    Fucoxanthin Enhances Cisplatin-Induced Cytotoxicity via NFκB-Mediated Pathway and Downregulates DNA Repair Gene Expression in Human Hepatoma HepG2 Cells

    Cisplain, a platinum-containing anticancer drug, has been shown to enhance DNA repair and to inhibit cell apoptosis, leading to drug resistance. Thus, the combination of anticancer drugs with nutritional factors is a potential strategy for improving the efficacy of cisplatin chemotherapy. In this study, we investigated the anti-proliferative effects of a combination of fucoxanthin, the major non-provitamin A carotenoid found in Undaria Pinnatifida, and cisplatin in human hepatoma HepG2 cells. We found that fucoxanthin (1–10 μΜ) pretreatment for 24 h followed by cisplatin (10 μΜ) for 24 h significantly decreased cell proliferation, as compared with cisplatin treatment alone. Mechanistically, we showed that fucoxanthin attenuated cisplatin-induced NFκB expression and enhanced the NFκB-regulated Bax/Bcl-2 mRNA ratio. Cisplatin alone induced mRNA expression of excision repair cross complementation 1 (ERCC1) and thymidine phosphorylase (TP) through phosphorylation of ERK, p38 and PI3K/AKT pathways. However, fucoxanthin pretreatment significantly attenuated cisplatin-induced ERCC1 and TP mRNA expression, leading to improvement of chemotherapeutic efficacy of cisplatin. The results suggest that a combined treatment with fucoxanthin and cisplatin could lead to a potentially important new therapeutic strategy against human hepatoma cells

    Spontaneous regression of cancer is a rare, but well documented, phenomenon. We present a unique case of an 82 year old Chinese male who experienced spontaneous regression of histologically-verified metastatic type II papillary renal cell carcinoma in the absence of intervening systemic therapy or surgery. This is the first reported case of spontaneous regression of papillary renal cell carcinoma. The mechanism of spontaneous regression remains unknown, and represents a challenge for existing oncology paradigms

    Sponges from North Borneo and their bioactivity against human colorectal cancer cells

    Sponges are major source of numerous cytotoxic compounds that are used for defence as well as adaptation to the environment. Numerous studies have discovered compounds from sponge extracts that were effective against a wide range of cancer cells. In this study, a total of 23 sponges comprising of 19 species were collected from Northeast Borneo. Sponges were treated and extracted using modified Folch extraction method, followed by cytotoxicity assay to determine their effectiveness against different colorectal cancer cells. Our results demonstrate that Monanchora clathrata, Dysidea sp., and Jaspis sp. possess different degrees of cytotoxicity against a wide range of human colorectal cancer cells. Monanchora clathrata (KDT07), Dysidea sp. (KDT09), and Jaspis sp. (KDT18) are among the demosponges which possess significant cytotoxicity against colorectal cancer cell lines, including HCT116, LoVo, SW480, and SW620. KDT08 and KDT21 which fall under the same genus Dysidea, possess insignificant cytotoxicity against colorectal cancer cells suggested environmental factors (symbiotic organisms) play a role in biosynthesizing bioactive compounds. Presented results suggested the importance of intensifying research on isolating and purifying natural products from marine sponges for useful applications

    Impact of Social Media On Consumer Purchase Behaviour During COVID-19 Pandemic

    Social media has been used by people to create and share content with the public. Consumers have changed their purchasing habits from offline to online purchases during the COVID-19 pandemic. Social media is also changing sellers' and buyers' specialised strategies. This study aimed to study the factors influencing consumer purchasing behaviour via social media during the COVID-19 outbreak. 215 young adults from three Malaysian states which are Johor, Melaka, and Selangor, have been invited to participate in this study by answering a set of questionnaires. The successful response rate is 93.02%, where 200 completed questionnaires were used for data analysis in this study. The four independent variables, price, convenience, product variety, and risk, are tested in relation to the dependent variable, consumer purchase behaviour during the COVID-19 outbreak. The measurement items of all the four independent variables and dependent variables were adopted from the past research studies. The collected data were entered into SPSS version 26 and then tested using Partial Least Square Structural Equation Modelling (PLS-SEM 3.3.3). All hypotheses were found to be supported by the analysis results. Lastly, the findings of this study will help marketers understand consumer purchasing behaviour via social media during the COVID-19 pandemic

    Bioactivity-guided isolation of anticancer agents from Bauhinia kockiana Korth

    Background: Flowers of Bauhinia kockiana were investigated for their anticancer properties. Methods: Gallic acid (1), and methyl gallate (2), were isolated via bioassay-directed isolation, and they exhibited anticancer properties towards several cancer cell lines, examined using MTT cell viability assay. Pyrogallol (3) was examined against the same cancer cell lines to deduce the bioactive functional group of the phenolic compounds. Results: The results showed that the phenolic compounds could exhibit moderate to weak cytotoxicity towards certain cell lines (GI50 30 - 86 µM), but were inactive towards DU145 prostate cancer cell (GI50 > 100 µM). Conclusion: It was observed that pyrogallol moiety was one of the essential functional structures of the phenolic compounds in exhibiting anticancer activity. Also, the carboxyl group of compound 1 was also important in anticancer activity. Examination of the PC-3 cells treated with compound 1 using fluorescence microscopy showed that PC-3 cells were killed by apoptosis

    Sesamin: A Naturally Occurring Lignan Inhibits CYP3A4 by Antagonizing the Pregnane X Receptor Activation

    Inconsistent expression and regulation of drug-metabolizing enzymes (DMEs) are common causes of adverse drug effects in some drugs with a narrow therapeutic index (TI). An important cytochrome, cytochrome P450 3A4 (CYP3A4), is predominantly regulated by a nuclear receptor, pregnane X receptor (PXR). Sesamin, a major lignan constituent in sesame seeds and oil, exhibits a variety of biological functions; however, the effect of sesamin on the modulation of CYP3A4 is not well understood. In this study, the effects of sesamin on the PXR-CYP3A4 pathway were characterized, as well as the underlying mechanisms of those effects. Sesamin potently attenuated CYP3A4 induction in a dose-dependent manner by blocking the activation of PXR. The PXR inducer-mediated inhibition of CYP3A4 was further evidenced by the ability of sesamin to attenuate the effects of several PXR ligands in the CYP3A4 reporter assay. Further mechanistic studies showed that sesamin inhibited PXR by interrupting the interacting with coregulators. These results may lead to the development of new therapeutic and dietary approaches to reduce the frequency of inducer-drug interaction. Sesamin was established as a novel inhibitor of PXR and may be useful for modulating DMEs expression and drug efficacies. Modification of CYP3A4 expression and activity by consumption of sesamin may have important implications for drug safety
