Epitope recognition of peptide-imprinted polymers for Regenerating protein 1 (REG1)

Molecularly imprinted polymers (MIPs) were developed to replace natural antibodies with a cost-effective and durable synthetic material. Molecular imprinting of proteins conventionally utilizes the whole protein as the template, which is complex (as many different epitopes may be imprinted) and expensive. In this work, seven peptides (13–18 amino acids) were synthesized and used as templates for the imprinting and recognition of Regenerating Protein 1 (REG1). REG1 is involved in the proliferation and differentiation of diverse cell types, and was recently described as a urinary biomarker for pancreatic ductal adenocarcinoma (PDAC). Peptide-imprinted poly(ethylene-co-vinyl alcohol)s (PIPs), containing four different mole fractions of ethylene were cast on screen-printed electrodes to find the optimum composition for both the sensing and the extraction of REG1 in an E. coli culture medium. Peptides with fewer than 16 amino acids and two or three aromatic and hydrophobic groups have a higher affinity for MIPs of poly(ethylene-co-vinyl alcohol) (EVAL) with 27 mol% of ethylene, while those with four aromatic and hydrophobic groups have a higher affinity for MIPs with EVALs that contain 32 mol% of ethylene. The peptide / EVAL combination that maximized both imprinting effectiveness and response to REG1B was the sequence NEDRETWVDADLY imprinted into 32 mol% EVAL. This EVAL composition and template peptide were then modified by incorporation of magnetic nanoparticles, thus extending applications for PIPs to include extraction of REG1 protein from E. coli culture medium

Pose selection for animated scenes and a case study of bas-relief generation

This paper aims to automate the process of generating a meaningful single still image from a temporal input of scene sequences. The success of our extraction relies on evaluating the optimal pose of characters selection, which should maximize the information conveyed. We define the information entropy of the still image candidates as the evaluation criteria. To validate our method and to demonstrate its effectiveness, we generated a relief (as a unique form of art creation) to narrate given temporal action scenes. A user study was conducted to experimentally compare the computer-selected poses with those selected by human participants. The results show that the proposed method can assist the selection of informative pose of character effectively

Lepton Mixing Matrix in Standard Model Extended by One Sterile Neutrino

We consider the simplest extension of the standard electroweak model by one sterile neutrino that allows for neutrino masses and mixing. We find that its leptonic sector contains much less free physical parameters than previously realized. In addition to the two neutrino masses, the lepton mixing matrix in charged current interactions involves (n-1) free physical mixing angles for n generations. The mixing matrix in neutral current interactions of neutrinos is completely fixed by the two masses. Both interactions conserve CP. We illustrate the phenomenological implications of the model by vacuum neutrino oscillations, tritium beta decay and neutrinoless double beta decay. It turns out that, due to the revealed specific structure in its mixing matrix, the model with any n generations cannot accommodate simultaneously the data by KamLAND, K2K and CHOOZ.Comment: 14 pages, no figures; version 2: (1) added a short paragraph at the end of subsec 2.2 to record the counting of physical parameters for any numbers of generations and sterile neutrinos for completeness; (2) added a note in ref list, item [18] to quote and comment on an earlier work; (3) added the second paper to ref list, item [17]; (4) fixed typo

The tomato cytochrome P450 CYP712G1 catalyzes the double oxidation of orobanchol <i>en route</i> to the rhizosphere signaling strigolactone, solanacol

Strigolactones (SLs) are rhizosphere signalling molecules and phytohormones. The biosynthetic pathway of SLs in tomato has been partially elucidated, but the structural diversity in tomato SLs predicts that additional biosynthetic steps are required. Here, root RNA-seq data and co-expression analysis were used for SL biosynthetic gene discovery. This strategy resulted in a candidate gene list containing several cytochrome P450s. Heterologous expression in Nicotiana benthamiana and yeast showed that one of these, CYP712G1, can catalyse the double oxidation of orobanchol, resulting in the formation of three didehydro-orobanchol (DDH) isomers. Virus-induced gene silencing and heterologous expression in yeast showed that one of these DDH isomers is converted to solanacol, one of the most abundant SLs in tomato root exudate. Protein modelling and substrate docking analysis suggest that hydroxy-orbanchol is the likely intermediate in the conversion from orobanchol to the DDH isomers. Phylogenetic analysis demonstrated the occurrence of CYP712G1 homologues in the Eudicots only, which fits with the reports on DDH isomers in that clade. Protein modelling and orobanchol docking of the putative tobacco CYP712G1 homologue suggest that it can convert orobanchol to similar DDH isomers as tomato

By analyzing the data sets of 17.3, 6.5 and 1.0 pb1^{-1} taken, respectively, at s=3.773\sqrt s= 3.773, 3.650 and 3.6648 GeV with the BES-II detector at the BEPC collider, we measure the observed cross sections for e+eπ+ππ0π0e^+e^-\to \pi^+\pi^-\pi^0\pi^0, K+Kπ0π0K^+K^-\pi^0\pi^0, 2(π+ππ0)2(\pi^+\pi^-\pi^0), K+Kπ+ππ0π0K^+K^-\pi^+\pi^-\pi^0\pi^0 and 3(π+π)π0π03(\pi^+\pi^-)\pi^0\pi^0 at the three energy points. Based on these cross sections we set the upper limits on the observed cross sections and the branching fractions for ψ(3770)\psi(3770) decay into these final states at 90% C.L..Comment: 7 pages, 2 figure

    Using 5.8×107J/ψ5.8 \times 10^7 J/\psi events collected in the BESII detector, the radiative decay J/ψγϕϕγK+KKS0KL0J/\psi \to \gamma \phi \phi \to \gamma K^+ K^- K^0_S K^0_L is studied. The ϕϕ\phi\phi invariant mass distribution exhibits a near-threshold enhancement that peaks around 2.24 GeV/c2c^{2}. A partial wave analysis shows that the structure is dominated by a 0+0^{-+} state (η(2225)\eta(2225)) with a mass of 2.240.02+0.030.02+0.032.24^{+0.03}_{-0.02}{}^{+0.03}_{-0.02} GeV/c2c^{2} and a width of 0.19±0.030.04+0.060.19 \pm 0.03^{+0.06}_{-0.04} GeV/c2c^{2}. The product branching fraction is: Br(J/ψγη(2225))Br(η(2225)ϕϕ)=(4.4±0.4±0.8)×104Br(J/\psi \to \gamma \eta(2225))\cdot Br(\eta(2225)\to \phi\phi) = (4.4 \pm 0.4 \pm 0.8)\times 10^{-4}.Comment: 11 pages, 4 figures. corrected proof for journa
