Целью работы является- проектирование технологического решения для бурения эксплуатационной наклонно-направленной скважины с горизонтальным участком глубиной 3894 метров. Для достижения цели были поставлены следующие задачи: 1. Детальный анализ общей геологической части месторождения; 2. Проектирование актуальной конструкции скважины для данных геологических условий; 3. Проектирование оптимального оборудования для бурения скважины параметров и свойств бурового раствора; 4. Проектирование процессов закачивания скважины; 5. Финансовый расчёт сметной стоимости бурения и крепления скважины; 6. Анализ вредных факторов на производстве и их влияние на человека.The object of the research is an operational directional borehole with a horizontal section 3894 meters deep. The purpose of the work is to design a technological solution for drilling an operational directional borehole with a horizontal section with a depth of 3894 meters. To achieve this goal, the following tasks were set: 1. A detailed analysis of the general geological part of the field; 2. Designing the actual design of the hole for given geological conditions; 3. Designing optimal equipment for drilling borehole parameters and properties of the drilling fluid; 4. Design of well injection processes; 5. Financial calculation of the estimated cost of drilling and borehole fixing; 6. Analysis of harmful factors in the workplace and their impact on humans

    A systematic review of non-productivity-related animal-based indicators of heat stress resilience in dairy cattle.

    INTRODUCTION:Projected temperature rise in the upcoming years due to climate change has increased interest in studying the effects of heat stress in dairy cows. Environmental indices are commonly used for detecting heat stress, but have been used mainly in studies focused on the productivity-related effects of heat stress. The welfare approach involves identifying physiological and behavioural measurements so as to start heat stress mitigation protocols before the appearance of impending severe health or production issues. Therefore, there is growing interest in studying the effects of heat stress on welfare. This systematic review seeks to summarise the animal-based responses to heat stress (physiological and behavioural, excluding productivity) that have been used in scientific literature. METHODS:Using systematic review guidelines set by PRISMA, research articles were identified, screened and summarised based on inclusion criteria for physiology and behaviour, excluding productivity, for animal-based resilience indicators. 129 published articles were reviewed to determine which animal-based indicators for heat stress were most frequently used in dairy cows. RESULTS:The articles considered report at least 212 different animal-based indicators that can be aggregated into body temperature, feeding, physiological response, resting, drinking, grazing and pasture-related behaviour, reactions to heat management and others. The most common physiological animal-based indicators are rectal temperature, respiration rate and dry matter intake, while the most common behavioural indicators are time spent lying, standing and feeding. CONCLUSION:Although body temperature and respiration rate are the animal-based indicators most frequently used to assess heat stress in dairy cattle, when choosing an animal-based indicator for detecting heat stress using scientific literature to establish thresholds, characteristics that influence the scale of the response and the definition of heat stress must be taken into account, e.g. breed, lactation stage, milk yield, system type, climate region, bedding type, diet and cooling management strategies

    Polymyxin B Conjugates with Bio-Inspired Synthetic Polymers of Different Nature

    The emergence and growth of bacterial resistance to antibiotics poses an enormous threat to humanity in the future. In this regard, the discovery of new antibiotics and the improvement of existing ones is a priority task. In this study, we proposed the synthesis of new polymeric conjugates of polymyxin B, which is a clinically approved but limited-use peptide antibiotic. In particular, three carboxylate-bearing polymers and one synthetic glycopolymer were selected for conjugation with polymyxin B (PMX B), namely, poly(α,L-glutamic acid) (PGlu), copolymer of L-glutamic acid and L-phenylalanine (P(Glu-co-Phe)), copolymer of N-vinyl succinamic acid and N-vinylsuccinimide (P(VSAA-co-VSI)), and poly(2-deoxy-2-methacrylamido-D-glucose) (PMAG). Unlike PGlu and PMAG, P(Glu-co-Phe) and P(VSAA-co-VSI) are amphiphilic and form nanoparticles in aqueous media. A number of conjugates with different polymyxin B loading were synthesized and characterized. In addition, the complex conjugates of PGLu or PMAG with polymyxin B and deferoxamine (siderophore) were obtained. A release of PMX B from Schiff base and amide-linked polymer conjugates was studied in model buffer media with pH 7.4 and 5.8. In both cases, a more pronounced release was observed under slightly acidic conditions. The cytotoxicity of free polymers and PMX B as well as their conjugates was examined in human embryonic kidney cells (HEK 293T cell line). All conjugates demonstrated reduced cytotoxicity compared to the free antibiotic. Finally, the antimicrobial efficacy of the conjugates against Pseudomonas aeruginosa was determined and compared. The lowest values of minimum inhibitory concentrations (MIC) were observed for polymyxin B and polymyxin B/deferoxamine conjugated with PMAG. Among the polymers tested, PMAG appears to be the most promising carrier for delivery of PMX B in conjugated form due to the good preservation of the antimicrobial properties of PMX B and the ability of controlled drug release

    Self-Assembled Nanoparticles Based on Block-Copolymers of Poly(2-Deoxy-2-methacrylamido-d-glucose)/Poly(N-Vinyl Succinamic Acid) with Poly(O-Cholesteryl Methacrylate) for Delivery of Hydrophobic Drugs

    The self-assembly of amphiphilic block-copolymers is a convenient way to obtain soft nanomaterials of different morphology and scale. In turn, the use of a biomimetic approach makes it possible to synthesize polymers with fragments similar to natural macromolecules but more resistant to biodegradation. In this study, we synthesized the novel bio-inspired amphiphilic block-copolymers consisting of poly(N-methacrylamido-d-glucose) or poly(N-vinyl succinamic acid) as a hydrophilic fragment and poly(O-cholesteryl methacrylate) as a hydrophobic fragment. Block-copolymers were synthesized by radical addition–fragmentation chain-transfer (RAFT) polymerization using dithiobenzoate or trithiocarbonate chain-transfer agent depending on the first monomer, further forming the hydrophilic block. Both homopolymers and copolymers were characterized by 1H NMR and Fourier transform infrared spectroscopy, as well as thermogravimetric analysis. The obtained copolymers had low dispersity (1.05–1.37) and molecular weights in the range of ~13,000–32,000. The amphiphilic copolymers demonstrated enhanced thermal stability in comparison with hydrophilic precursors. According to dynamic light scattering and nanoparticle tracking analysis, the obtained amphiphilic copolymers were able to self-assemble in aqueous media into nanoparticles with a hydrodynamic diameter of approximately 200 nm. An investigation of nanoparticles by transmission electron microscopy revealed their spherical shape. The obtained nanoparticles did not demonstrate cytotoxicity against human embryonic kidney (HEK293) and bronchial epithelial (BEAS-2B) cells, and they were characterized by a low uptake by macrophages in vitro.     CT-based temperature monitoring during hepatic RF ablation: Feasibility in an animal model

    Purpose: The aim of this paper was to establish non-invasive CT-based temperature monitoring during hepatic radiofrequency (RF) ablation in an ex vivo porcine model followed by transfer of the technique into a feasibility in vivo experiment. Materials and methods: Bipolar RF ablations were performed in 10 specimens of porcine liver. Parallel to the needle-shaped RF applicator three optical temperature probes were inserted into the liver specimens at fixed distances of 5, 10 and 15mm from the RF probe. During energy application (20W) unenhanced sequential MSCT scans were acquired using the following scan protocol: 140 kV tube voltage, 300 mAs/rotation tube current time product, collimation 24 x 1.2 mm, rotation time 0.5 s. Axial image data was reconstructed using a soft tissue convolution kernel. Temperature data was recorded during every CT scan. Using a circular 0.5 cm(2) region of interest local CT values were measured at the tips of the temperature probes and matched with the measured temperatures. Regression analysis was performed to analyse the relationship between local temperatures and CT values for each temperature probe position. Furthermore, the same experimental design was used in four anaesthetised female pigs in order to investigate the potential of this technique for an in vivo application. Results: A negative correlation was found for the relationship between temperature and CT value. Regression coefficients were -0.44 (5 mm), -0.35 (10mm) and -0.37 (15mm) for ex vivo data. Analysis of in vivo experiments showed regression coefficients between -0.025 and -0.434. Conclusion: Multislice computed tomography is able to depict temperature changes in liver tissue during RFA
