    Modelling wave-induced current at haeundae beach on orthogonal curvilinear grid by using CST3D

    Haeundae Beach is sandy beach at Busan, Korea. Haeundae Beach is about 1.5 km- long enclosed by two headlands. The normal direction of the beach is between south and south-east. Tidal current at the site is not negligible, and waves are relatively high during monsoon and winter seasons. Currents were measured at nine points along Haeundae Beach on 4 June 2008 by using GPS-equipped drogues. Bathymetry around Haeundae Beach was surveyed on 7 August 2007 and 12 November 2007 by using an echo-sounder; wave, tide, and tidal current were measured between the two survey days, so that bathymetric change at the beach was obtained from the two surveys. The measured current vectors may include wave- induced current element, tidal current element, and wind-induced current. Seasonal sediment transport pattern at Haeundae Beach, Busan, Korea is known to be simple, according to previous observation. Typically sediment moves eastward, while sediment moves westward in winter. Tidal current contributes to long-term bathymetric change at the beach by transporting small-sized sediment

    Synaptic Pattern of KA1 and KA2 upon the Direction-Selective Ganglion Cells in Developing and Adult Mouse Retina

    The detection of image motion is important to vision. Direction-selective retinal ganglion cells (DS-RGCs) respond strongly to stimuli moving in one direction of motion and are strongly inhibited by stimuli moving in the opposite direction. In this article, we investigated the distributions of kainate glutamate receptor subtypes KA1 and KA2 on the dendritic arbors of DS-RGCs in developing (5, 10) days postnatal (PN) and adult mouse retina to search for anisotropies. The distribution of kainate receptor subtypes on the DS-RGCs was determined using antibody immunocytochemistry. To identify their characteristic morphology, DS-RGCs were injected with Lucifer yellow. The triple-labeled images of dendrites, kinesin II, and receptors were visualized by confocal microscopy and were reconstructed from high-resolution confocal images. We found no evidence of asymmetry in any of the kainate receptor subunits examined on the dendritic arbors of both the On and Off layers of DS-RGCs in all periods of developing and adult stage that would predict direction selectivity

    Abstract Live algae carrying hydrophobic xenobiotics can be an effective vector candidate for the chemical translocation to filter feeders in the laboratory toxicity test, but information on their application is lacking. Time-course uptake and elimination of polychlorinated biphenyls (PCBs) (0, 50, 100, and 500 ng g -1 ) by two key algal foods, Isochrysis galbana and Tetraselmis suecica, were measured. Both of the algae achieved maximum concentration in an hour after PCBs exposure regardless the chemical concentrations in our time-course measurements (0, 1, 5, 10, 24, 48 and 72 hrs). Once achieved the maximum concentration, the algae shortly exhibited elimination or eliminating tendency depending on the chemical concentrations. Algae exposed to the chemical for 1 and 24 hrs (hereafter 1 and 24 hr vectors, respectively) were then evaluated as a chemical translocation vector by feeding test to larval and spat Crassostrea gigas. In the feeding test the 24 hr vector, which contained lower chemicals than the 1-hr vector, appeared to be more damaging the early lives of the oyster. This was particularly significant for vectors of higher PCBs (p<0.05), probably due to algal reduction in food value by the prolonged chemical stress. These findings imply that 1 hr exposure is long enough for a generation of algal vector for laboratory toxicity test, minimizing data error resulted from reduction in food value by longer chemical stress

    Collagen Immobilization on Ultra-thin Nanofiber Membrane to Promote In Vitro Endothelial Monolayer Formation

    The endothelialization on the poly (epsilon-caprolactone) nanofiber has been limited due to its low hydrophilicity. The aim of this study was to immobilize collagen on an ultra-thin poly (epsilon-caprolactone) nanofiber membrane without altering the nanofiber structure and maintaining the endothelial cell homeostasis on it. We immobilized collagen on the poly (epsilon-caprolactone) nanofiber using hydrolysis by NaOH treatment and 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide/sulfo-N-hydroxysulfosuccinimide reaction as a cost-effective and stable approach. NaOH was first applied to render the poly (epsilon-caprolactone) nanofiber hydrophilic. Subsequently, collagen was immobilized on the surface of the poly (epsilon-caprolactone) nanofibers using 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide/sulfo-N-hydroxysulfosuccinimide. Scanning electron microscopy, Fourier transform infrared spectroscopy, transmission electron microscopy, and fluorescence microscopy were used to verify stable collagen immobilization on the surface of the poly (epsilon-caprolactone) nanofibers and the maintenance of the original structure of poly (epsilon-caprolactone) nanofibers. Furthermore, human endothelial cells were cultured on the collagen-immobilized poly (epsilon-caprolactone) nanofiber membrane and expressed tight junction proteins with the increase in transendothelial electrical resistance, which demonstrated the maintenance of the endothelial cell homeostasis on the collagen-immobilized-poly (epsilon-caprolactone) nanofiber membrane.     Intra-wave-phase cross-shore profile modelling by using boundary-fitted slowly moving grid

    Coastal bed profile change is described by bed load, pick-up, and settling on a boundary-fitted moving grid. Existing bed load formula is modified by changing threshold bed shear stress to reflect local bed slope. A numerical model system adopting the above function is developed to simulate cross-shore sediment transport around swash zone with steep bed slope as well as surf zone. The model system adopts a moving boundary grid which fits bed boundary slope, and other horizontal grid lines are parallel to the bed grid line. The model system is composed of flow module and sediment transport module. The flow module solves continuity equation and Reynolds-average Navier-Stokes momentum equations in intra-wave-phase manner. The flow module provides detailed flow information including near-bed fluid velocity which varies asymmetrically within a regular wave period near sea-bed and wave-phase average undertow profile of main fluid body including wave boundary layer. The sediment transport module solves sediment mass conservation equation. Exchange of sediment mass between bed itself and fluid column containing suspended sediment happens through pick-up and deposition. Wild bed level undulation is controlled by using a smoothing method. Model system is applied to two extreme cases of sand experiments by Kajima et al., and reasonable agreements between measurements and computations are obtained for both onshore-dominant and offshore-dominant cases

    A functional regulatory variant of MYH3 influences muscle fiber-type composition and intramuscular fat content in pigs

    Muscle development and lipid accumulation in muscle critically affect meat quality of livestock. However, the genetic factors underlying myofiber-type specification and intramuscular fat (IMF) accumulation remain to be elucidated. Using two independent intercrosses between Western commercial breeds and Korean native pigs (KNPs) and a joint linkage-linkage disequilibrium analysis, we identified a 488.1-kb region on porcine chromosome 12 that affects both reddish meat color (a*) and IMF. In this critical region, only the MYH3 gene, encoding myosin heavy chain 3, was found to be preferentially overexpressed in the skeletal muscle of KNPs. Subsequently, MYH3-transgenic mice demonstrated that this gene controls both myofiber-type specification and adipogenesis in skeletal muscle. We discovered a structural variant in the promotor/regulatory region of MYH3 for which Q allele carriers exhibited significantly higher values of a* and IMF than q allele carriers. Furthermore, chromatin immunoprecipitation and cotransfection assays showed that the structural variant in the 5-flanking region of MYH3 abrogated the binding of the myogenic regulatory factors (MYF5, MYOD, MYOG, and MRF4). The allele distribution of MYH3 among pig populations worldwide indicated that the MYH3 Q allele is of Asian origin and likely predates domestication. In conclusion, we identified a functional regulatory sequence variant in porcine MYH3 that provides novel insights into the genetic basis of the regulation of myofiber type ratios and associated changes in IMF in pigs. The MYH3 variant can play an important role in improving pork quality in current breeding programs

    Clinical Features of Probable Cluster Headache: A Prospective, Cross-Sectional Multicenter Study

    Background: Epidemiological data of probable cluster headaches (CH) are scarce in the relevant literature. Here, we sought to assess the prevalence and clinical characteristics of probable CH in comparison with definite CH.Methods: Data used in this study were obtained from the Korean Cluster Headache Registry (KCHR), a prospective, cross-sectional, multicenter headache registry that collected data from consecutive patients diagnosed with CH.Results: In total, 159 patients were enrolled in this study; 20 (12.6%) were diagnosed with probable CH. The most common unfulfilled criterion in patients with probable CH was the duration of attack, which was found in 40% of patients with probable CH. Among clinical characteristics, the number of autonomic symptoms tended to be lower in probable CH than in definite CH (1.7 ± 1.2 vs. 2.4 ± 1.5, p = 0.051) and conjunctival injection and lacrimation showed an increased odds ratio (OR) [OR = 3.03; 95% confidence interval (CI): 1.03–8.33] in definite CH. The groups did not differ with regard to baseline demographic characteristics, disability, impact on life, or treatment response.Conclusions: Probable CH is relatively common among CH disorders, with a clinical impact similar to that of definite CH
