    Este trabalho foi apresentado no I Seminário de Arqueologia Memória e Patrimônio, realizado pelo Museu de Ciências Naturais da PUC Minas. Seu objetivo é refletir sobre a relação entre os homens viventes no período chamado por nós de pré-história e o mundo envolvente, a partir da análise de distribuição dos conjuntos gráficos rupestres da região de Diamantina. Parte dos dados aqui apresentados foram construídos em dissertação de mestrado defendida em 2008 a partir de uma abordagem da Arqueologia da Paisagem. Mas o tempo decorrido desde então ofereceu algumas modificações ao texto original em função de novas questões e inspirações

    Resumo: a Lapa do Caboclo, em Diamantina (Minas Gerais), apresenta em suas ocupações do Holoceno Superior, quatro estruturas funerárias, correspondendo a sepultamentos secundários de composição sofisticada. Apresentamos os estudos bioarqueológicos, elementos do processo de escavação e características das estruturas (constituídas por estojos de cascas de árvores, com ossos recobertos de pigmentos, resina, penas e couro), construindo interpretações sobre as práticas funerárias.     Genetic determinants of gut microbiota composition and bile acid profiles in mice.

    The microbial communities that inhabit the distal gut of humans and other mammals exhibit large inter-individual variation. While host genetics is a known factor that influences gut microbiota composition, the mechanisms underlying this variation remain largely unknown. Bile acids (BAs) are hormones that are produced by the host and chemically modified by gut bacteria. BAs serve as environmental cues and nutrients to microbes, but they can also have antibacterial effects. We hypothesized that host genetic variation in BA metabolism and homeostasis influence gut microbiota composition. To address this, we used the Diversity Outbred (DO) stock, a population of genetically distinct mice derived from eight founder strains. We characterized the fecal microbiota composition and plasma and cecal BA profiles from 400 DO mice maintained on a high-fat high-sucrose diet for ~22 weeks. Using quantitative trait locus (QTL) analysis, we identified several genomic regions associated with variations in both bacterial and BA profiles. Notably, we found overlapping QTL for Turicibacter sp. and plasma cholic acid, which mapped to a locus containing the gene for the ileal bile acid transporter, Slc10a2. Mediation analysis and subsequent follow-up validation experiments suggest that differences in Slc10a2 gene expression associated with the different strains influences levels of both traits and revealed novel interactions between Turicibacter and BAs. This work illustrates how systems genetics can be utilized to generate testable hypotheses and provide insight into host-microbe interactions

    Transforming Environmental Water Management to Adapt to a Changing Climate

    Environmental water management has become a global imperative in response to environmental degradation and the growing recognition that human well-being and livelihoods are critically dependent on freshwater ecosystems and the ecological functions and services they provide. Although a wide range of techniques and strategies for planning and implementing environmental flows has developed, many remain based on assumptions of hydrologic stationarity, typically focusing on restoring freshwater ecosystems to pre-development or “natural” conditions. Climate change raises major challenges to this conventional approach, in part because of increasing uncertainties in patterns of water supply and demand. In such a rapidly changing world, the implementation of, and capacity of water managers to deliver flow regimes resembling historical hydrological patterns may be both unfeasible and undesirable. Additionally, as emphasis shifts from species-focused water allocation plans toward a greater appreciation of freshwater ecological functions and services, many of which will be influenced by climate change, a thorough re-evaluation of the conventional objectives, planning, delivery and monitoring of environmental water, including its role in the broader context of water and environmental management, is essential. Here, we identify the major challenges posed by climate change to environmental water management and discuss key adaptations and research needed to meet these challenges to achieve environmental and societal benefits and avoid maladaptation

    Genetic mapping of microbial and host traits reveals production of immunomodulatory lipids by Akkermansia muciniphila in the murine gut.

    The molecular bases of how host genetic variation impacts the gut microbiome remain largely unknown. Here we used a genetically diverse mouse population and applied systems genetics strategies to identify interactions between host and microbe phenotypes including microbial functions, using faecal metagenomics, small intestinal transcripts and caecal lipids that influence microbe-host dynamics. Quantitative trait locus (QTL) mapping identified murine genomic regions associated with variations in bacterial taxa; bacterial functions including motility, sporulation and lipopolysaccharide production and levels of bacterial- and host-derived lipids. We found overlapping QTL for the abundance of Akkermansia muciniphila and caecal levels of ornithine lipids. Follow-up in vitro and in vivo studies revealed that A. muciniphila is a major source of these lipids in the gut, provided evidence that ornithine lipids have immunomodulatory effects and identified intestinal transcripts co-regulated with these traits including Atf3, which encodes for a transcription factor that plays vital roles in modulating metabolism and immunity. Collectively, these results suggest that ornithine lipids are potentially important for A. muciniphila-host interactions and support the role of host genetics as a determinant of responses to gut microbes

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data
