A microfluidic 2×2 optical switch
A 2×2 microfluidic-based optical switch is proposed and demonstrated. The switch is made of an optically clear silicon elastomer, Polydimethylsiloxane (PDMS), using soft lithography. It has insertion loss smaller than 1 dB and extinction ratio on the order of 20 dB. The device is switching between transmission (bypass) and reflection (exchange) modes within less than 20 m
Language models align with human judgments on key grammatical constructions
Do Large Language Models (LLMs) make human-like linguistic generalizations?
Dentella et al. (2023; "DGL") prompt several LLMs ("Is the following sentence
grammatically correct in English?") to elicit grammaticality judgments of 80
English sentences, concluding that LLMs demonstrate a "yes-response bias" and a
"failure to distinguish grammatical from ungrammatical sentences". We
re-evaluate LLM performance using well-established practices and find that
DGL's data in fact provide evidence for just how well LLMs capture human
behaviors. Models not only achieve high accuracy overall, but also capture
fine-grained variation in human linguistic judgments.Comment: Response to Dentella et al. (2023

Splaying Preorders and Postorders
Let be a binary search tree. We prove two results about the behavior of
the Splay algorithm (Sleator and Tarjan 1985). Our first result is that
inserting keys into an empty binary search tree via splaying in the order of
either 's preorder or 's postorder takes linear time. Our proof uses the
fact that preorders and postorders are pattern-avoiding: i.e. they contain no
subsequences that are order-isomorphic to and ,
respectively. Pattern-avoidance implies certain constraints on the manner in
which items are inserted. We exploit this structure with a simple potential
function that counts inserted nodes lying on access paths to uninserted nodes.
Our methods can likely be extended to permutations that avoid more general
patterns. Second, if is any other binary search tree with the same keys as
and is weight-balanced (Nievergelt and Reingold 1973), then splaying
's preorder sequence or 's postorder sequence starting from takes
linear time. To prove this, we demonstrate that preorders and postorders of
balanced search trees do not contain many large "jumps" in symmetric order, and
exploit this fact by using the dynamic finger theorem (Cole et al. 2000). Both
of our results provide further evidence in favor of the elusive "dynamic
optimality conjecture.
The mammalian LINC complex regulates genome transcriptional responses to substrate rigidity
Mechanical integration of the nucleus with the extracellular matrix (ECM) is established by linkage between the cytoskeleton and the nucleus. This integration is hypothesized to mediate sensing of ECM rigidity, but parsing the function of nucleus-cytoskeleton linkage from other mechanisms has remained a central challenge. Here we took advantage of the fact that the LINC (linker of nucleoskeleton and cytoskeleton) complex is a known molecular linker of the nucleus to the cytoskeleton, and asked how it regulates the sensitivity of genome-wide transcription to substratum rigidity. We show that gene mechanosensitivity is preserved after LINC disruption, but reversed in direction. Combined with myosin inhibition studies, we identify genes that depend on nuclear tension for their regulation. We also show that LINC disruption does not attenuate nuclear shape sensitivity to substrate rigidity. Our results show for the first time that the LINC complex facilitates mechano-regulation of expression across the genome
Sept8/SEPTIN8 involvement in cellular structure and kidney damage is identified by genetic mapping and a novel human tubule hypoxic model.
Chronic kidney disease (CKD), which can ultimately progress to kidney failure, is influenced by genetics and the environment. Genes identified in human genome wide association studies (GWAS) explain only a small proportion of the heritable variation and lack functional validation, indicating the need for additional model systems. Outbred heterogeneous stock (HS) rats have been used for genetic fine-mapping of complex traits, but have not previously been used for CKD traits. We performed GWAS for urinary protein excretion (UPE) and CKD related serum biochemistries in 245 male HS rats. Quantitative trait loci (QTL) were identified using a linear mixed effect model that tested for association with imputed genotypes. Candidate genes were identified using bioinformatics tools and targeted RNAseq followed by testing in a novel in vitro model of human tubule, hypoxia-induced damage. We identified two QTL for UPE and five for serum biochemistries. Protein modeling identified a missense variant within Septin 8 (Sept8) as a candidate for UPE. Sept8/SEPTIN8 expression increased in HS rats with elevated UPE and tubulointerstitial injury and in the in vitro hypoxia model. SEPTIN8 is detected within proximal tubule cells in human kidney samples and localizes with acetyl-alpha tubulin in the culture system. After hypoxia, SEPTIN8 staining becomes diffuse and appears to relocalize with actin. These data suggest a role of SEPTIN8 in cellular organization and structure in response to environmental stress. This study demonstrates that integration of a rat genetic model with an environmentally induced tubule damage system identifies Sept8/SEPTIN8 and informs novel aspects of the complex gene by environmental interactions contributing to CKD risk
Sun exposure among teenage and young adult cancer survivors in the United Kingdom
Skin cancers are a common form of second malignant neoplasm among teenage and young adult cancer survivors (TYACS). The Children's Oncology Group specifies that TYACS should adhere to safe sun practices and be screened for skin cancer annually. Cross-sectional self-report data collected by our group indicate over a third of TYACS (n = 229; mean age: 19.8 years) intentionally sunbathe, with many reporting sunburn. TYACS sunbathing, sunburn, and sunbed use are similar to the general population (P > 0.05). These data suggest TYACS require intervention to limit sun exposure and improve their sun safety habits
Status Report of the DPHEP Study Group: Towards a Global Effort for Sustainable Data Preservation in High Energy Physics
Data from high-energy physics (HEP) experiments are collected with
significant financial and human effort and are mostly unique. An
inter-experimental study group on HEP data preservation and long-term analysis
was convened as a panel of the International Committee for Future Accelerators
(ICFA). The group was formed by large collider-based experiments and
investigated the technical and organisational aspects of HEP data preservation.
An intermediate report was released in November 2009 addressing the general
issues of data preservation in HEP. This paper includes and extends the
intermediate report. It provides an analysis of the research case for data
preservation and a detailed description of the various projects at experiment,
laboratory and international levels. In addition, the paper provides a concrete
proposal for an international organisation in charge of the data management and
policies in high-energy physics
Spatial Analysis of Environmental Factors Related to Lyme Disease in Alabama by Means of NASA Earth Observation Systems
This slide presentation reviews the epidemiology of Lyme Disease that accounts for more than 95% or vector borne diseases in the United States. The history, symptoms and the life cycle of the tick, the transmitting agent of Lyme Disease, a map that shows the cases reported to the CDC between1990 and 2006 and the number of cases in Alabama by year from 1986 to 2007. A NASA project is described, the goals of which are to (1) Demonstrate the presence of the chain of infection of Lyme disease in Alabama (2) Identify areas with environmental factors that support tick population using NASA Earth Observation Systems data in selected areas of Alabama and (3) Increase community awareness of Lyme disease and recommend primary and secondary prevention strategies. The remote sensing methods included: Analyzed Advanced Spaceborne Thermal Emission and Reflection Radiometer (ASTER) and DigitalGlobe Quickbird satellite imagery from summer months and Performed image analyses in ER Mapper 7.1. Views from the ASTER and Quickbird land cover are shown, the Normalized Difference Vegetation Index (NDVI) algorithm was applied to all ASTER and Quickbird imagery. The use of the images to obtain the level of soil moisture is reviewed, and this analysis was used along with the NDVI, was used to identify the areas that support the tick population
