    O estudo da diversidade biolĂłgica sofreu vĂĄrias modificaçÔes ao longo dos tempos. Desde Darwin todos os mĂ©todos para classificar os seres vivos deveriam abordar as relaçÔes evolutivas dos grupos estudados. Um desses mĂ©todos compreende a construção de cladogramas (grĂĄficos contendo hipĂłteses de relaçÔes filogenĂ©ticas). Com o emprego desse mĂ©todo e seu desenvolvimento as classificaçÔes biolĂłgicas tradicionais sofreram modificaçÔes profundas. Grupos como peixes, invertebrados e gimnospermas nĂŁo sĂŁo mais considerado grupos vĂĄlidos e tiveram profundas modificaçÔes em sua classificação. No ensino mĂ©dio no Brasil, o estudo de Zoologia e BotĂąnica geralmente ainda nĂŁo Ă© realizado dentro de um enfoque evolutivo, mas sim a partir dos modelos da classificação tradicional. Algumas experiĂȘncias demonstraram que o ensino de Zoologia e BotĂąnica no ensino mĂ©dio dentro de uma perspectiva evolucionĂĄria se torna mais dinĂąmico e interessante, pois aborda o conhecimento sobre a diversidade biolĂłgica considerando as hipĂłteses filogenĂ©ticas para propor modelos de classificação dos organismos

    Aptasensor for the detection of Moraxella catarrhalis adhesin UspA2

    Innovative point-of-care (PoC) diagnostic platforms are desirable to surpass the deficiencies of conventional laboratory diagnostic methods for bacterial infections and to tackle the growing antimicrobial resistance crisis. In this study, a workflow was implemented, comprising the identification of new aptamers with high affinity for the ubiquitous surface protein A2 (UspA2) of the bacterial pathogen Moraxella catarrhalis and the development of an electrochemical biosensor functionalized with the best-performing aptamer as a bioreceptor to detect UspA2. After cell-systematic evolution of ligands by exponential enrichment (cell-SELEX) was performed, next-generation sequencing was used to sequence the final aptamer pool. The most frequent aptamer sequences were further evaluated using bioinformatic tools. The two most promising aptamer candidates, Apt1 and Apt1_RC (Apt1 reverse complement), had Kd values of 214.4 and 3.4 nM, respectively. Finally, a simple and label-free electrochemical biosensor was functionalized with Apt1_RC. The aptasensor surface modifications were confirmed by impedance spectroscopy and cyclic voltammetry. The ability to detect UspA2 was evaluated by square wave voltammetry, exhibiting a linear detection range of 4.0 × 104–7.0 × 107 CFU mL−1, a square correlation coefficient superior to 0.99 and a limit of detection of 4.0 × 104 CFU mL−1 at pH 5.0.     Electrochemical aptasensor for the detection of the key virulence factor YadA of Yersinia enterocolitica

    New point-of-care (POC) diagnosis of bacterial infections are imperative to overcome the deficiencies of conventional methods, such as culture and molecular methods. In this study, we identified new aptamers that bind to the virulence factor Yersinia adhesin A (YadA) of Yersinia enterocolitica using cell-systematic evolution of ligands by exponential enrichment (cell-SELEX). Escherichia coli expressing YadA on the cell surface was used as a target cell. After eight cycles of selection, the final aptamer pool was sequenced by high throughput sequencing using the Illumina Novaseq platform. The sequencing data, analyzed using the Geneious software, was aligned, filtered and demultiplexed to obtain the key nucleotides possibly involved in the target binding. The most promising aptamer candidate, Apt1, bound specifically to YadA with a dissociation constant (Kd) of 11 nM. Apt1 was used to develop a simple electrochemical biosensor with a two-step, label-free design towards the detection of YadA. The sensor surface modifications and its ability to bind successfully and stably to YadA were confirmed by cyclic voltammetry, impedance spectroscopy and square wave voltammetry. The biosensor enabled the detection of YadA in a linear range between 7.0 × 104 and 7.0 × 107 CFU mL−1 and showed a square correlation coefficient >0.99. The standard deviation and the limit of detection was ~2.5% and 7.0 × 104 CFU mL−1, respectively. Overall, the results suggest that this novel biosensor incorporating Apt1 can potentially be used as a sensitive POC detection system to aid the diagnosis of Y. enterocolitica infections.     Is the WHO End TB Strategy at Risk?

However, the COVID-19 pandemic has severely impacted global public health and the strict measures to control the coronavirus spread can affect the management of other diseases, such as TB. Herein, we aimed to assess the impact of the COVID-19 pandemic on the diagnosis of TB in Brazil, during 2020. Methods: We carried out an ecological and population-based study, using spatial analysis techniques. The variables used were the new cases of TB, pulmonary tuberculosis (PTB), and also baciloscopy-positive (BP) cases in Brazil between 2015 and 2020. The percentage of changes (% change) was calculated to verify if there was an increase or decrease of TB cases in 2020, along with time trend analyses given by Joinpoint regression model. Also, interrupted time series analyses were used to assess the trend of TB diagnosis before and after the onset of the COVID-19 in Brazil. Spatial distribution maps were elaborated, considering the % change of each Brazilian state. Findings: Data analyses showed a reduction in the diagnosis of TB (−8.3%) and PTB (−8.1%) in Brazil after the irruption of the COVID-19 pandemic. Likewise, 22 states depicted a reduction in TB diagnosis. An expressive reduction of BP cases (−17.1%) was also observed. Interestingly, interrupted time series analysis showed decline in TB and PTB diagnoses from March 2020. Spatial analyses revealed that all states had a progressive reduction of TB, PTB and PB cases, from March on, with the highest percentages of reduction in December (−100% to −75%). Interpretation: Taken together, our analyses demonstrated a reduction in TB diagnosis after the irruption of the COVID-19 pandemic in Brazil and its regions, signaling a serious impact on the WHO “End TB Strategy” global plan.publishersversionpublishe

    O cuidado prestado por doulas em uma maternidade pĂșblica: o olhar das puĂ©rperas

    Objetivo: Analisar a percepção das mulheres sobre o cuidado prestado por doulas durante o trabalho de parto, o parto e pĂłs-parto imediato em uma maternidade pĂșblica de JoĂŁo Pessoa - PB. MĂ©todo: Estudo exploratĂłrio, descritivo, com abordagem qualitativa, realizado com oito puĂ©rperas, aprovado pelo ComitĂȘ de Ética em Pesquisa do CCS/UFPB, sob parecer 808.256. O material empĂ­rico foi produzido por meio de entrevistas semiestruturadas e analisado pela tĂ©cnica de anĂĄlise do discurso de Fiorin. Resultados: As puĂ©rperas compreendem o trabalho da doula como adjuvante para diminuição da dor e desconforto no trabalho de parto e no parto. Enunciaram que o vĂ­nculo estabelecido contribuiu para tornar o parto um momento positivo e afetuoso.     Development and validation of the Brazilian Version of Lay Persons’ Social Judgements about Cleft-lip Scale (B-LSojCleft-S)

    Abstract The aim of this study was to develop and analyze the psychometric properties of the Brazilian Version of Lay Person's Social Judgement about Cleft-lip Scale (B-LSojCleft-S). A standardized photograph of a 16-year-old girl was manipulated by using photo editing software, to simulate different cleft-lip situations. The cleft-free image was used as a control. The researchers structured an initial questionnaire that was evaluated by experts and a sample of the target population to evaluate the construct. After analysis by the researchers, a final version of the B-LSojCleft-S containing 14 items was generated. Each answer was awarded score from 1 to 3 points, yielding a total score of 14 to 42 points. Higher scores represented better social judgements. The B-LSojCleft-S was applied online to 103 test participants and 73 retest participants with an interval of 20 days between application. Aspects of acceptability, discriminant property, internal consistency, reliability and construct validity were evaluated. Construct validity was assessed using the Friedman test (p 0.70, and internal consistency using Cronbach's alpha > 0.81. The B-LSojCleft-S showed high acceptability, strong discriminant property, excellent internal consistency and reliability, but had a floor and ceiling effect. The instrument reached valid and reliable scores and had acceptable psychometric properties to evaluate the social judgments of lay persons about different cleft lip situations in a Brazilian population

MĂ©todos: Os testes foram feitos em triplicata e a Concentração InibitĂłria MĂ­nima foi determinada pelo mĂ©todo da microdiluição em caldo, onde as concentraçÔes testadas do extrato variaram de 1024 a 1 ”g/mL. O ensaio de modulação foi realizado utilizando a nistatina e o benzoilmetronidazol em concentraçÔes variando de 512 a 1 ”g/mL. Resultados: L. venustum apresentou atividade antifĂșngica contra Candida krusei e potencializador da ação do antifĂșngico nistatina frente a Candida tropicalis. ConclusĂŁo: O produto natural tem efeito sobrealgumas linhagens de fungos do gĂȘnero Candida, validando cientificamente o potencial antifĂșngico da espĂ©cie, junto ao uso popular.
