    Proyek Sekolah Tinggi Musik Klasik di Surabaya ini adalah sekolah tinggi yang khusus mengajarkan musik klasik, mulai dari teori, teknik bermain musik baik secara individu maupun berkelompok, serta pelajaran literatur yang diperlukan dalam pemahaman mahasiswa mengenai musik tertentu yang sedang dipelajari. Latar belakang lahirnya gagasan proyek ini adalah langkanya pemain musik klasik yang berkualitas, serta menurunnya peminat musik klasik di Indonesia. Oleh karena itu, melalui proyek ini, diharapkan akan lahir pemain musik yang lebih berkualitas dan mampu bersaing dengan dunia Internasional, serta menumbuhkan minat masyarakat terhadap musik klasik. Konsep yang digunakan dalam proyek ini adalah "perform", dimana tujuan utama dari seorang pemain musik klasik adalah untuk tampil dalam suatu pertunjukan orchestra. Dalam penampilan sebuah orkestra yang baik, dibutuhkan kesatuan dari banyak alat musik serta kesatuan pemahaman akan musik yang sedang dimainkan. Oleh karena itu, dapat dikatakan bahwa inti dari sebuah musik klasik adalah sebuah pertunjukan

    Catecholaminergic modulation of indices of cognitive flexibility:a pharmaco-tDCS study

    Background: Dopaminergic activity within the dorsolateral prefrontal cortex (dlPFC) has been implicated in the control of cognitive flexibility. Much of the evidence for a causative relationship between cognitive flexibility and dopamine has come from animal studies, whilst human data have largely been correlational. Objective/Hypothesis:The current study examines whether changes in dopamine levels through tyrosine administration and suppression of dlPFC activity via cathodal tDCS could be causally related to cognitive flexibility as measured by task switching and reversal learning. Methods: Using a crossover, double-blind, sham controlled, counterbalanced, randomized trial, we tested the effects of combining cathodal tDCS with tyrosine, a catecholaminergic precursor, with appropriate drug and tDCS placebo controls, on two measures of cognitive flexibility: probabilistic reversal learning, and task switching. Results: While none of the manipulations had an effect on task switching, there was a significant main effect of cathodal tDCS and tyrosine on reversal learning. Reversal learning performance was significantly worsened by cathodal tDCS compared with sham tDCS, whilst tyrosine significantly improved performance compared with placebo. However, there was no significant tDCS × drugs interaction. Interestingly, and as predicted by our model, the combined administration of tyrosine with cathodal tDCS resulted in performance that was equivalent to the control condition (i.e. tDCS sham + placebo). Conclusions: Our results suggest a causative role for dopamine signalling and dorsolateral prefrontal cortex activity in regulating indices of cognitive flexibility in humans

    Surgical approaches for lung volume reduction in emphysema

    Lung volume reduction surgery (LVRS) for chronic obstructive pulmonary disease (COPD) is recommended in both British and international guidelines because trials have shown improvement in survival in selected patients with poor baseline exercise capacity and upper lobe-predominant emphysema. Despite this, few procedures are carried out, possibly because of historical concerns about high levels of morbidity and mortality associated with the operation. The authors reviewed data on lung volume reduction procedures at their institution between January 2000 and September 2012. There were no deaths within 90 days of unilateral LVRS (n=81), bullectomy (n=20) or intracavity drainage procedures (n=14). These data suggest that concerns about surgical mortality should not discourage LVRS in selected patients with COPD, provided that it is undertaken within a multidisciplinary team environment involving appropriate patient selection. © Royal College of Physicians 2014.     Folic acid supplementation normalizes the endothelial progenitor cell transcriptome of patients with type 1 diabetes: a case-control pilot study

    Background: Endothelial progenitor cells play an important role in vascular wall repair. Patients with type 1 diabetes have reduced levels of endothelial progenitor cells of which their functional capacity is impaired. Reduced nitric oxide bioavailability and increased oxidative stress play a role in endothelial progenitor cell dysfunction in these patients. Folic acid, a B-vitamin with anti-oxidant properties, may be able to improve endothelial progenitor cell function. In this study, we investigated the gene expression profiles of endothelial progenitor cells from patients with type 1 diabetes compared to endothelial progenitor cells from healthy subjects. Furthermore, we studied the effect of folic acid on gene expression profiles of endothelial progenitor cells from patients with type 1 diabetes. Methods: We used microarray analysis to investigate the gene expression profiles of endothelial progenitor cells from type 1 diabetes patients before (n = 11) and after a four week period of folic acid supplementation (n = 10) compared to the gene expression profiles of endothelial progenitor cells from healthy subjects (n = 11). The probability of genes being differentially expressed among the classes was computed using a random-variance t-test. A multivariate permutation test was used to identify genes that were differentially expressed among the two classes. Functional classification of differentially expressed genes was performed using the biological process ontology in the Gene Ontology database. Results: Type 1 diabetes significantly modulated the expression of 1591 genes compared to healthy controls. These genes were found to be involved in processes regulating development, cell communication, cell adhesion and localization. After folic acid treatment, endothelial progenitor cell gene expression profiles from diabetic patients were similar to those from healthy controls. Genes that were normalized by folic acid played a prominent role in development, such as the transcription factors ID1 and MAFF. Few oxidative-stress related genes were affected by folic acid. Conclusion: Folic acid normalizes endothelial progenitor cell gene expression profiles of patients with type 1 diabetes. Signaling pathways modulated by folic acid may be potential therapeutic targets to improve endothelial progenitor cell function

    The architecture of Cidec-mediated interfaces between lipid droplets.

    Lipid droplets (LDs) are intracellular organelles responsible for storing surplus energy as neutral lipids. Their size and number vary enormously. In white adipocytes, LDs can reach 100 μm in diameter, occupying >90% of the cell. Cidec, which is strictly required for the formation of large LDs, is concentrated at interfaces between adjacent LDs and facilitates directional flux of neutral lipids from the smaller to the larger LD. The mechanism of lipid transfer is unclear, in part because the architecture of interfaces between LDs remains elusive. Here we visualize interfaces between LDs by electron cryo-tomography and analyze the kinetics of lipid transfer by quantitative live fluorescence microscopy. We show that transfer occurs through closely apposed monolayers, is slowed down by increasing the distance between the monolayers, and follows exponential kinetics. Our data corroborate the notion that Cidec facilitates pressure-driven transfer of neutral lipids through two "leaky" monolayers between LDs

    Protistan Communities Within the Galápagos Archipelago With an Emphasis on Micrograzers

    The Galápagos Archipelago is a globally significant biodiversity hotspot. However, compared to the relatively well-known megafauna, the distribution and ecological significance of marine protists in this system are poorly understood. To gain an understanding of the protistan assemblages across trophic modes, an intensive oceanographic survey was conducted in the Galápagos Marine Reserve (GMR) in October of 2018. The Equatorial Undercurrent (EUC)-influenced region had higher chlorophyll-a (Chl-a) concentrations than those of the eastern regions of the archipelago, along with higher abundances of protistan grazers. Specifically, proportions of autotrophic and potentially mixotrophic dinoflagellates were higher in the EUC, whereas in the eastern regions, heterotrophic dinoflagellates and chlorophytes dominated. Taxonomic composition and biochemical indicators suggested proportions of micrograzers and their associated heterotrophic biomass was higher in the oligotrophic, low Chl-a regions in the east. We also report observations from a dinoflagellate bloom in the western archipelago, which was heavily influenced by upwelling of the EUC. The red tide-forming dinoflagellate Scrippsiella lachrymosa was highly detected through light microscopy and DNA amplicon sequencing. In addition, the heterotrophic dinoflagellate Polykrikos kofoidii was detected and, based on cell densities observed in this study and grazing rates obtained from the literature, estimated to potentially graze up to 62% of S. lachrymosa bloom population. Our findings thus provide new insights into the composition of micrograzers and their potential roles in structuring protistan communities in the Galápagos Archipelago

Surrounding issues come from various factors such as the usage of fossil fuels that lead to global warming and climate change which slowly assassinate human beings. This article will focus on efficiency comparison between using paper money, plastic cards and cryptocurrency. The main goal is to prove which of the three financial payments is the most energy-efficient in the past, present, and future. The parameters used in this study are electricity usage and CO2 emissions. The whole analysis is based on a quantitative-descriptive method which provides in depth analysis of electricity usage and CO2 emissions level among financial equates from every generation. Even though plastic cards have the highest efficiency level initially, over time, cryptocurrency is going revolutionary, making them more efficient than paper money and plastic cards. This proves that cryptocurrency could be the most energy-efficient payment method in the future

    Loss of RAF kinase inhibitor protein is involved in myelomonocytic differentiation and aggravates RAS-driven myeloid leukemogenesis

    RAS-signaling mutations induce the myelomonocytic differentiation and proliferation of hematopoietic stem and progenitor cells. Moreover, they are important players in the development of myeloid neoplasias. RAF kinase inhibitor protein (RKIP) is a negative regulator of RAS-signaling. As RKIP loss has recently been described in RAS-mutated myelomonocytic acute myeloid leukemia, we now aimed to analyze its role in myelomonocytic differentiation and RAS-driven leukemogenesis. Therefore, we initially analyzed RKIP expression during human and murine hematopoietic differentiation and observed that it is high in hematopoietic stem and progenitor cells and lymphoid cells but decreases in cells belonging to the myeloid lineage. By employing short hairpin RNA knockdown experiments in CD34+ umbilical cord blood cells and the undifferentiated acute myeloid leukemia cell line HL-60, we show that RKIP loss is indeed functionally involved in myelomonocytic lineage commitment and drives the myelomonocytic differentiation of hematopoietic stem and progenitor cells. These results could be confirmed in vivo, where Rkip deletion induced a myelomonocytic differentiation bias in mice by amplifying the effects of granulocyte macrophage-colony-stimulating factor. We further show that RKIP is of relevance for RAS-driven myelomonocytic leukemogenesis by demonstrating that Rkip deletion aggravates the development of a myeloproliferative disease in NrasG12D-mutated mice. Mechanistically, we demonstrate that RKIP loss increases the activity of the RAS-MAPK/ERK signaling module. Finally, we prove the clinical relevance of these findings by showing that RKIP loss is a frequent event in chronic myelomonocytic leukemia, and that it co-occurs with RAS-signaling mutations. Taken together, these data establish RKIP as novel player in RAS-driven myeloid leukemogenesis

    Characterizing the Near-infrared Spectra of Flares from TRAPPIST-1 During JWST Transit Spectroscopy Observations

    We present the first analysis of JWST near-infrared spectroscopy of stellar flares from TRAPPIST-1 during transits of rocky exoplanets. Four flares were observed from 0.6--2.8 μ\mum with NIRISS and 0.6--3.5 μ\mum with NIRSpec during transits of TRAPPIST-1b, f, and g. We discover Pα\alpha and Brβ\beta line emission and characterize flare continuum at wavelengths from 1--3.5 μ\mum for the first time. Observed lines include Hα\alpha, Pα\alpha-Pϵ\epsilon, Brβ\beta, He I λ\lambda0.7062μ\mum, two Ca II infrared triplet (IRT) lines, and the He I IRT. We observe a reversed Paschen decrement from Pα\alpha-Pγ\gamma alongside changes in the light curve shapes of these lines. The continuum of all four flares is well-described by blackbody emission with an effective temperature below 5300 K, lower than temperatures typically observed at optical wavelengths. The 0.6--1 μ\mum spectra were convolved with the TESS response, enabling us to measure the flare rate of TRAPPIST-1 in the TESS bandpass. We find flares of 1030^{30} erg large enough to impact transit spectra occur at a rate of 3.6+2.11.3\substack{+2.1 \\ -1.3} flare d1^{-1}, \sim10×\times higher than previous predictions from K2. We measure the amount of flare contamination at 2 μ\mum for the TRAPPIST-1b and f transits to be 500±\pm450 and 2100±\pm400 ppm, respectively. We find up to 80% of flare contamination can be removed, with mitigation most effective from 1.0--2.4 μ\mum.
