    Tetramethyl pyrazine exerts anti-apoptotic and antioxidant effects in a mouse model of MPTP-induced Parkinson's disease via regulation of the expressions of Bax, Bcl-2, Nrf2 and GCLC

    Purpose: To investigate the effect of tetramethyl pyrazine (TMP) on MPTP)-mediated neuronal apoptosis and oxidative imbalance in mice, and the mechanism of action involved. Methods: Forty-five mice were assigned evenly to blank control, MPTP and TMP groups. The protein concentrations of Bax, Bcl-2, cytochrome C (Cyt c), Nrf2, GCLC and cleaved caspase-3; and levels of glutathione (GSH) and thiobarbituric acid reactive products (TBARS) were evaluated and compared amongst the groups. Results: Cyt c, Bax, and cleaved caspase-3 protein levels in TMP group were significantly lower than those in MPTP group, while Bcl-2 protein expression was higher in TMP group than in MPTP mice (p < 0.05). Furthermore, TBARS was lower in TMP group than in MPTP group, while GSH level increased, relative to MPTP mice. The levels of Nrf2 and GCLC were significantly higher in TMP group than in MPTP group (p < 0.05). Conclusion: Tetramethyl pyrazine exerts anti-apoptotic and antioxidant effects on MPTP-mediated Parkinsonism via regulation of the expressions of Bax, Bcl-2, Nrf2 and glutamate-cysteine ligase catalytic subunit. Thus, TMP has potential for use in the treatment Parkinson’s disease

    Exploration of the Modulatory Property Mechanism of ELeng Capsule in the Treatment of Endometriosis Using Transcriptomics Combined With Systems Network Pharmacology

    Endometriosis is a common gynecological disease and causes severe chronic pelvic pain and infertility. Growing evidence showed that traditional Chinese medicine (TCM) plays an active role in the treatment of endometriosis. ELeng Capsule (ELC) is a Chinese medicine formula used for the treatment of endometriosis for several years. However, the mechanisms of ELC have not been fully characterized. In this study, network pharmacology and mRNA transcriptome analysis were used to study various therapeutic targets in ELC. As a result, 40 compounds are identified, and 75 targets overlapped with endometriosis-related proteins. The mechanism of ELC for the treatment of endometriosis is based on the function modules of inducing apoptosis, inhibiting angiogenesis, and regulating immunity mainly through signaling molecules and interaction (neuroactive ligand–receptor interaction), immune system–associated pathways (toll-like receptor signaling pathway), vascular endothelial growth factor (VEGF) signaling, and MAPK signaling pathway based on network pharmacology. In addition, based on RNA-sequence analysis, we found that the mechanism of ELC was predominantly associated with the regulation of the function modules of actin and cytoskeleton, epithelial–mesenchymal transition (EMT), focal adhesion, and immunity-associated pathways. In conclusion, ELC exerted beneficial effects on endometriosis, and the potential mechanism could be realized through functional modules, such as inducing apoptosis and regulating angiogenesis, cytoskeleton, and EMT. This work not only provides insights into the therapeutic mechanism of TCM for treating endometriosis but also offers an efficient way for drug discovery and development from herbal medicine

    Molecular level one-step activation of agar to activated carbon for high performance supercapacitors

    Activated carbon was synthesized by a simple one-step calcination of deoxygenated agar in a hot KOH aqueous solution, in which KOH plays both deoxidant and activation agent. The deoxygenation course omits high temperature carbonization in the conventional technology and leads to molecular level activation of agar in subsequent one-step calcination. The one-step activated carbon has superior specific surface area of 1672 m2 g−1 and total pore volume of 0.81 cm3 g−1. It also shows a maximum specific capacitance of 226 F g−1 in the KOH electrolyte, which is 1.4 times as high as that for the activated carbon by the conventional two-step method. This study provides potentially economical and effective means for the production of commercial activated carbon with high porosity for supercapacitors

    High speed self-testing quantum random number generation without detection loophole

    Quantum mechanics provides means of generating genuine randomness that is impossible with deterministic classical processes. Remarkably, the unpredictability of randomness can be certified in a self-testing manner that is independent of implementation devices. Here, we present an experimental demonstration of self-testing quantum random number generation based on an detection-loophole free Bell test with entangled photons. In the randomness analysis, without the assumption of independent identical distribution, we consider the worst case scenario that the adversary launches the most powerful attacks against quantum adversary. After considering statistical fluctuations and applying an 80 Gb ×\times 45.6 Mb Toeplitz matrix hashing, we achieve a final random bit rate of 114 bits/s, with a failure probability less than 10−510^{-5}. Such self-testing random number generators mark a critical step towards realistic applications in cryptography and fundamental physics tests.Comment: 34 pages, 10 figure

    Efficacy and Safety of Electroacupuncture on Treating Depression Related Sleep Disorders: Study Protocol of a Randomized Controlled Trial

    Background. Depression is frequently accompanied by sleep disturbances including insomnia. Insomnia may persist even after mood symptoms have been adequately treated. Acupuncture is considered to be beneficial to adjust the state of body and mind and restore the normal sleep-awake cycle. This trial is aimed at evaluating the efficacy and safety of electroacupuncture on treating insomnia in patients with depression. Methods. We describe a protocol for a randomized, single-blinded, sham controlled trial. Ninety eligible patients will be randomly assigned to one of 3 treatment groups: treatment group (acupuncture), control A group (superficial acupuncture at sham points), and control B group (sham acupuncture). All treatment will be given 3 times per week for 8 weeks. The primary outcome is the Pittsburgh Sleep Quality Index (PSQI). The secondary outcomes are sleep parameters recorded in the Actigraphy, Hamilton Rating Scale for Depression (HAMD), and Self-Rating Depression Scale (SDS). All adverse effects will be accessed by the Treatment Emergent Symptom Scale (TESS). Outcomes will be evaluated at baseline, 4 weeks after treatment, 8 weeks after treatment, and 4 weeks of follow-up. Ethics. This trial has been approved by the Ethics Committee of Shanghai Municipal Hospital of Traditional Chinese Medicine (2015SHL-KY-21) and is registered with ChiCTR-IIR-16008058

    Cinnamomum cassia Bark in Two Herbal Formulas Increases Life Span in Caenorhabditis elegans via Insulin Signaling and Stress Response Pathways

    Background: Proving the efficacy and corresponding mode of action of herbal supplements is a difficult challenge for evidence-based herbal therapy. A major hurdle is the complexity of herbal preparations, many of which combine multiple herbs, particularly when the combination is assumed to be vitally important to the effectiveness of the herbal therapy. This issue may be addressed through the use of contemporary methodology and validated animal models. Methods and Principal Findings: In this study, two commonly used traditional herbal formulas, Shi Quan Da Bu Tang (SQDB) and Huo Luo Xiao Ling Dan (HLXL) were evaluated using a survival assay and oxidative stress biomarkers in a well-established C. elegans model of aging. HLXL is an eleven herb formula modified from a top-selling traditional herbal formula for the treatment of arthritic joint pain. SQDB consists of ten herbs often used for fatigue and energy, particularly in the aged. We demonstrate here that SQDB significantly extend life span in a C. elegans model of aging. Among all individual herbs tested, two herbs Cinnamomum cassia bark (Chinese pharmaceutical name: Cinnamomi Cortex, CIN) and Panax ginseng root (Chinese pharmaceutical name: Ginseng Radix, GS) significantly extended life span in C. elegans. CIN in both SQDB and HLXL formula extended life span via modulation of multiple longevity assurance genes, including genes involved in insulin signaling and stress response pathways. All the life-span-extending herbs (SQDB, CIN and GS) also attenuated levels of H2O2 and enhanced small heat shock protein expression. Furthermore, the life spanextending herbs significantly delayed human amyloid beta (Aβ)-induced toxicity in transgenic C. elegans expressing human Aβ. Conclusion/Significance:These results validate an invertebrate model for rapid, systematic evaluation of commonly used Chinese herbal formulations and may provide insight for designing future evidence-based herbal therapy(s).     Retrospective evaluation of whole exome and genome mutation calls in 746 cancer samples

    Funder: NCI U24CA211006Abstract: The Cancer Genome Atlas (TCGA) and International Cancer Genome Consortium (ICGC) curated consensus somatic mutation calls using whole exome sequencing (WES) and whole genome sequencing (WGS), respectively. Here, as part of the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium, which aggregated whole genome sequencing data from 2,658 cancers across 38 tumour types, we compare WES and WGS side-by-side from 746 TCGA samples, finding that ~80% of mutations overlap in covered exonic regions. We estimate that low variant allele fraction (VAF < 15%) and clonal heterogeneity contribute up to 68% of private WGS mutations and 71% of private WES mutations. We observe that ~30% of private WGS mutations trace to mutations identified by a single variant caller in WES consensus efforts. WGS captures both ~50% more variation in exonic regions and un-observed mutations in loci with variable GC-content. Together, our analysis highlights technological divergences between two reproducible somatic variant detection efforts
