Combined overexpression of chitinase and defensin genesin transgenic tomato enhances resistance to Botrytis cinerea
The rice chitinase gene (CHI), the alfalfa defensin gene (alfAFP) and their bivalent gene (CHI-AFP) were introduced into tomato line Micro-Tom via Agrobacterium-mediated gene transfer method. Transformants were obtained and confirmed by GFP, PCR and Southern blot hybridization. One to four copies of transgene were integrated into the tomato nuclear genome. Transcription levels of chitinase, alfAFP and their bivalent gene CHI-AFP in various transgenic lines were determined using Northern blotand Western blot analysis. Performance test of resistance analyses to Botrytis cinerea with T1 generation transgenic tomato lines showed the transgenic lines exhibited higher resistance to the pathogens infected than that of the non-transgenic plants and the resistance levels were related toexpression levels of the transgene, showing dosage-effect. The transgenic tomato harboring CHI-AFP cassette showed the highest disease resistance; it suggested that co-transformation with alfAFP and chitinase gene was more effective than individual transformations on the resistance to B. cinerea. Some independent lines with high disease resistance, low variability and stable expression of transgenes could be selected for the further studies and molecular breeding
Trophic strategy of diverse methanogens across a river-to-sea gradient
Methanogens are an important biogenic source of methane, especially in estuarine waters across a river-to-sea gradient. However, the diversity and trophic strategy of methanogens in this gradient are not clear. In this study, the diversity and trophic strategy of methanogens in sediments across the Yellow River (YR) to the Bohai Sea (BS) gradient were investigated by high-throughput sequencing based on the 16S rRNA gene. The results showed that the diversity of methanogens in sediments varied from multitrophic communities in YR samples to specific methylotrophic communities in BS samples. The methanogenic community in YR samples was dominated by Methanosarcina, while that of BS samples was dominated by methylotrophic Methanococcoides. The distinct methanogens suggested that the methanogenic community of BS sediments did not originate from YR sediment input. High-throughput sequencing of the mcrA gene revealed that active Methanococcoides dominated in the BS enrichment cultures with trimethylamine as the substrate, and methylotrophic Methanolobus dominated in the YR enrichment cultures, as detected to a limited amount in in situ sediment samples. Methanosarcina were also detected in this gradient sample. Furthermore, the same species of Methanosarcina mazei, which was widely distributed, was isolated from the area across a river-to-sea gradient by the culture-dependent method. In summary, our results showed that a distribution of diverse methanogens across a river-to-sea gradient may shed light on adaption strategies and survival mechanisms in methanogens
Heterogeneous activation of peroxymonosulfate by a biochar-supported Co3O4 composite for efficient degradation of chloramphenicols
Herein, a new peroxymonosulfate (PMS) activation system was established using a biochar (BC)-supported Co3O4 composite (Co3O4-BC) as a catalyst to enhance chloramphenicols degradation. The effects of the amount of Co3O4 load on the BC, Co3O4-BC amount, PMS dose and solution pH on the degradation of chloramphenicol (CAP) were investigated. The results showed that the BC support could well disperse Co3O4 particles. The degradation of CAP (30 mg/L) was enhanced in the Co3O4-BC/PMS system with the apparent degradation rate constant increased to 5.1, 19.4 and 7.2 times of that in the Co3O4/PMS, BC/PMS and PMS-alone control systems, respectively. Nearly complete removal of CAP was achieved in the Co3O4-BC/PMS system under the optimum conditions of 10 wt% Co3O4 loading on BC, 0.2 g/L. Co3O4-BC, 10 mM PMS and pH 7 within 10 min. The Co3O4/BC composites had a synergistic effect on the catalytic activity possibly because the conducting BC promoted electron transfer between the Co species and HSO5- and thus accelerated the Co3+/Co(2+)redox cycle. Additionally, over 85.0 +/- 1.5% of CAP was still removed in the 10th run. Although both SO4 center dot- and OH center dot were identified as the main active species, SO4 center dot- played a dominant role in CAP degradation. In addition, two other chloramphenicols, i.e., florfenicol (FF) and thiamphenicol (TAP), were also effectively degraded with percentages of 86.4 +/- 13% and 71.8 +/- 1.0%, respectively. This study provides a promising catalyst Co3O4-BC to activate PMS for efficient and persistent antibiotics degradation. (C) 2019 Elsevier Ltd. All rights reserved
Microstructural Characterization of Graphite Spheroids in Ductile Iron
The present work brings new insights by transmission electron microscopy allowing disregarding or supporting some of the models proposed for spheroidal growth of graphite in cast irons. Nodules consist of sectors made of graphite plates elongated along a hai direction and stack on each other with their c axis aligned with the radial direction. These plates are the elementary units for spheroidal growth and a calculation supports the idea that new units continuously nucleate at the ledge between sectors
Effects of serum proteins on corrosion behavior of ISO 5832–9 alloy modified by titania coatings
Stainless steel ISO 5832–9 type is often used to
perform implants which operate in protein-containing physiological
environments. The interaction between proteins and
surface of the implant may affect its corrosive properties. The
aim of this work was to study the effect of selected serum
proteins (albumin and γ-globulins) on the corrosion of ISO
5832–9 alloy (trade name M30NW) which surface was modified
by titania coatings. These coatings were obtained by sol–
gel method and heated at temperatures of 400 and 800 °C. To
evaluate the effect of the proteins, the corrosion tests were
performed with and without the addition of proteins with
concentration of 1 g L−1 to the physiological saline solution
(0.9 % NaCl, pH 7.4) at 37 °C. The tests were carried out
within 7 days. The following electrochemical methods were
used: open circuit potential, linear polarization resistance, and
electrochemical impedance spectroscopy. In addition, surface
analysis by optical microscopy and X-ray photoelectron spectroscopy
(XPS) method was done at the end of weekly corrosion
tests. The results of corrosion tests showed that M30NW
alloy both uncoated and modified with titania coatings exhibits
a very good corrosion resistance during weekly exposition
to corrosion medium. The best corrosion resistance in
0.9 % NaCl solution is shown by alloy samples modified by
titania coating annealed at 400 °C. The serumproteins have no
significant effect onto corrosion of investigated biomedical
steel. The XPS results confirmed the presence of proteins on
the alloy surface after 7 days of immersion in proteincontaining
solutions. The investigations were supported by the National Science Centre project No. N N507 501339. The authors gratefully acknowledge Dr. Janusz

Ameliorating effect of Erxian decoction combined with Fructus Schisandrae chinensis (Wu Wei Zi) on menopausal sweating and serum hormone profiles in a rat model.
Background Modified Erxian decoction (MEXD), i.e., Erxian decoction (EXD) with Fructus Schisandrae chinensis (Wu Wei Zi) added, has been used to alleviate menopausal symptoms. This study aimed to investigate the effects of MEXD on menopausal sweating and serum hormone levels in a rat model of menopause after oral administration of MEXD. Methods Quality control of MEXD was conducted by employing a reversed-phase high performance liquid chromatography column. The three treatment groups received oral administration of MEXD in 0.5% sodium carboxylmethyl cellulose (CMC-Na) at three different doses (5.5, 11, and 22 g/kg body weight) once-daily for 6 consecutive weeks, with 10 animals per group. Huangqijing oral liquor (5 mL/kg) prepared from the roots of Huang qi (Astragalus membranaceus) with an antiperspirant effect was used as a positive control. The negative control group received the same volume of vehicle (0.5% CMC-Na). Ten 3-month-old Sprague–Dawley rats were used as a young group for comparison with the treatment groups (12–14 months old rats). Blood was collected from all animals after 3–6 weeks of treatment. At the end of the treatment, the uterine weight, ovarian weight, and body weight were recorded. Serum malondialdehyde contents and superoxide dismutase activities were determined by thiobarbituric acid colorimetric assays and chemoluminescence assays, respectively. Serum levels of estradiol, follicle-stimulating hormone, and luteinizing hormone were measured by radioimmunoassays. Rat foot pad assays were used to determine the antiperspirant activity of MEXD and histological examinations were conducted on plantar sweat glands. Results Treatment with MEXD (11 g/kg) significantly inhibited sweat excretion in the menopause model rats after treatment for 3 (P = 0.0026) and 6 (P < 0.0001) weeks. The decoction markedly decreased the number of secretory cells in plantar sweat glands. In addition, MEXD (11 g/kg) significantly increased the serum estradiol levels (P < 0.001) and superoxide dismutase activities (P = 0.0405). Furthermore, MEXD (11 g/kg) markedly decreased the serum levels of follicle-stimulating hormone (P = 0.001), luteinizing hormone (P = 0.0213), and malondialdehyde (P = 0.01). Conclusion Modified Erxian decoction significantly inhibited sweat excretion, regulated serum levels of pituitary gonadotropins and estradiol, and exhibited antioxidative effects in a rat model of menopause.published_or_final_versio
Bank Political Connections and Performance in China
We examine the effects of bank’s political connection on bank performance and risk in China. We use hand-collected information on CEOs’ professional background to identify their political affiliations, and find that banks whose CEOs have former government experiences have higher return on assets, lower default risk, and lower credit risk. Additionally, politically connected banks have disproportionally higher performance when the CEOs previous worked in the same city where the current bank’s headquarter locates, had past banking experiences, spend more on entertainment and travel costs, and have higher previous administrative rankings (e.g., at the provincial or state level). These results suggest that politically connected banks have better access to lending to politically connected firms, which are high yield assets and more likely to be bailed out when in distress. Our results offer a mechanism of political rent seeking, consistent with the institutional environment of China’s banking and political system
On the General Ericksen-Leslie System: Parodi's Relation, Well-posedness and Stability
In this paper we investigate the role of Parodi's relation in the
well-posedness and stability of the general Ericksen-Leslie system modeling
nematic liquid crystal flows. First, we give a formal physical derivation of
the Ericksen-Leslie system through an appropriate energy variational approach
under Parodi's relation, in which we can distinguish the
conservative/dissipative parts of the induced elastic stress. Next, we prove
global well-posedness and long-time behavior of the Ericksen-Leslie system
under the assumption that the viscosity is sufficiently large. Finally,
under Parodi's relation, we show the global well-posedness and Lyapunov
stability for the Ericksen-Leslie system near local energy minimizers. The
connection between Parodi's relation and linear stability of the
Ericksen-Leslie system is also discussed
Observation of a ppb mass threshoud enhancement in \psi^\prime\to\pi^+\pi^-J/\psi(J/\psi\to\gamma p\bar{p}) decay
The decay channel
is studied using a sample of events collected
by the BESIII experiment at BEPCII. A strong enhancement at threshold is
observed in the invariant mass spectrum. The enhancement can be fit
with an -wave Breit-Wigner resonance function with a resulting peak mass of
and a
narrow width that is at the 90% confidence level.
These results are consistent with published BESII results. These mass and width
values do not match with those of any known meson resonance.Comment: 5 pages, 3 figures, submitted to Chinese Physics
