Insulin Receptor Isoform A and Insulin-like Growth Factor II as Additional Treatment Targets in Human Osteosarcoma
Abstract
Despite the frequent presence of an insulin-like growth factor I receptor (IGFIR)-mediated autocrine loop in osteosarcoma (OS), interfering with this target was only moderately effective in preclinical studies. Here, we considered other members of the IGF system that might be involved in the molecular pathology of OS. We found that, among 45 patients with OS, IGF-I and IGFBP-3 serum levels were significantly lower, and IGF-II serum levels significantly higher, than healthy controls. Increased IGF-II values were associated with a decreased disease-free survival. After tumor removal, both IGF-I and IGF-II levels returned to normal values. In 23 of 45 patients, we obtained tissue specimens and found that all expressed high mRNA level of IGF-II and >IGF-I. Also, isoform A of the insulin receptor (IR-A) was expressed at high level in addition to IGFIR and IR-A/IGFIR hybrids receptors (HRA). These receptors were also expressed in OS cell lines, and simultaneous impairment of IGFIR, IR, and Hybrid-Rs by monoclonal antibodies, siRNA, or the tyrosine kinase inhibitor BMS-536924, which blocks both IGFIR and IR, was more effective than selective anti-IGFIR strategies. Also, anti–IGF-II-siRNA treatment in low-serum conditions significantly inhibited MG-63 OS cells that have an autocrine circuit for IGF-II. In summary, IGF-II rather than IGF-I is the predominant growth factor produced by OS cells, and three different receptors (IR-A, HRA, and IGFIR) act complementarily for an IGF-II–mediated constitutive autocrine loop, in addition to the previously shown IGFIR/IGF-I circuit. “I luoghi di Mercalli”: a travelling exhibition as a tool for scientists to dialogue with the public on volcanoes and earthquakes
On March 19, 1914 Giuseppe Mercalli, a seismologist and volcanologist, well-known around the world for the
Intensity scale of earthquakes bearing his name, died tragically. A hundred years after, the Istituto Nazionale di
Geofisica e Vulcanologia (INGV) has promoted a variety of activities and cultural events that will take place under the
Patronage of the President of the Italian Republic within a year, the so called “Anno Mercalliano” (the Year of
Mercalli).
The opening ceremony took place in Naples, Italy, on March 19, 2014, in the Convitto Nazionale Vittorio Emanuele
II. A scientific conference was held with the participation of experts from INGV and the university of Milano –
Bicocca, and presentations of students. On that day the exhibition entitled “I luoghi di Mercalli” (Mercalli's places) was
also inaugurated, at the presence of local authorities.
The exhibition, organized by INGV, was realized in collaboration with the high school Vittorio Emanuele II, where
Mercalli has been teaching for 19 years, and the Università degli Studi Suor Orsola Benincasa, where he was professor
of natural sciences.
A biographical and geographical description of the places where Mercalli operated introduces the exhibition, which
is organized in sections:
- Mercalli educator (he taught at high schools in Reggio Calabria and Naples);
- Mercalli volcanologist (Mercalli studied Vesuvius volcanic activity for more than twenty years, he was a scientific
witness of the Vesuvius 1906 eruption, and of the eruptions occurred at Vulcano (1888-90) and Stromboli (1891)
islands.
- Mercalli seismologist (Mercalli Intensity scale definition, based on his experience as witness of catastrophic
earthquakes, such as Casamicciola in 1883 and Messina in 1908).
Another section deals with the Vesuvius Observatory, directed by Mercalli between 1911 and 1914, and the
description of the three active volcanoes of the Campania region (Vesuvius, Campi Flegrei and Ischia island), which
have been the subject of studies by the well-known scientist. The exhibition is enriched by documents, manuscripts,
photos and field notebooks of Mercalli.
It is not intended to be only a celebratory exhibition; rather it is designed as a tool for dissemination of scientific
culture and to raise awareness about seismic and volcanic hazards.
In the exhibition path a continuous thread between the figure of Mercalli as a researcher and the role of an Earth
Science researcher today is highlighted, pointing to the development of scientific knowledge in the past century. The
goal is to improve the capability of learning from the disasters occurred in the past to implement preventive actions to
safely deal with future events.

Immunological signatures unveiled by integrative systems vaccinology characterization of dengue vaccination trials and natural infection
Introduction: Dengue virus infection is a global health problem lacking specific therapy, requiring an improved understanding of DENV immunity and vaccine responses. Considering the recent emerging of new dengue vaccines, here we performed an integrative systems vaccinology characterization of molecular signatures triggered by the natural DENV infection (NDI) and attenuated dengue virus infection models (DVTs). Methods and results: We analyzed 955 samples of transcriptomic datasets of patients with NDI and attenuated dengue virus infection trials (DVT1, DVT2, and DVT3) using a systems vaccinology approach. Differential expression analysis identified 237 common differentially expressed genes (DEGs) between DVTs and NDI. Among them, 28 and 60 DEGs were up or downregulated by dengue vaccination during DVT2 and DVT3, respectively, with 20 DEGs intersecting across all three DVTs. Enriched biological processes of these genes included type I/II interferon signaling, cytokine regulation, apoptosis, and T-cell differentiation. Principal component analysis based on 20 common DEGs (overlapping between DVTs and our NDI validation dataset) distinguished dengue patients by disease severity, particularly in the late acute phase. Machine learning analysis ranked the ten most critical predictors of disease severity in NDI, crucial for the anti-viral immune response. Conclusion: This work provides insights into the NDI and vaccine-induced overlapping immune response and suggests molecular markers (e.g., IFIT5, ISG15, and HERC5) for anti-dengue-specific therapies and effective vaccination development
Efficacy and safety of growth hormone treatment in children with short stature: the Italian cohort of the GeNeSIS clinical study
Purpose: We examined auxological changes in growth hormone (GH)-treated children in Italy using data from the Italian cohort of the multinational observational Genetics and Neuroendocrinology of Short Stature International Study (GeNeSIS) of pediatric patients requiring GH treatment. Methods: We studied 711 children (median baseline age 9.6 years). Diagnosis associated with short stature was as determined by the investigator. Height standard deviation score (SDS) was evaluated yearly until final or near-final height (n = 78). Adverse events were assessed in all GH-treated patients. Results: The diagnosis resulting in GH treatment was GH deficiency (GHD) in 85.5 % of patients, followed by Turner syndrome (TS 6.6 %). Median starting GH dose was higher in patients with TS (0.30 mg/kg/week) than patients with GHD (0.23 mg/kg/week). Median (interquartile range) GH treatment duration was 2.6 (0.6\u20133.7) years. Mean (95 % confidence interval) final height SDS gain was 2.00 (1.27\u20132.73) for patients with organic GHD (n = 18) and 1.19 (0.97\u20131.40) for patients with idiopathic GHD (n = 41), but lower for patients with TS, 0.37 ( 120.03 to 0.77, n = 13). Final height SDS was > 122 for 94 % of organic GHD, 88 % of idiopathic GHD and 62 % of TS patients. Mean age at GH start was lower for organic GHD patients, and treatment duration was longer than for other groups, resulting in greater mean final height gain. GH-related adverse events occurred mainly in patients diagnosed with idiopathic GHD. Conclusions: Data from the Italian cohort of GeNeSIS showed auxological changes and safety of GH therapy consistent with results from international surveillance databases
El ASIS en el contexto de la intervención comunitaria y su rol pedagógico: Redes intersectoriales en el Barrio Nuevo de Ringuelet (La Plata, 2018-2019)
El presente trabajo tiene como objetivo sistematizar y comunicar los resultados del desarrollo de un Análisis de Situación Integral de Salud (ASIS) en el Barrio Nuevo de Ringuelet (La Plata) -territorio surgido a partir de una relocalización de asentamientos populares- entre los meses de julio de 2018 y octubre de 2019. Como Residencia de Medicina General del Hospital Gutiérrez del Centro de Atención Primaria de la Salud (CAPS) n° 15, la pregunta de la que partimos es la siguiente: ¿Cuáles son las características sociodemográficas y las problemáticas de salud de la población que habita en Barrio Nuevo después de la relocalización?
El propósito de esta investigación es conocer a la población, sus problemáticas, necesidades y deseos para incidir en la calidad de vida de la misma desde una perspectiva sanitaria. Para el equipo de trabajo esto significa acompañar procesos participativos y colectivos en vistas de transformar la situación de salud del territorio partiendo de sus propias configuraciones culturales. A Deep Catalogue of Protein-Coding Variation in 983,578 Individuals
Rare coding variants that substantially affect function provide insights into the biology of a gene1-3. However, ascertaining the frequency of such variants requires large sample sizes4-8. Here we present a catalogue of human protein-coding variation, derived from exome sequencing of 983,578 individuals across diverse populations. In total, 23% of the Regeneron Genetics Center Million Exome (RGC-ME) data come from individuals of African, East Asian, Indigenous American, Middle Eastern and South Asian ancestry. The catalogue includes more than 10.4 million missense and 1.1 million predicted loss-of-function (pLOF) variants. We identify individuals with rare biallelic pLOF variants in 4,848 genes, 1,751 of which have not been previously reported. From precise quantitative estimates of selection against heterozygous loss of function (LOF), we identify 3,988 LOF-intolerant genes, including 86 that were previously assessed as tolerant and 1,153 that lack established disease annotation. We also define regions of missense depletion at high resolution. Notably, 1,482 genes have regions that are depleted of missense variants despite being tolerant of pLOF variants. Finally, we estimate that 3% of individuals have a clinically actionable genetic variant, and that 11,773 variants reported in ClinVar with unknown significance are likely to be deleterious cryptic splice sites.
