    Promoting Farmers\u27 Markets: Preferences of Farmers\u27 Market Leaders

    Extension professionals across disciplines are involved with farmers\u27 markets, and reports have indicated an increase in the number of farmers\u27 markets across the country. We explored perspectives of farmers\u27 market leaders regarding topics and data of interest and capacity and willingness to collect data related to market promotion. The purpose of our work was to provide Extension educators with information that may guide programming around farmers\u27 markets. We collected data through an online survey of Wisconsin farmers\u27 market leaders in spring 2017. Market leaders were most interested in learning how to encourage word-of-mouth communication between customers and engage in other low-cost strategies, such as having partners help promote a market

    The SbcCD complex and its homologues play important roles in DNA repair and in the maintenance of genome stability. In Escherichia coli, the in vitro functions of SbcCD have been well characterized, but its exact cellular role remains elusive. This work investigates the regulation of the sbcDC operon and the cellular localization of the SbcC and SbcD proteins. Transcription of the sbcDC operon is shown to be dependent on starvation and RpoS protein. Overexpressed SbcC protein forms foci that colocalize with the replication factory, while overexpressed SbcD protein is distributed through the cytoplasm

    Barriers to infection of human cells by feline leukemia virus: insights into resistance to zoonosis

    The human genome displays a rich fossil record of past gamma-retrovirus infections, yet no current epidemic is evident, despite environmental exposure to viruses that infect human cells in vitro. Feline leukemia viruses (FeLVs) rank high on this list, but domestic or workplace exposure has not been associated with detectable serological responses. Non-specific inactivation of gamma-retroviruses by serum factors appears insufficient to explain these observations. To investigate further we explored the susceptibility of primary and established human cell lines to FeLV-B, the most likely zoonotic variant. Fully permissive infection was common in cancer-derived cell lines, but was also a feature of non-transformed keratinocytes and lung fibroblasts. Cells of haematopoietic origin were less generally permissive and formed discrete groups on the basis of high or low intracellular protein expression and virion release. Potent repression was observed in primary human blood mononuclear cells and a subset of leukemia cell lines. However, the early steps of reverse transcription and integration appear to be unimpaired in non-permissive cells. FeLV-B was subject to G->A hypermutation with a predominant APOBEC3G signature in partially permissive cells but was not mutated in permissive cells or in non-permissive cells that block secondary viral spread. Distinct cellular barriers that protect primary human blood cells are likely to be important in protection against zoonotic infection with FeLV

    A microanalytical study of iron, aluminium and organic matter relationships in soils with contrasting hydrological regimes

    It is recognised that interactions between mineral oxides and soil organic matter (SOM) are an important factor in the stabilisation of soil organic carbon (SOC). The nature of these interactions is particularly complex in gleyed soils that experience periodic waterlogging and changeable redox conditions. This study explores the complex patterns of iron (Fe) (hydr)oxides and SOM in three soils with contrasting hydrological regimes (Gleysol, Stagnosol and Cambisol). Micromorphological examination of undisturbed soil thin sections was teamed with SEM-EDS analysis and sequential dissolution of Fe pedofeatures to gain a better understanding of the mechanisms involved in SOM stabilisation by mineral oxides. All soils contained a diverse range of particulate SOM forms and Fe pedofeatures; the degree of impregnation of the Fe pedofeatures was found to increase with depth and a strong correlation between the presence of SOM and Fe pedofeatures was found to exist through all soils. Weakly crystalline Fe (hydr)oxides were found in association with partially degraded tissue residues and amorphous fine organic matter (OM). Strongly crystalline Fe (hydr)oxides were found in all impregnative Fe pedofeatures and high Fe/C ratios suggested precipitative processes rather than sorption dominate SOC sequestration in these features. In addition, at the core of some strongly impregnated Fe nodules, occluded well preserved organic tissues were identified. The study highlights the range of processes and complexity involved in SOC sequestration over mm to cm scales and untangling this complexity is vital to understanding and modelling terrestrial C fluxes. Whilst the methods used here are not without their complications, the value of micro-scale studies of undisturbed soil thin sections is clearly demonstrated

    Effectiveness, inequality and ethos in three English schools

    Many school effectiveness studies have focussed on schools serving disadvantaged communities. ‘Schools matter most for underprivileged and/or initially low achieving students. Effective or ineffective schools are especially effective or ineffective for these students’ (Scheerens & Bosker 1997: 96). Analysing how schools can improve in socially disadvantaged communities is essential. The nature of a school cannot, of course, remove social inequality or social exclusion, but it can have a significant positive impact (Sammons 2007). In a study of improving schools in disadvantaged settings, Muijs et al. (2004) found they focus on a number of factors: teaching and learning; enhancing leadership capacity; creating an information rich environment; creating a positive school culture; building a learning community; promoting continuous professional development; involving parents; and engaging external support. This study of three English schools supports Muijs et al.’s findings, but points also to the importance of an ‘ethos of inclusion’. It also draws attention to role of schools serving students with Special Educational Needs

    Defining a roadmap for harmonizing quality indicators in Laboratory Medicine: A consensus statement on behalf of the IFCC Working Group "laboratory Error and Patient Safety" and EFLM Task and Finish Group "performance specifications for the extra-analytical phases"

    The improving quality of laboratory testing requires a deep understanding of the many vulnerable steps involved in the total examination process (TEP), along with the identification of a hierarchy of risks and challenges that need to be addressed. From this perspective, the Working Group \u201cLaboratory Errors and Patient Safety\u201d (WG-LEPS) of International Federation of Clinical Chemistry and Laboratory Medicine (IFCC) is focusing its activity on implementation of an efficient tool for obtaining meaningful information on the risk of errors developing throughout the TEP, and for establishing reliable information about error frequencies and their distribution. More recently, the European Federation of Clinical Chemistry and Laboratory Medicine (EFLM) has created the Task and Finish Group \u201cPerformance specifications for the extraanalytical phases\u201d (TFG-PSEP) for defining performance specifications for extra-analytical phases. Both the IFCC and EFLM groups are working to provide laboratories with a system to evaluate their performances and recognize the critical aspects where improvement actions are needed. A Consensus Conference was organized in Padova, Italy, in 2016 in order to bring together all the experts and interested parties to achieve a consensus for effective harmonization of quality indicators (QIs). A general agreement was achieved and the main outcomes have been the release of a new version of model of quality indicators (MQI), the approval of a criterion for establishing performance specifications and the definition of the type of information that should be provided within the report to the clinical laboratories participating to the QIs project

    Reflections and Experiences of a Co-Researcher involved in a Renal Research Study

    Background Patient and Public Involvement (PPI) is seen as a prerequisite for health research. However, current Patient and public involvement literature has noted a paucity of recording of patient and public involvement within research studies. There have been calls for more recordings and reflections, specifically on impact. Renal medicine has also had similar criticisms and any reflections on patient and public involvement has usually been from the viewpoint of the researcher. Roles of patient and public involvement can vary greatly from sitting on an Advisory Group to analysing data. Different PPI roles have been described within studies; one being a co-researcher. However, the role of the co-researcher is largely undefined and appears to vary from study to study. Methods The aims of this paper are to share one first time co-researcher's reflections on the impact of PPI within a mixed methods (non-clinical trial) renal research study. A retrospective, reflective approach was taken using data available to the co-researcher as part of the day-to-day research activity. Electronic correspondence and documents such as meeting notes, minutes, interview thematic analysis and comments on documents were re-examined. The co-researcher led on writing this paper. Results This paper offers a broad definition of the role of the co-researcher. The co-researcher reflects on undertaking and leading on the thematic analysis of interview transcripts, something she had not previously done before. The co-researcher identified a number of key themes; the differences in time and responsibility between being a coresearcher and an Advisory Group member; how the role evolved and involvement activities could match the co-researchers strengths (and the need for flexibility); the need for training and support and lastly, the time commitment. It was also noted that it is preferable that a co-researcher needs to be involved from the very beginning of the grant application. Conclusions The reflections, voices and views of those undertaking PPI has been largely underrepresented in the literature. The role of co-researcher was seen to be rewarding but demanding, requiring a large time commitment. It is hoped that the learning from sharing this experience will encourage others to undertake this role, and encourage researchers to reflect on the needs of those involved.Peer reviewedFinal Published versio
