    Evaluation of the hazard from exposure to electron irradiation simulating that in the synchronous orbit

    The electron spectrum predicted for the synchronous orbit was simulated to determine the effects that might occur to astroscientists exposed to such irradiation while on a prolonged space station mission in that region. Miniature pigs were exposed to monoenergetic and spectral-fractionated irradiations with 0.5 to 2.1 MeV electrons. Clinical and pathological alterations observed in biopsies were correlated with depth-dose pattern and length of post irradiation period up to one year. With monoenergetic electrons, the lowest dose causing a recognizable lesion was 1450 rad and with increasing dose lesions appeared earlier and were more severe. At the highest dose given, 2650 rad, ulceration extending into the dermis was present by twenty one days and required about four months for complete healing. Spectral-fractionated irradiations, in which the total dose range was essentially comparable to that of the monoenergetic series, resulted in very minimal outer dermis edema at 1790 rad and at no dose employed did necrosis of epidermis or ulceration into dermis occur

    Calibration of liquid argon and neon detectors with 83Krm^{83}Kr^m

    We report results from tests of 83^{83}Krm^{\mathrm{m}}, as a calibration source in liquid argon and liquid neon. 83^{83}Krm^{\mathrm{m}} atoms are produced in the decay of 83^{83}Rb, and a clear 83^{83}Krm^{\mathrm{m}} scintillation peak at 41.5 keV appears in both liquids when filling our detector through a piece of zeolite coated with 83^{83}Rb. Based on this scintillation peak, we observe 6.0 photoelectrons/keV in liquid argon with a resolution of 6% (σ\sigma/E) and 3.0 photoelectrons/keV in liquid neon with a resolution of 19% (σ\sigma/E). The observed peak intensity subsequently decays with the 83^{83}Krm^{\mathrm{m}} half-life after stopping the fill, and we find evidence that the spatial location of 83^{83}Krm^{\mathrm{m}} atoms in the chamber can be resolved. 83^{83}Krm^{\mathrm{m}} will be a useful calibration source for liquid argon and neon dark matter and solar neutrino detectors.Comment: 7 pages, 12 figure

    Click Here Now: The Impact of Color and Size on Advertisements

    Businesses and companies are always trying to find the most effective way to advertise their products to create higher click-through rates, increasing the intent to purchase, and consequently raising sales and revenue. This research examines the effect that several different design elements of an online advertisement have on the attitudes or behaviors of consumers. An online survey was sent out to students at Gettysburg College, consisting of pairs of advertisements differing either in color, hue, or size. Participants in the online survey were asked to click on the image that they preferred. All three of these categories had a significant effect on click-through rates on advertisements. Highly-saturated advertisements were significantly preferred over the low-saturated advertisements. There was a significant difference in size preference, but the average preference was evenly split between square and vertical rectangle sizes in the shape category. Warm colors were marginally preferred over cool colors in the images that were used. These findings are important for businesses in order for them to receive the greatest number of clicks on products advertised because those clicks lead to purchases of the product. Further research could also include the examination of other characteristics of advertisements

    Cortical actin recovery at the immunological synapse leads to termination of lytic granule secretion in cytotoxic T lymphocytes.

    CD8+ cytotoxic T lymphocytes (CTLs) eliminate virally infected cells through directed secretion of specialized lytic granules. Because a single CTL can kill multiple targets, degranulation must be tightly regulated. However, how CTLs regulate the termination of granule secretion remains unclear. Previous work demonstrated that centralized actin reduction at the immune synapse precedes degranulation. Using a combination of live confocal, total internal reflection fluorescence, and superresolution microscopy, we now show that, after granule fusion, actin recovers at the synapse and no further secretion is observed. Depolymerization of actin led to resumed granule secretion, suggesting that recovered actin acts as a barrier preventing sustained degranulation. Furthermore, RAB27a-deficient CTLs, which do not secrete cytotoxic granules, failed to recover actin at the synapse, suggesting that RAB27a-mediated granule secretion is required for actin recovery. Finally, we show that both actin clearance and recovery correlated with synaptic phosphatidylinositol 4,5-bisphosphate (PIP2) and that alterations in PIP2 at the immunological synapse regulate cortical actin in CTLs, providing a potential mechanism through which CTLs control cortical actin density. Our work provides insight into actin-related mechanisms regulating CTL secretion that may facilitate serial killing during immune responses

    The organization of engaged and quiescent translocons in the endoplasmic reticulum of mammalian cells

    Protein translocons of the mammalian endoplasmic reticulum are composed of numerous functional components whose organization during different stages of the transport cycle in vivo remains poorly understood. We have developed generally applicable methods based on fluorescence resonance energy transfer (FRET) to probe the relative proximities of endogenously expressed translocon components in cells. Examination of substrate-engaged translocons revealed oligomeric assemblies of the Sec61 complex that were associated to varying degrees with other essential components including the signal recognition particle receptor TRAM and the TRAP complex. Remarkably, these components not only remained assembled but also had a similar, yet distinguishable, organization both during and after nascent chain translocation. The persistence of preassembled and complete translocons between successive rounds of transport may facilitate highly efficient translocation in vivo despite temporal constraints imposed by ongoing translation and a crowded cellular environment
