    The Safety After Fifty Evaluation trial: Evaluation of the safety and efficacy of antihypertensive therapy with metoprolol in patients 50 to 75 years of age: Study design

    Hypertension increases in prevalence with advancing age and is a major risk factor for the development of cardiovascular disease in elderly patients. However, the presence of coexisting illness, altered drug metabolism, enhanced susceptibility to drug side effects, and physiologic changes such as reduced plasma volume and lower plasma renin levels make truatment of hypertension in elderly patients more difficult. Nonetheless, several studies have now demonstrated the beneficial effects of antihypertensive drug therapy in older parlents. The Safety After Fifty Evaluation trial was designed to determine the short-term efficacy and tolerabillty of once-daily therapy with the cardioselective [beta]-blocker metoprolol anone or in combination with hydrochlorothiazide in the treatment of mild hypertension in patients 50 to 75 years of age. A total of 24,816 patients were enrolled in the trial by 2821 practicing physicians from across the United States. This article describes the detalls of the Safety After Fiffy Evaluation study design.     System for Secure Integration of Aviation Data

    The Aviation Data Integration System (ADIS) of Ames Research Center has been established to promote analysis of aviation data by airlines and other interested users for purposes of enhancing the quality (especially safety) of flight operations. The ADIS is a system of computer hardware and software for collecting, integrating, and disseminating aviation data pertaining to flights and specified flight events that involve one or more airline(s). The ADIS is secure in the sense that care is taken to ensure the integrity of sources of collected data and to verify the authorizations of requesters to receive data. Most importantly, the ADIS removes a disincentive to collection and exchange of useful data by providing for automatic removal of information that could be used to identify specific flights and crewmembers. Such information, denoted sensitive information, includes flight data (here signifying data collected by sensors aboard an aircraft during flight), weather data for a specified route on a specified date, date and time, and any other information traceable to a specific flight. The removal of information that could be used to perform such tracing is called "deidentification." Airlines are often reluctant to keep flight data in identifiable form because of concerns about loss of anonymity. Hence, one of the things needed to promote retention and analysis of aviation data is an automated means of de-identification of archived flight data to enable integration of flight data with non-flight aviation data while preserving anonymity. Preferably, such an automated means would enable end users of the data to continue to use pre-existing data-analysis software to identify anomalies in flight data without identifying a specific anomalous flight. It would then also be possible to perform statistical analyses of integrated data. These needs are satisfied by the ADIS, which enables an end user to request aviation data associated with de-identified flight data. The ADIS includes client software integrated with other software running on flight-operations quality-assurance (FOQA) computers for purposes of analyzing data to study specified types of events or exceedences (departures of flight parameters from normal ranges). In addition to ADIS client software, ADIS includes server hardware and software that provide services to the ADIS clients via the Internet (see figure). The ADIS server receives and integrates flight and non-flight data pertaining to flights from multiple sources. The server accepts data updates from authorized sources only and responds to requests from authorized users only. In order to satisfy security requirements established by the airlines, (1) an ADIS client must not be accessible from the Internet by an unauthorized user and (2) non-flight data as airport terminal information system (ATIS) and weather data must be displayed without any identifying flight information. ADIS hardware and software architecture as well as encryption and data display scheme are designed to meet these requirements. When a user requests one or more selected aviation data characteristics associated with an event (e.g., a collision, near miss, equipment malfunction, or exceedence), the ADIS client augments the request with date and time information from encrypted files and submits the augmented request to the server.     Stratified inclined duct: direct numerical simulations

    The stratified inclined duct (SID) experiment consists of a zero-net-volume exchange flow in a long tilted rectangular duct, which allows the study of realistic stratified shear flows with sustained internal forcing. We present the first three-dimensional direct numerical simulations (DNS) of SID to explore the transitions between increasingly turbulent flow regimes first described by Meyer \& Linden (\textit{J. Fluid Mech.} \textbf{753}, 242-253, 2014). We develop a numerical set-up that faithfully reproduces the experiments and sustains the flow for arbitrarily long times at minimal computational cost. We recover the four qualitative flow regimes found experimentally in the same regions of parameter space: laminar flow, waves, intermittent turbulence, and fully-developed turbulence. We find good qualitative and quantitative agreement between DNS and experiments and highlight the added value of DNS to complement experimental diagnostics and increase our understanding of the transition to turbulence, both temporally (laminar/turbulent cycles) and parametrically (as the tilt angle of the duct and the Reynolds number are increased). These results demonstrate that numerical studies of SID -- and deeper integration between simulations and experiments -- have the potential to lead to a better understanding of stratified turbulence in environmental flows

    Systemic Disease-Induced Salivary Biomarker Profiles in Mouse Models of Melanoma and Non-Small Cell Lung Cancer

    Background: Saliva (oral fluids) is an emerging biofluid poised for detection of clinical diseases. Although the rationale for oral diseases applications (e.g. oral cancer) is intuitive, the rationale and relationship between systemic diseases and saliva biomarkers are unclear. Methodology/Principal Findings: In this study, we used mouse models of melanoma and non-small cell lung cancer and compared the transcriptome biomarker profiles of tumor-bearing mice to those of control mice. Microarray analysis showed that salivary transcriptomes were significantly altered in tumor-bearing mice vs. controls. Significant overlapping among transcriptomes of mouse tumors, serum, salivary glands and saliva suggests that salivary biomarkers have multiple origins. Furthermore, we identified that the expression of two groups of significantly altered transcription factors (TFs) Runx1, Mlxipl, Trim30 and Egr1, Tbx1, Nr1d1 in salivary gland tissue of melanoma-bearing mice can potentially be responsible for 82.6 % of the up-regulated gene expression and 62.5 % of the down-regulated gene expression, respectively, in the saliva o

    PAndAS' cubs: discovery of two new dwarf galaxies in the surroundings of the Andromeda and Triangulum galaxies

    We present the discovery of two new dwarf galaxies, Andromeda XXI and Andromeda XXII, located in the surroundings of the Andromeda and Triangulum galaxies (M31 and M33). These discoveries stem from the first year data of the Pan-Andromeda Archaeological Survey (PAndAS), a photometric survey of the M31/M33 group conducted with the Megaprime/MegaCam wide-field camera mounted on the Canada-France-Hawaii Telescope. Both satellites appear as spatial overdensities of stars which, when plotted in a color-magnitude diagram, follow metal-poor, [Fe/H]=-1.8, red giant branches at the distance of M31/M33. Andromeda XXI is a moderately bright dwarf galaxy (M_V=-9.9+/-0.6), albeit with low surface brightness, emphasizing again that many relatively luminous M31 satellites still remain to be discovered. It is also a large satellite, with a half-light radius close to 1 kpc, making it the fourth largest Local Group dwarf spheroidal galaxy after the recently discovered Andromeda XIX, Andromeda II and Sagittarius around the Milky Way, and supports the trend that M31 satellites are larger than their Milky Way counterparts. Andromeda XXII is much fainter (M_V=-6.5+/-0.8) and lies a lot closer in projection to M33 than it does to M31 (42 vs. 224 kpc), suggesting that it could be the first Triangulum satellite to be discovered. Although this is a very exciting possibility in the context of a past interaction of M33 with M31 and the fate of its satellite system, a confirmation will have to await a good distance estimate to confirm its physical proximity to M33. Along with the dwarf galaxies found in previous surveys of the M31 surroundings, these two new satellites bring the number of dwarf spheroidal galaxies in this region to 20.Comment: 10 pages, 6 figures, accepted for publication in ApJ; v2: minor typographical correction
