    Changes in DNA bending and flexing due to tethered cations detected by fluorescence resonance energy transfer

    Local DNA deformation arises from an interplay among sequence-related base stacking, intrastrand phosphate repulsion, and counterion and water distribution, which is further complicated by the approach and binding of a protein. The role of electrostatics in this complex chemistry was investigated using tethered cationic groups that mimic proximate side chains. A DNA duplex was modified with one or two centrally located deoxyuracils substituted at the 5-position with either a flexible 3-aminopropyl group or a rigid 3-aminopropyn-1-yl group. End-to-end helical distances and duplex flexibility were obtained from measurements of the time-resolved Förster resonance energy transfer between 5′- and 3′-linked dye pairs. A novel analysis utilized the first and second moments of the G(t) function, which encompasses only the energy transfer process. Duplex flexibility is altered by the presence of even a single positive charge. In contrast, the mean 5′–3′ distance is significantly altered by the introduction of two adjacently tethered cations into the double helix but not by a single cation: two adjacent aminopropyl groups decrease the 5′–3′ distance while neighboring aminopropynyl groups lengthen the helix

    llinois otter trapping was reinstated during the 2012-2013 Illinois Furbearer Trapping season. During 2014-2015, thethirdyear of reinstatement, 495 Illinois residents who trapped an otter were provided anotter permit; each otter trapper was mailed a 4-page questionnaire. We received 342 (74%) usable questionnaires. During the 2014-15 season, an average of 30 traps were set per trapper over the course of 43 days or nights. Respondents harvested an average of 2.2 otters (Lontra canadensis) for a total of 723 otters, which represented a 25% decrease from 2013-14. Participants also harvested 15,513 raccoons (Procyon lotor), 7,410 muskrat (Ondatra zibethicus), and 2,557 opossum (Didelphis virginiana) during the 2014-2015 Illinois trapping season.     Buried Black Hole Growth in IR-selected Mergers: New Results from Chandra

    Observations and theoretical simulations suggest that a significant fraction of merger-triggered accretion onto supermassive black holes is highly obscured, particularly in late-stage galaxy mergers, when the black hole is expected to grow most rapidly. Starting with the Wide-Field Infrared Survey Explorer all-sky survey, we identified a population of galaxies whose morphologies suggest ongoing interaction and which exhibit red mid-infrared colors often associated with powerful active galactic nuclei (AGNs). In a follow-up to our pilot study, we now present Chandra/ACIS and XMM-Newton X-ray observations for the full sample of the brightest 15 IR-preselected mergers. All mergers reveal at least one nuclear X-ray source, with 8 out of 15 systems exhibiting dual nuclear X-ray sources, highly suggestive of single and dual AGNs. Combining these X-ray results with optical line ratios and with near-IR coronal emission line diagnostics, obtained with the near-IR spectrographs on the Large Binocular Telescope, we confirm that 13 out of the 15 mergers host AGNs, two of which host dual AGNs. Several of these AGNs are not detected in the optical. All X-ray sources appear X-ray weak relative to their mid-infrared continuum, and of the nine X-ray sources with sufficient counts for spectral analysis, eight reveal strong evidence of high absorption with column densities of NH≳1023N_\mathrm{H} \gtrsim 10^{23}~cm−2^{-2}. These observations demonstrate that a significant population of single and dual AGNs are missed by optical studies, due to high absorption, adding to the growing body of evidence that the epoch of peak black hole growth in mergers occurs in a highly obscured phase.Comment: 29 pages, 22 figures; (Main text: 17 pages, 4 figures

    Subjective Well-Being and Cardiometabolic Health: An 8-11 Year Study of Midlife Adults

    Objective: Individuals who are satisfied and experience frequent positive emotions tend to have reduced risk for coronary heart disease (CHD). However, conflicting evidence exists and little research has investigated whether well-being is associated with early-warning indicators of biological risk that precede CHD. We investigated whether life satisfaction and positive emotions longitudinally predicted reduced risk of incident cardiometabolic conditions and healthier cardiometabolic risk scores, which may provide insight into underlying mechanisms and novel prevention targets. Methods: Initially healthy men and women (N=754–854) reported their baseline life satisfaction and positive emotions. During follow-up, presence of manifest cardiometabolic conditions was assessed and a separate cardiometabolic risk score was constructed from eight biomarkers. Poisson and linear regression analyses tested whether life satisfaction and positive emotionswere associated with reduced incident disease risk and lower cardiometabolic risk scores 8–11 years later. Results: Life satisfaction and positive emotions were each prospectively associated with reduced risk of manifest conditions, controlling for demographics and family history of CHD. Associations were attenuated for positive emotions after adjusting for depressive symptoms and for life satisfaction after adjusting for health behaviors. Life satisfactionwas associated with lower cardiometabolic risk scores until adding health behaviors, but positive emotions were not (regardless of the included covariates). Conclusion: Well-being, particularly life satisfaction, is associated with reduced risk for incident cardiometabolic conditions in minimally-adjusted models. However, accounting for underlying behavioral pathways attenuates the association. Low levels of life satisfaction (but not positive emotions) may also provide early warning of cardiometabolic risk prior to disease development

    Methods & proposal for metadata guiding principles for scholarly communications

    This article describes an international community-based effort to create metadata guiding principles for adopting and using richer metadata and advancing its application in scholarly communications. These principles can facilitate the dissemination, discoverability and use/reuse of many types of research and scholarly outputs. While much work remains to be done, these principles serve as a starting point for the evolution of processes that span communities including publishers, researchers, scholars, authors and other creators, librarians, curators, custodians, and consumers of scholarly works. These aspirational Metadata 2020 Principles are designed to encompass the needs of our entire community while ensuring thoughtful, purposeful, and reusable metadata resources. They provide a framework for all of us to be good metadata citizens. They also provide a foundation for considering related work from Metadata 2020 and must be interpreted within the legal and practical context in which we operate. They are intended to guide the broadest possible cross-section of our community in improving research communications, publishing, and discoverability

    The Association between Optimism and Serum Antioxidants in the Midlife in the United States Study

    Objective Psychological and physical health are often conceptualized as the absence of disease, but less research addresses positive psychological and physical functioning. For example, optimism has been linked with reduced disease risk and biological dysfunction, but very little research has examined associations with markers of healthy biological functioning. Thus, we investigated the association between two indicators of positive health: optimism and serum antioxidants. Methods The cross-sectional association between optimism and antioxidant concentrations was examined in 982 men and women from the Midlife in the United States study. Primary measures included self-reported optimism (assessed with the revised Life Orientation Test) and serum concentrations of nine different antioxidants (carotenoids and Vitamin E). Regression analyses examined the relationship between optimism and antioxidant concentrations in models adjusted for demographics, health status, and health behaviors. Results For every standard deviation increase in optimism, carotenoid concentrations increased by 3–13% in age-adjusted models. Controlling for demographic characteristics and health status attenuated this association. Fruit and vegetable consumption and smoking status were identified as potential pathways underlying the association between optimism and serum carotenoids. Optimism was not significantly associated with Vitamin E. Conclusions Optimism was associated with greater carotenoid concentrations and this association was partially explained by diet and smoking status. The direction of effects cannot be conclusively determined. Effects may be bidirectional given that optimists are likely to engage in health behaviors associated with more serum antioxidants, and more serum antioxidants are likely associated with better physical health that enhances optimism

    Prey range and genome evolution of Halobacteriovorax marinus predatory bacteria from an estuary

To understand how Halobacteriovorax strains impact ecosystems and develop them as biocontrol agents, it is important to characterize variation in predation phenotypes and investigate Halobacteriovorax genome evolution. We isolated Halobacteriovorax marinus BE01 from an estuary in Rhode Island using Vibrio from the same site as prey. Small, fast-moving, attack-phase BE01 cells attach to and invade prey cells, consistent with the intraperiplasmic predation strategy of the H. marinus type strain, SJ. BE01 is a prey generalist, forming plaques on Vibrio strains from the estuary, Pseudomonas from soil, and Escherichia coli. Genome analysis revealed extremely high conservation of gene order and amino acid sequences between BE01 and SJ, suggesting strong selective pressure to maintain the genome in this H. marinus lineage. Despite this, we identified two regions of gene content difference that likely resulted from horizontal gene transfer. Analysis of modal codon usage frequencies supports the hypothesis that these regions were acquired from bacteria with different codon usage biases than H. marinus. In one of these regions, BE01 and SJ carry different genes associated with mobile genetic elements. Acquired functions in BE01 include the dnd operon, which encodes a pathway for DNA modification, and a suite of genes involved in membrane synthesis and regulation of gene expression that was likely acquired from another Halobacteriovorax lineage.     Conservation and Use of the North American Plant Cornucopia: The Way Forward

    The pages of this extensive book document the potential of a great many North American plants to enhance the productivity, sustainability, and nutritional quality of crops or to be further developed into important cultivated species in their own right. But this potential can only be realized if the plants are adequately conserved to ensure their survival and availability for research, invested in to promote their development, and marketed so as to be attractive to producers and consumers. We outline some of the key steps needed to boost the conservation and use of our regional cornucopia. In situ and ex situ conservation of North America’s useful plants are being accomplished by a variety of institutions with different mandates, but habitat destruction and other threats to wild populations continue to negatively impact many species. Information sharing, coordinating efforts, filling research gaps for wild plants, and increasing support for conservation will be necessary to more comprehensively safeguard these plants and to make them available for use. Technologies enabling more efficient exploration of the diversity within these species are rapidly advancing and offer the potential to contribute to quick advances in improvement of cultivars, but considerable further research and partnerships are needed to generate and share the results widely. Marketing of new crops can take advantage of the increasing public interest in diverse and nutritious foods, learning from successful collaborations between producers, researchers, and consumers. As a whole, North America already possesses a strong foundation from which the conservation and use of its flora can be enhanced. This includes many protected areas, strong conservation institutions, innovative research, and the willingness to collaborate across fields, institutions, and borders. There are still many silos that need to be broken down and reorganized through innovative partnerships to better conserve and benefit from the North American cornucopia. But given the incredible diversity of interesting and useful plants in the region, the remarkable efforts for many decades by many organizations to care for these plants and share them with humanity, and the increasing public interest in more diverse, healthy, and resilient food and agricultural systems, there is reason for hope
