    Polymorphisms of the bovine chemokine receptor-like 1 gene and their associations with meat quality traits in Qinchuan cattle ( Bos taurus )

    Background: We investigated the polymorphisms of the bovine chemokine receptor-like 1(CMKLR1) gene. The coding region of CMKLR1 was screened in Qinchuan cattle by PCR-RFLP technology. Results: In this study, we discovered two single nucleotide polymorphisms (SNPs) (264G > C and 762C > T) in the coding region of the CMKLR1 gene. Hence, we described the BmgT120l and Pdm1 PCR-RFLP methods for detecting the 64G > C and 762C > T mutations, respectively. PCR-RFLP and sequencing were used to analyze the two loci of CMKLR1 gene in 324 individuals, which were randomly selected from breeding populations. Furthermore, meat quality traits in another 80 Qinchuan individuals were analyzed by the comparison between the genotypes and their phenotypic data. Conclusions: The results showed that the G264C SNP and C762T SNP of bovine CMKLR1 were significantly associated with backfat thickness (BFT) and water holding capacity (WHC), respectively

    Spatial variation in grain-size population of surface sediments from northern Bering Sea and western Arctic Ocean: implications for provenance and depositional mechanisms

    In general, sediments in nature comprise populations of various diameters. Accurate information regarding the sources and depositional mechanisms of the populations can be obtained through their temporal and spatial comparisons. In this study, the grain size distribution of surface sediments from the Bering Sea and western Arctic Ocean were fitted and partitioned into populations using a log-normal distribution function. The spatial variations in the populations indicate differences in their sources and deposition mechanisms. The sediments on most of the Bering Sea Shelf originated from the Yukon River, and were transported westward by waves and currents. However, the presence of a coarser population outside Anadyr Bay was the result of Anadyr River transport. Additionally, a northward transport trend of fine suspended particles was observed on the west side of the Bering Sea Shelf. The sediments in Hope Valley in the south Chukchi Sea also originated from the Yukon River. The coarser population on the central Chukchi Sea Shelf originated from coast of Alaska to the east, not the Yukon River, and was transported by sea ice and bottom brine water. The populations of sediments from the Chukchi Basin and the base of the Chukchi Sea Slope are the result of sea ice and eddy action. Surface sediments from the western high Arctic Ocean predominantly comprised five populations, and two unique populations with mode diameters of 50–90 μm and 200–400 μm, respectively, were ubiquitous in the glacial and interglacial sediments. It was difficult to distinguish whether these two populations originated from sea ice or icebergs. Therefore, caution should be exercised when using either the > 63 μm or > 250 μm fractions in sediments as a proxy index for iceberg and ice sheet variation in the high Arctic Ocean

    A genetic study of the NOS3 gene for ischemic stroke in a Chinese population

    We recruited 560 unrelated patients with ischemic stroke and 153 unrelated controls to undertake a genetic analysis for association between the NOS3 gene and ischemic stroke. All the subjects were Chinese of Han descent. Because the NOS3 gene spans about 21 kb of DNA and contains 26 exons, we selected a single nucleotide polymorphism (SNP) rs3918181, an A to G base change located in intron 14 of the gene, as a DNA marker. PCR-based restriction fragment length polymorphism analysis was applied to genotype rs3918181 (RsaI site). The chi-square (χ2) goodness-of-fit test showed that the genotypic distributions of the marker were not deviated from Hardy-Weinberg equilibrium in both the patient group (χ2 = 0.166, p = 0.684) and the control group (χ2 = 0.421, p = 0.517). The cocaphase analysis showed allelic association of rs3918181 with ischemic stroke in males (χ2 = 4.04, p = 0.044, OR = 1.43, 95% CI 1.01∼2.02) and frequency of allele A was significantly higher in male patients than male control subjects. The χ2 test revealed genotypic association between rs3918181 and ischemic stroke in males (χ2 = 4.26, df = 1, p = 0.039, OR = 1.61, 95% CI 1.02∼2.53) but not in females. The present work suggests that rs3918181 is associated with ischemic stroke in male patients. This finding gives further evidence in support of the eNOS association with ischemic stroke in the Chinese population

    Expression of CD44 3′-untranslated region regulates endogenous microRNA functions in tumorigenesis and angiogenesis

    The non-coding 3′-untranslated region (UTR) plays an important role in the regulation of microRNA (miRNA) functions, since it can bind and inactivate multiple miRNAs. Here, we show the 3′-UTR of CD44 is able to antagonize cytoplasmic miRNAs, and result in the increased translation of CD44 and downstream target mRNA, CDC42. A series of cell function assays in the human breast cancer cell line, MT-1, have shown that the CD44 3′-UTR inhibits proliferation, colony formation and tumor growth. Furthermore, it modulated endothelial cell activities, favored angiogenesis, induced tumor cell apoptosis and increased sensitivity to Docetaxel. These results are due to the interaction of the CD44 3′-UTR with multiple miRNAs. Computational algorithms have predicted three miRNAs, miR-216a, miR-330 and miR-608, can bind to both the CD44 and CDC42 3′-UTRs. This was confirmed with luciferase assays, western blotting and immunohistochemical staining and correlated with a series of siRNA assays. Thus, the non-coding CD44 3′-UTR serves as a competitor for miRNA binding and subsequently inactivates miRNA functions, by freeing the target mRNAs from being repressed

    High-resolution circa-2020 map of urban lakes in China

    Abstract Urban lakes provide important ecological services to local communities, such as flood mitigation, biodiversity, and recreation. With rapid urbanization, urban lakes are significantly affected by socio-economic development and urgently need attention. Yet there is still a lack of datasets that include tiny urban lakes on a global or national scale. This study aims to produce a high-resolution circa-2020 map of urban lakes (≥0.001 km2) in China. The 10-m-resolution Sentinel-2 imagery and a simple but robust water extraction method was used to generate waterbodies. The accuracy of this national-scale dataset was evaluated by comparing it with manually sampled urban units, with the average accuracy of 81.85% in area and 93.35% in count. The database totally inventories 1.11 × 106 urban lakes in China, with a net area of ~2.13 × 103 km2. Overall, the spatial distribution of urban lakes in China showed strongly heterogeneous characteristics. This dataset will enhance our understanding of the distribution pattern of China’s urban lakes and contribute to better ecological and environmental management as well as sustainable urban development planning

    Early Peritonitis is an Independent Risk Factor for Mortality in Elderly Peritoneal Dialysis Patients

    Background/Aims: The impact of early peritonitis on the outcome of elderly peritoneal dialysis (PD) patients has not been studied. We aimed to research the influence of early peritonitis on patient outcomes in elderly PD patients. Methods: This study involved elderly PD patients (age ≥65) who underwent PD between Jan 1, 2004 and Jul 31, 2013. Patient characteristics were collected in our database. Early peritonitis was defined as peritonitis within 6 months after the initiation of PD. Patient survival and technique were compared among the non-peritonitis, early peritonitis and late peritonitis groups using Cox regression analysis. Results: There were 155 subjects involved in this study. The patients were divided among a non-peritonitis group (n=78), early peritonitis group (n=32) and late peritonitis group (n=45). The organisms causing first peritonitis in the two groups did not differ significantly. After adjustment for age, diabetes, serum albumin and residual renal function, multivariable Cox regression model revealed that compared with the early peritonitis group, both the non-peritonitis group (HR 0.57, RI 0.32-0.99, p=0.046) and the late peritonitis group (HR 0.37, RI 0.16-0.75, p=0.004) exhibited a lower patient mortality rate. Conclusions: Early peritonitis is an independent risk factor for mortality in elderly peritoneal dialysis patients
