopidogrel or placebo in PCI patients. The occurrence of the primary endpoint of death, myocardial infarction, or ischemia-driven revascularization (IDR) was assessed in patients with and without PAD. GUSTO severe bleeding at 48 h was also evaluated. There were 1720 (7%) patients with PAD and 22,802 (93%) without PAD. After adjustment for differences in baseline variables, PAD patients, compared with those without PAD, experienced increased odds of the primary endpoint (OR [95% CI] = 1.27 [0.91, 1.77], P = 0.16) and GUSTO severe bleeding (OR [95% CI] = 3.24 [1.28, 8.21], P = 0.01). In PAD patients, the primary endpoint was 4.7% with cangrelor vs. 7.2% with clopidogrel (OR [95% CI] = 0.64 [0.42,0.96]); in patients without PAD the primary endpoint was 3.5% with cangrelor vs. 4.2% with clopidogrel (OR [95% CI] = 0.83 [0.72,0.95]), P-interaction 0.23. Among patients with or without PAD, there was no significant difference in the rate of GUSTO severe bleeding with cangrelor compared with control, P-interaction 0.86. Conclusions In a pooled analysis of the CHAMPION studies, PAD was associated with increased rates of ischemic and bleeding complications. Cangrelor reduced the odds of ischemic events, without increasing GUSTO severe bleeding. On exact solutions for quintessential (inflationary) cosmological models with exponential potentials
We first study dark energy models with a minimally-coupled scalar field and
exponential potentials, admitting exact solutions for the cosmological
equations: actually, it turns out that for this class of potentials the
Einstein field equations exhibit alternative Lagrangians, and are completely
integrable and separable (i.e. it is possible to integrate the system
analytically, at least by quadratures). We analyze such solutions, especially
discussing when they are compatible with a late time quintessential expansion
of the universe. As a further issue, we discuss how such quintessential scalar
fields can be connected to the inflationary phase, building up, for this class
of potentials, a quintessential inflationary scenario: actually, it turns out
that the transition from inflation toward late-time exponential quintessential
tail admits a kination period, which is an indispensable ingredient of this
kind of theoretical models. All such considerations have also been done by

Comprehensive in vitro and in vivo studies of novel melt-derived Nb-substituted 45S5 bioglass reveal its enhanced bioactive properties for bone healing
The present work presents and discusses the results of a comprehensive study on the bioactive properties of Nb-substituted silicate glass derived from 45S5 bioglass. In vitro and in vivo experiments were performed. We undertook three different types of in vitro analyses: (i) investigation of the kinetics of chemical reactivity and the bioactivity of Nb-substituted glass in simulated body fluid (SBF) by 31P MASNMR spectroscopy, (ii) determination of ionic leaching profiles in buffered solution by inductively coupled plasma optical emission spectrometry (ICP-OES), and (iii) assessment of the compatibility and osteogenic differentiation of human embryonic stem cells (hESCs) treated with dissolution products of different compositions of Nb-substituted glass. The results revealed that Nb-substituted glass is not toxic to hESCs. Moreover, adding up to 1.3 mol% of Nb2O5 to 45S5 bioglass significantly enhanced its osteogenic capacity. For the in vivo experiments, trial glass rods were implanted into circular defects in rat tibia in order to evaluate their biocompatibility and bioactivity. Results showed all Nb-containing glass was biocompatible and that the addition of 1.3 mol% of Nb2O5, replacing phosphorous, increases the osteostimulation of bioglass. Therefore, these results support the assertion that Nb-substituted glass is suitable for biomedical applications
