    Neural Stem Cells Engineered to Express Three Therapeutic Factors Mediate Recovery from Chronic Stage CNS Autoimmunity

    © The American Society of Gene and Cell Therapy. Treatment of chronic neurodegenerative diseases such as multiple sclerosis (MS) remains a major challenge. Here we genetically engineer neural stem cells (NSCs) to produce a triply therapeutic cocktail comprising IL-10, NT-3, and LINGO-1-Fc, thus simultaneously targeting all mechanisms underlie chronicity of MS in the central nervous system (CNS): persistent inflammation, loss of trophic support for oligodendrocytes and neurons, and accumulation of neuroregeneration inhibitors. After transplantation, NSCs migrated into the CNS inflamed foci and delivered these therapeutic molecules in situ. NSCs transduced with one, two, or none of these molecules had no or limited effect when injected at the chronic stage of experimental autoimmune encephalomyelitis; cocktail-producing NSCs, in contrast, mediated the most effective recovery through inducing M2 macrophages/microglia, reducing astrogliosis, and promoting axonal integrity and endogenous oligodendrocyte/neuron differentiation. These engineered NSCs simultaneously target major mechanisms underlying chronicity of multiple sclerosis (MS) and encephalomyelitis (EAE), thus representing a novel and potentially effective therapy for the chronic stage of MS, for which there is currently no treatment available

    Although several studies have been published on the gut microbiota composition, they are mainly focused on bacteria. Therefore, the world of gut yeasts, the "gut mycome", is still unclear. Over the last years, brand new gut microbiota analysis techniques have been applied to the study of yeasts, with exciting results both in health and in disease. A therapeutic potential for many gastrointestinal and extra-intestinal diseases has been recognized for selected yeast strains, such as Saccharomyces boulardii.     Objetivo: Avaliar os efeitos do CLA associado à dieta hiperlipídica sobre o perfil lipídico e composição corporal de camundongos knockout para o gene da ApoE exercitados. Métodos: 32 camundongos foram divididos e submetidos à dieta normo e hiperlipídica suplementadas ou não com 1% de CLA. Todos realizaram exercício físico em esteira, durante 12 semanas. Utilizou-se o teste de Kruskal-Wallis ao nível de significância de 5%. Resultados: Os animais que ingeriram dieta hiperlipídica com CLA aumentaram o colesterol total e o LDL, comparado com os alimentados com dieta normolipídica associada ou não ao CLA. Observou-se aumento no ganho de peso dos camundongos que ingeriram dieta hiperlipídica com CLA, comparado com os normolipídicos suplementados com CLA. Conclusões: O CLA na concentração de 1% não alterou nenhum dos parâmetros analisados, independente da dieta, e não foi protetor contra os efeitos da dieta hiperlipídica

    Gut microbial flora, prebiotics and probiotics in IBD: their current usage and utility

    Inflammatory bowel diseases are chronic diseases affecting the gastrointestinal tract, whose major forms are represented by Crohn's disease (CD) and ulcerative colitis (UC). Their etiology is still unclear, although several factors have been identified as major determinants for induction or relapses. Among these, the role of the "forgotten organ", gut microbiota, has become more appreciated in recent years. The delicate symbiotic relationship between the gut microbiota and the host appears to be lost in IBD. In this perspective, several studies have been conducted to assess the role of prebiotics and probiotics in gut microbiota modulation. This is a minireview aimed to address in an easy format (simple questions-simple answers) some common issues about the theme. An update on the role of selected constituents of gut microbiota in the pathogenesis of IBD is presented together with the analysis of the efficacy of gut microbiota modulation by prebiotics and probiotics administration in the management of IBD
