    Effectiveness of Air Cleaning Systems in Crushing Zones of Dressing Mills

    Body sensor networks (BSN) are an important research topic due to various advantages over conventional measurement equipment. One main advantage is the feasibility to deploy a BSN system for 24/7 health monitoring applications. The requirements for such an application are miniaturization of the network nodes and the use of wireless data transmission technologies to ensure wearability and ease of use. Therefore, the reliability of such a system depends on the quality of the wireless data transmission. At present, most BSNs use ZigBee or other IEEE 802.15.4 based transmission technologies. Here, we evaluated the performance of a wireless transmission system of a novel BSN for biomedical applications in the 433 MHz ISM band, called Integrated Posture and Activity NEtwork by Medit Aachen (IPANEMA) BSN. The 433 MHz ISM band is used mostly by implanted sensors and thus allows easy integration of such into the BSN. Multiple measurement scenarios have been assessed, including varying antenna orientations, transmission distances and the number of network participants. The mean packet loss rate (PLR) was 0.63% for a single slave, which is comparable to IEEE 802.15.4 BSNs in the proximity of Bluetooth or WiFi networks. Secondly, an enhanced version is evaluated during on-body measurements with five slaves. The mean PLR results show a comparable good performance for measurements on a treadmill (2.5%), an outdoor track (3.4%) and in a climate chamber (1.5%)

    DNA methylation patterns change dynamically during mammalian development and lineage specification, yet scarce information is available about how DNA methylation affects gene expression profiles upon differentiation. Here we determine genome-wide transcription profiles during undirected differentiation of severely hypomethylated (Dnmt1⁻/⁻) embryonic stem cells (ESCs) as well as ESCs completely devoid of DNA methylation (Dnmt1⁻/⁻;Dnmt3a⁻/⁻;Dnmt3b⁻/⁻ or TKO) and assay their potential to transit in and out of the ESC state. We find that the expression of only few genes mainly associated with germ line function and the X chromosome is affected in undifferentiated TKO ESCs. Upon initial differentiation as embryoid bodies (EBs) wild type, Dnmt1⁻/⁻ and TKO cells downregulate pluripotency associated genes and upregulate lineage specific genes, but their transcription profiles progressively diverge upon prolonged EB culture. While Oct4 protein levels are completely and homogeneously suppressed, transcription of Oct4 and Nanog is not completely silenced even at late stages in both Dnmt1⁻/⁻ and TKO EBs. Despite late wild type and Dnmt1⁻/⁻ EBs showing a much higher degree of concordant expression, after EB dissociation and replating under pluripotency promoting conditions both Dnmt1⁻/⁻ and TKO cells, but not wild type cells rapidly revert to expression profiles typical of undifferentiated ESCs. Thus, while DNA methylation seems not to be critical for initial activation of differentiation programs, it is crucial for permanent restriction of developmental fate during differentiation

    Stability analysis of chemically modified mRNA using micropattern-based single-cell arrays

    The measurement of mRNA turnover in living cells plays an important role in the search for stable mRNA constructs for RNA-based therapies. Here we show that automated time-lapse microscopy combined with micropatterned arrays allows for efficient high-throughput monitoring of fluorescent reporter protein expression at the single-cell level. The fluorescence time courses after mRNA transfection yield the distribution of individual mRNA expression and degradation rates within a population. We compare mRNA constructs with combinations of 5′ and 3′ UTR sequences and find a systematic broadening and shift towards longer functional half-lives for UTR stabilized mRNA. At the same time the life time distribution of the destabilized EGFP reporter protein was found to be constant and narrowly distributed. Using mathematical modeling, we show that mRNA functional life-time predicts the time-integrated protein level, i.e. the area under the curve (AUC) of mRNA translation. Our approach paves the way for quantitative assessment of hitherto unexplored mRNA functional life time heterogeneity, possibly predicated on multiple mRNA secondary structures and its dependence on UTR sequences

    Quantum Computing with Continuous-Variable Clusters

    Continuous-variable cluster states offer a potentially promising method of implementing a quantum computer. This paper extends and further refines theoretical foundations and protocols for experimental implementation. We give a cluster-state implementation of the cubic phase gate through photon detection, which, together with homodyne detection, facilitates universal quantum computation. In addition, we characterize the offline squeezed resources required to generate an arbitrary graph state through passive linear optics. Most significantly, we prove that there are universal states for which the offline squeezing per mode does not increase with the size of the cluster. Simple representations of continuous-variable graph states are introduced to analyze graph state transformations under measurement and the existence of universal continuous-variable resource states.Comment: 17 pages, 5 figure

    In a highly globalised world where all production and consumption activities are internationally intertwined and the environmental consequences of those actions are hard to identify, rethinking the role of work in our societies according to sustainability principles is a complex but highly necessary task. Salaried work has become one of the crucial indicators to analyse any country in the world. By looking at the proportion of the population that is employed, the working conditions they have, and how productive they are when performing their tasks, it is possible to produce an image of a country's society to assist in the understanding of the levels of well-being of its citizens. Work and labour markets not only largely structure the way the economy and society function, they also heavily influence an individual's life satisfaction and happiness; virtually the entire life of a person is designed around their work. Given the relevance work has at all levels, diving into the concept of sustainable work is a crucial project due to the urgency of environmental matters. The biggest role humanity faces is how to transform our societies so that they are sustainable from a social, ecological and economic perspective. For the sustainable society vision, work would need to be drastically altered in order to adapt it to the multi-dimensional sustainability requirements. This research aims to contribute to this enterprise by identifying the conditions that define the sustainability of work and then present an overview of seven European countries from this perspective. The present document introduces our conceptualisation of work and explains its main components. These are designed around the idea of the sustainable society and are composed of individuals' needs, equity and planetary boundaries. The final section concludes and introduces the different country-case studies.Series: WWWforEurop
