    PARTITION OF THE ORGANOCHLORINE INSECTICIDE LINDANE INTO THE HUMAN SPERM SURFACE INDUCES MEMBRANE DEPOLARIZATION AND CALCIUM INFLUX

    The effects of the insecticide lindane (the gamma-isomer of 1,2,3,4,5,6-hexachlorocyclohexane) on membrane potential, cytosolic free Ca2+ concentration ([Ca2+]i) and surface biophysical properties were studied in human spermatozoa. The insecticide induces rapid, transient and reproducible membrane depolarization and opening of voltage-dependent Ca2+ channels leading to an increase in [Ca2+]i. In contrast with the effect in somatic cells, lindane did not affect gamma-aminobutyric acid receptor-linked Cl- currents. Ca2+ and K+ currents were found to drive lindane-induced membrane depolarization and repolarization respectively, whereas Na+ and Cl- fluxes appear not to have a role in the phenomenon. The insecticide was still able to produce membrane depolarization both in the combined absence of extracellular Ca2+ and Na+ and in high-K+ buffer, suggesting that lindane alters the membrane dipole potential. In agreement with this, Laurodan and Prodan fluorescence spectroscopy revealed that lindane partition into the sperm plasma membrane lowers water molecular dynamics in the uppermost region of the membrane external leaflet, probably as the result of reordering of water dipoles. We propose that the first effect of lindane partitioning into the sperm plasma membrane is a change in the membrane dipole potential, which results in the activation of membrane-located Ca2+-influx pathways

    Shocks near Jamming

    Non-linear sound is an extreme phenomenon typically observed in solids after violent explosions. But granular media are different. Right when they jam, these fragile and disordered solids exhibit a vanishing rigidity and sound speed, so that even tiny mechanical perturbations form supersonic shocks. Here, we perform simulations in which two-dimensional jammed granular packings are dynamically compressed, and demonstrate that the elementary excitations are strongly non-linear shocks, rather than ordinary phonons. We capture the full dependence of the shock speed on pressure and impact intensity by a surprisingly simple analytical model.Comment: Revised version. Accepted for publication in Phys. Rev. Let

    Late pleistocene carnivores (Carnivora: Mammalia) from a cave sedimentary deposit in northern Brazil

    The Brazilian Quaternary terrestrial Carnivora are represented by the following families: Canidae, Felidae, Ursidae, Procyonidae Mephitidae and Mustelidae. Their recent evolutionary history in South America is associated with the uplift of the Panamanian Isthmus, and which enabled the Great American Biotic Interchange (GABI). Here we present new fossil records of Carnivora found in a cave in Aurora do Tocantins, Tocantins, northern Brazil. A stratigraphical controlled collection in the sedimentary deposit of the studied cave revealed a fossiliferous level where the following Carnivora taxa were present: Panthera onca, Leopardus sp., Galictis cuja, Procyon cancrivorus, Nasua nasua and Arctotherium wingei. Dating by Electron Spinning Resonance indicates that this assemblage was deposited during the Last Glacial Maximum (LGM), at least, 22.000 YBP. The weasel, G. cuja, is currently reported much further south than the record presented here. This may suggest that the environment around the cave was relatively drier during the LGM, with more open vegetation, and more moderate temperatures than the current Brazilian Cerrado.Facultad de Ciencias Naturales y Muse

    A Kolmogorov-Zakharov Spectrum in AdSAdS Gravitational Collapse

    We study black hole formation during the gravitational collapse of a massless scalar field in asymptotically AdSDAdS_D spacetimes for D=4,5D=4,5. We conclude that spherically symmetric gravitational collapse in asymptotically AdSAdS spaces is turbulent and characterized by a Kolmogorov-Zakharov spectrum. Namely, we find that after an initial period of weakly nonlinear evolution, there is a regime where the power spectrum of the Ricci scalar evolves as ωs\omega^{-s} with the frequency, ω\omega, and s1.7±0.1s\approx 1.7\pm 0.1.Comment: 5 pages, 4 figures. v2: Typos, other initial profile considered for universality, error analysis, close to PRL versio

    Stochastic pulse coded arithmetic

    Among the different pulse codification techniques, stochastic pulse codification has its own arithmetic based on the similarity between Boolean algebra and statistical algebra. Summation and multiplication are the two basic arithmetic operations treated in depth in the literature. In this paper we present two digital stochastic circuits that extend traditional stochastic algebra: a division circuit and a square-root circuit, and the interfaces between the analog and stochastic domain. As a result, we are able to process analog input signals with a simple and complete processing system. These circuits can be implemented in low-cost and low-power digital programmable devices

    Hadronic Density of States from String Theory

    Exactly soluble string theories describing a particular hadronic sector of certain confining gauge theories have been obtained recently as Penrose-Gueven limits of the dual supergravity backgrounds. The effect of taking the Penrose-Gueven limit on the gravity side translates, in the gauge theory side, into an effective truncation to hadrons of large U(1) charge (annulons). We present an exact calculation of the finite temperature partition function for the hadronic states corresponding to a Penrose-Gueven limit of the Maldacena-Nunez embedding of N=1 SYM into string theory. It is established that the theory exhibits a Hagedorn density of states. Motivated by this exact calculation we propose a semiclassical string approximation to the finite temperature partition function for confining gauge theories admitting a supergravity dual, by performing an expansion around classical solutions characterized by temporal windings. This semiclassical approximation reveals a hadronic energy density of states of Hagedorn type, with the coefficient determined by the gauge theory string tension as expected for confining theories. We argue that our proposal captures primarily information about states of pure N=1 SYM, given that this semiclassical approximation does not entail a projection onto states of large U(1) charge.Comment: 15 page

    Notes are provided on the morphology of males and females of the enigmatic Lithurgus huberi Ducke (Lithurginae: Lithurgini), a species historically believed to have been introduced into South America from Asia and to be a possible synonym of the more widespread L. atratus Smith. Distinctive differences are documented between L. huberi and L. atratus, perhaps indicative of separate species.     Multi‐target directed ligands (Mtdls) binding the σ1 receptor as promising therapeutics: State of the art and perspectives

    The sigma‐1 (σ1) receptor is a ‘pluripotent chaperone’ protein mainly expressed at the mitochondria–endoplasmic reticulum membrane interfaces where it interacts with several client proteins. This feature renders the σ1 receptor an ideal target for the development of multifunctional ligands, whose benefits are now recognized because several pathologies are multifactorial. Indeed, the current therapeutic regimens are based on the administration of different classes of drugs in order to counteract the diverse unbalanced physiological pathways associated with the pathology. Thus, the multi‐targeted directed ligand (MTDL) approach, with one molecule that exerts polypharmacological actions, may be a winning strategy that overcomes the pharmacokinetic issues linked to the administration of diverse drugs. This review aims to point out the progress in the development of MTDLs directed toward σ1 receptors for the treatment of central nervous system (CNS) and cancer diseases, with a focus on the perspectives that are proper for this strategy. The evidence that some drugs in clinical use unintentionally bind the σ1 protein (as off‐target) provides a proof of concept of the potential of this strategy, and it strongly supports the promise that the σ1 receptor holds as a target to be hit in the context of MTDLs for the therapy of multifactorial pathologies

    HPLC-UV and LC-MS/MS analysis to study formulation and long-term stability of some anticancer drugs in elastomeric pumps

    Drug stability evaluations in elastomeric pump represent the first step to certify the safety of a therapeutic treatment in oncology. Since the stability of several anticancer molecules is due to their reactivity and stability in elastomeric pumps, made up of different materials, several experimental conditions, such as temperature, pH, concentration and possible chemical interactions among drugs in a single formulation, should be always investigated. Galenic preparation of anticancer drugs is an important prerogative of Anticancer Units within hospital pharmacies and, considering the burden of COVID-19 pandemic event, specific guidelines for therapeutic administration in elastomeric pumps reducing hospitalization both for post-surgical treatment and for therapeutic treatment have been worldwide elaborated. In the present study, the stability of Doxorubicin and Vincristine as a single formulation at different experimental conditions has been investigated. Moreover, we report a systematic study of 5-FU, which is known to be largely used in these medical devices, although its criticisms in terms of solubility, pH effect, storage time and conditions. Our results demonstrate that doxorubicin and vincristine can be mixed safely as a single formulation and 5-FU is stable for 32 days at different temperatures and concentrations in elastomeric pumps
