Pathogenesis of Huntington’s Disease: How to Fight Excitotoxicity and Transcriptional Dysregulation

Huntington’s disease (HD) is a neurodegenerative disorder caused by an expanded CAG repeat in the exon-1 of the huntingtin (htt) gene. The presence of mutant htt (mhtt) results in multiple physiopathological changes, including protein aggregation, transcriptional deregulation, decreased trophic support, alteration in signaling pathways and excitotoxicity. Indeed, the presence of mhtt induces changes in the activities/levels of different kinases, phosphatases and transcription factors that can impact on cell survival. Many studies have provided evidence that transcription may be a major target of mhtt, as gene dysregulation occurs before the onset of symptoms. The greatest number of downregulated genes in HD has led to test the ability of a large number of compounds to restore gene transcription in mouse models of HD. On the other hand, mhtt engenders multiple cellular dysfunctions including an increase of pathological glutamate-mediated excitotoxicity. For that reason, targeting the excess of glutamate has been the goal for many promising drugs leading to clinical trials. Although advances in developing effective therapies are evident, currently, there is no known cure for HD and existing symptomatic treatments are limited

Xenobiotics that affect oxidative phosphorylation alter differentiation of human adipose-derived stem cells at concentrations that are found in human blood

Adipogenesis is accompanied by differentiation of adipose tissuederived stem cells to adipocytes. As part of this differentiation, biogenesis of the oxidative phosphorylation system occurs. Many chemical compounds used in medicine, agriculture or other human activities affect oxidative phosphorylation function. Therefore, these xenobiotics could alter adipogenesis. We have analyzed the effects on adipocyte differentiation of some xenobiotics that act on the oxidative phosphorylation system. The tested concentrations have been previously reported in human blood. Our results show that pharmaceutical drugs that decrease mitochondrial DNA replication, such as nucleoside reverse transcriptase inhibitors, or inhibitors of mitochondrial protein synthesis, such as ribosomal antibiotics, diminish adipocyte differentiation and leptin secretion. By contrast, the environmental chemical pollutant tributyltin chloride, which inhibits the ATP synthase of the oxidative phosphorylation system, can promote adipocyte differentiation and leptin secretion, leading to obesity and metabolic syndrome as postulated by the obesogen hypothesis

Children’s agency located: An analysis from the experiences of girls and boys working in contexts of social inequality in Argentina

El trabajo realizado por niñas y niños es uno de los fenómenos que articulan preocupaciones sobre las infancias en América Latina y su análisis ha dado lugar a fructíferosdebates que exceden las perspectivas abolicionistas y regulacionistas. El objetivo deeste artículo es contribuir a este campo de estudios con nuevas preguntas y maticesa partir del análisis comparativo de experiencias de investigación con niñas y niñosque venden, piden y cuidan en contextos de desigualdad en Argentina. A conceptual approach from Latin America for the study of contemporary childhoods

Este artículo ofrece una aproximación conceptual para pensar las infancias contemporáneas tomando como base una serie de estudios desarrollados en contextos de desigualdad social en Argentina y la región, desde los interrogantes que coloca el escenario de la ¿pos?pandemia. Para ello, se analizan cuestiones claves en las experiencias de las infancias. Por un lado, el carácter central de niñas/os en la producción y reproducción de lazos sociales con pares, adultos e instituciones y por otro, el papel de las políticas públicas destinadas a las infancias que conllevan concepciones sobre modos adecuados de ser niño/a así como de crianza y cuidado. Se aboga por una perspectiva relacional para abordar la experiencia infantil que favorezca la comprensión de los/as niños/as como actores sociales inmersos en contextos moldeados por la interrelacionalidad y por procesos de producción de jerarquías y desigualdades. En particular, se focaliza en las prácticas relacionales de cuidado y las formas en que éstas son reguladas por los Estados. Éstos se presentan como nodos idóneos para reflexionar sobre, por un lado, las tensiones entre nociones universalizantes de niño/a y el reconocimiento de la diversidad de infancias y, por otro la problematización de las fronteras entre lo público y lo privado.This article offers a conceptual approach to think about contemporary childhoods based on a series of studies developed in contexts of social inequality in Argentina and the region, from the questions posed by the ¿pos?pandemic scenario. To this end, key issues in the experiences of children are analyzed. On the one hand, the centrality of children in the production and reproduction of social ties with peers, adults and institutions, and on the other hand, the role of public policies aimed at children that entail conceptions about appropriate ways of being a child as well as of upbringing and care. A relational perspective is advocated to address the childhood experience that favors the understanding of children as social actors immersed in contexts shaped by interrelationality and by processes of production of hierarchies and inequalities. In particular, it focuses on the relational practices of care and the ways in which these are regulated by the states, since they are presented as suitable nodes to reflect on, on the one hand, the tensions between universalizing notions of the child and the recognition of the diversity of childhoods and, on the other hand, the problematization of the boundaries between the public and the private spheres.Fil: Frasco Zuker, Laura.
