    Analisi biomolecolare di ceppi di Canine Distemper Virus (CDV) in furetti domestici (Mustela putorius furo)

    L\u2019agente eziologico del cimurro (CDV) appartiene alla famiglia Paramyxoviridae, genere Morbillivirus ed \ue8 causa di una patologia infettiva e contagiosa in canidi, mustelidi e procionidi. Gli Autori, descrivono un caso clinico di cimurro in due furetti domestici vaccinati con un ceppo Onderstepoort avianizzato. e la caratterizzazione biomolecolare del CDV isolato. I furetti presentavano un quadro clinico caratterizzato da dermatite pruriginosa e squamo-purulenta, alla regione del mento, peri-labiale, peri-vulvare ed al condotto uditivo esterno. I soggetti venivano a morte tre settimane dopo l\u2019esordio dei sintomi. I campioni di croste prelevati sono stati sottoposti ad estrazione degli acidi nucleici ed analizzati mediante RT-PCR per CDV. E\u2019 stata effettuata l\u2019analisi con enzimi di restrizione (RFLP-PCR) di conferma, per discriminare i ceppi vaccinali dai ceppi di campo. I prodotti di amplificazione sono stati purificati e sottoposti a sequenziamento. Le sequenze di 1823 nt del gene H del CDV sono state comparate mediante Clustal X con analoghe sequenze di ceppi di campo e ceppi vaccinali disponibili su GeneBank. Sulla base dei dati ottenuti le sequenze sono risultate appartenenti al lineage Europa, ben segregato dal lineage America-1, che raccoglie i principali ceppi vaccinali avianizzati, e differente dal cluster dei ceppi Rockborn-like. Il sequenziamento ha confermato il risultato ottenuto con la RFLP-PCR e la mancanza di relazione con il ceppo vaccinale inoculato. Le due sequenze ottenute, presentando un\u2019omologia nucleotidica elevata per ceppi isolati da cani in Italia, sono risultate differenti dal lineage Wildlife. Nel furetto l\u2019infezione da cimurro ha un esito fatale quasi nel 100% dei casi e viene controllata attraverso l\u2019uso di vaccini. Questo lavoro rappresenta un contributo alla esigua disponibilit\ue0 bibliografica in merito alle infezioni da CDV nei mustelidi ed implementa i dati disponibili sui ceppi virali circolanti.     OBTAINING MESENCHYMAL STEM CELLS FROM ADIPOSE TISSUE OR MURIN ORIGIN: EXPERIMENTAL STUDY.

    The aim of this study was to isolate and characterize rat adipose Derived Mesenchymal Stem Cells (AD-MSCs) in order to evaluate their proliferative potential and their ability to different cell types. AD-MSCs and Derived Mesenchymal Stem Cells (BM-MSCs) have the same characteristic in terms of plasticity. The advantage of adipose tissue is that it is an easier accessible source and it offers a large amount of MSCs by less invasive surgical tecniques. MSCs were obtained from subcutaneous adipose tissue of Wistar rats. first of all microbiological controls were made to exclude the presence of bacteria of fungi in then tissue. Adipose tissue was mechanically and enzimatically fragmented and stromal cell fraction was seeded in adherent culture flasks in DMEM 20% FBS. After 48 h the medium was replaced. Cells were characterized by evaluating:1)their ability tho adhere to the plastic; 2) the clonogenic potential by Colony Forming Unit (CFU) assay, 3) their ability to differentiate in 3 mesodermal lineages (adipocytes, osteocytes and chondrocytes). AD-MSCs are able to differentiate in adipocytes, osteocytes and chondrocytes as confirmed by oil red'O staining, von Kossa staining and histological analuysis respectively. This first characterization is essential for the second part of our study in which we are planning to use AD-MSCs in vivo to restore renal function after an induced ischemic damage in experimental animals

    Adherence issues related to sublingual immunotherapy as perceived by allergists

    Objectives: Sublingual immunotherapy (SLIT) is a viable alternative to subcutaneous immunotherapy to treat allergic rhinitis and asthma, and is widely used in clinical practice in many European countries. The clinical efficacy of SLIT has been established in a number of clinical trials and meta-analyses. However, because SLIT is self-administered by patients without medical supervision, the degree of patient adherence with treatment is still a concern. The objective of this study was to evaluate the perception by allergists of issues related to SLIT adherence. Methods: We performed a questionnaire-based survey of 296 Italian allergists, based on the adherence issues known from previous studies. The perception of importance of each item was assessed by a VAS scale ranging from 0 to 10. Results: Patient perception of clinical efficacy was considered the most important factor (ranked 1 by 54% of allergists), followed by the possibility of reimbursement (ranked 1 by 34%), and by the absence of side effects (ranked 1 by 21%). Patient education, regular follow-up, and ease of use of SLIT were ranked first by less than 20% of allergists. Conclusion: These findings indicate that clinical efficacy, cost, and side effects are perceived as the major issues influencing patient adherence to SLIT, and that further improvement of adherence is likely to be achieved by improving the patient information provided by prescribers. © 2010 Scurati et al, publisher and licensee Dove Medical Press Ltd

    Parapoxvirus - ORF virus (ORFV) is the etiological agent of contagious ecthyma, a severe zoonotic exanthematic disease that affects small ruminants. It is usually characterized by highly infectious pustules and papillomatous lesions on the skin of the lips, tongue, and around the mouth. In more severe cases, the skin of the eyes, feet, vulva or udder may also be affected. Orthopoxvirus has been sometimes associated with naturally spreading disease in domestic and wild animals. Parapoxvirus infection can be complicate by opportunistic agents, mainly Fusiformis necrophorus, Dermatophilus congolensis or be invaded by the larvae of flies. Little is reported on co-infection with other viruses.     Cosa è possibile dire, oggi, del soggetto, e soprattutto del suo darsi come soggetto sessuato? Il presente volume intende provare a dare una risposta a tale domanda, interrogando quelle condizioni, esistenziali prima che cliniche, quali il transessualismo, il transgenderismo ed i disordini della differenziazione sessuale, caratterizzate da una disarticolazione di ciò che viene a darsi nell’anatomia dei corpi come riferibile alla differenza dei sessi ed il vissuto soggettivo di appartenenza ad un dato genere sessuale. Più in particolare, nella prima parte del testo viene offerta al lettore, per la prima volta in lingua italiana, la traduzione di un estratto dell’opera di Magnus Hirschfeld “Die Transvestiten” e dell’articolo pubblicato nel 1949 da David Cauldwell intitolato “Psychopathia Transexualis”. All’interno di un orizzonte eminentemente clinico, vengono, quindi, di seguito, ospitati tre contributi aventi ad oggetto la condizione transessuale: i primi due all’interno di una prospettiva psicoanalitica ed il terzo all’interno di una prospettiva daseinsanalitica. Attraverso un graduale mutamento prospettico, viene, quindi, proposta una rilettura del fenomeno transgender, eleggendo quest’ultimo a figura emblematica della condizione postmoderna, e, quindi, successivamente, attraverso gli strumenti propri alla ricerca sociologica, viene proposta un’analisi di tale categoria e delle condizioni intersessuali. Un successivo capitolo propone una rilettura in chiave antropologica e psicoanalitica della figura del Femminiello Napoletano, mentre, in quello ancora successivo, viene riattraversato l’immaginario cinematografico della transessualità. Infine, gli ultimi due capitoli del volume offrono una panoramica critica di quei dispositivi giuridici, italiani ed europei, che ad oggi regolamentano le pratiche proprie al campo del transessualismo
