    United States Air Force Academy get-away-special flexible beam experiment

    The Department of Astronautics at the United States Air Force Academy is currently planning to fly an experiment in a NASA Get-Away-Special (GAS) canister. The experiment was named the flex beam experiment. The primary technical objective of the flex beam experiment is to measure the damping of a thin beam in the vacuum and zero G environment of space. By measuring the damping in space, it is hoped to determine the amount of damping the beam normally experiences due to the gravitational forces present on Earth. This will allow validation of models which predict the dynamics of thin beams in the space environment. The experiment will also allow the Academy to develop and improve its ability to perform experiments within the confines of a NASA GAS canister. Several experiments, of limited technical difficulty, were flown by the Academy. More complex experiments are currently planned and it is hoped to learn techniques with each space shuttle flight

    In good company: risk, security and choice in young people's drug decisions

    This article draws on original empirical research with young people to question the degree to which 'individualisation of risk', as developed in the work of Beck and Giddens, adequately explains the risks young people bear and take. It draws on alternative understandings and critiques of 'risk' not to refute the notion of the reflexive individual upon which 'individualisation of risk' is based but to re-read that reflexivity in a more hermeneutic way. It explores specific risk-laden moments – young people's drug use decisions – in their natural social and cultural context of the friendship group. Studying these decisions in context, it suggests, reveals the meaning of 'risk' to be not given, but constructed through group discussion, disagreement and consensus and decisions taken to be rooted in emotional relations of trust, mutual accountability and common security. The article concludes that 'the individualisation of risk' fails to take adequate account of the significance of intersubjectivity in risk-decisions. It argues also that addressing the theoretical overemphasis on the individual bearer of risk requires not only further empirical testing of the theory but appropriate methodological reflection

    Nondestructive Evaluation of the J-2X Direct Metal Laser Sintered Gas Generator Discharge Duct

    The J-2X program at NASA's Marshall Space Flight Center (MSFC) procured a direct metal laser sintered (DMLS) gas generator discharge duct from Pratt & Whitney Rocketdyne and Morris Technologies for a test program that would evaluate the material properties and durability of the duct in an engine-like environment. DMLS technology was pursued as a manufacturing alternative to traditional techniques, which used off nominal practices to manufacture the gas generator duct's 180 degree turn geometry. MSFC's Nondestructive Evaluation (NDE) Team performed radiographic, ultrasonic, computed tomographic, and fluorescent penetrant examinations of the duct. Results from the NDE examinations reveal some shallow porosity but no major defects in the as-manufactured material. NDE examinations were also performed after hot-fire testing the gas generator duct and yielded similar results pre and post-test and showed no flaw growth or development

    Kinetic evaluation of human cloned coproporphyrinogen oxidase using a ring isomer of the natural substrate

    Background: The enzyme coproporphyrinogen oxidase (copro\u27gen oxidase) converts coproporphyrinogen-Ill (GIII) to protoporphyrinogen-IX via an intermediary monovinyl porphyrinogen. The A ring isomer coproporphyrinogen-IV (C-IV) has previously been shown to be a substrate for copro\u27gen oxidase derived from avian erythrocytes. In contrast to the authentic substrate (GIII) where only a small amount of the monovinyl intermediate is detected, C-IV gives rise to a monovinyl intermediate that accumulates before being converted to an isomer of protoporphyrinogen-IX. No kinetic studies have been carried out using the purified human copro\u27gen oxidase to evaluate its ability to process both the authentic substrate as well as analogs. Material/Methods: Therefore, purified, cloned human copro\u27gen oxidase was incubated with GIII or C-IV at 37 degrees C with various substrate concentrations (from 0.005 mu M to 3.5 mu M). The Km (an indication of molecular recognition) and Kcat (turnover number) values were determined. Results: The Km value for total product formation was about the same with either C-III or C-IV indicating the same molecular recognition. However, the catalytic efficiency (Kcat/Km) of the enzyme for total product formation was not more than two fold higher using GIII relative to C-IV. Conclusions: Since the Km values are about the same for either substrate and the total Kcat/Km values are within two fold of each other, this could correlate with the increase of severity of porphyrias with monovinyl accumulation. The ability of the increased levels of C-IV to compete with the authentic substrate has important implications for clinical porphyrias

    This is a detailed case history of one of London’s iconic new media companies, AMX Studios. Some of the changes in this firm, we assume, are not untypical for other firms in this sector. Particularly we want to draw attention to two transformations. The first change in AMX and in London’s new media industry more generally refers to the field of industrial relations. What can be observed is a shift from a rather heterarchical towards a more hierarchical organized new media industry, a shift from short-term project networks to long-term client dependency. The second change refers to new media products and services. We want to argue for a shift from cool content production towards consultancy and interactive communications solutions

    Hot dense capsule implosion cores produced by z-pinch dynamic hohlraum radiation

    Hot dense capsule implosions driven by z-pinch x-rays have been measured for the first time. A ~220 eV dynamic hohlraum imploded 1.7-2.1 mm diameter gas-filled CH capsules which absorbed up to ~20 kJ of x-rays. Argon tracer atom spectra were used to measure the Te~ 1keV electron temperature and the ne ~ 1-4 x10^23 cm-3 electron density. Spectra from multiple directions provide core symmetry estimates. Computer simulations agree well with the peak compression values of Te, ne, and symmetry, indicating reasonable understanding of the hohlraum and implosion physics.Comment: submitted to Phys. Rev.     Investigation of the catalytic and structural roles of conserved histidines of human coproporphyrinogen oxidase using site-directed mutagenesis

    Background: The catalytic contribution of four conserved histidines of human coproporphyrinogen oxidase (CPO) has been investigated using site-directed mutagenesis to change histidine (H) into alanine (A). Material/Methods: The wild-type and mutant enzyme forms were analyzed for their ability to utilize coproporphyrinogen-III, mesoporphvrinogen-VI, and harderoporphyrinogen as substrates. Results: Wild-type CPO had specific activities of 4.9 +/- 0.9 nmole product/min/mg for coproporphyrinogen-III, 1.7 +/- 0.7 nmole ptoduct/min/mg for mesoporphyrinogen-VI, and 5.1 +/- 1.8 nmole product/min/mg for harderoporphyrinogen. The four mutant enzymes were catalytically competent With all three substrates, but to varying degrees. The most affected Mutant was the H158A enzyme which exhibited approximately 50-fold lower activity than wild-type recombinant CPO. Conclusions: Thus, His 158 of human CPO may have a role ill the active site, but none of the conserved histidine residues of human coproporphyrinogen oxidase is essential for catalytic activity although changes in histidines have been implicated in the disease state hereditary coproporphyria

    Exploring the role of professional associations in collective learning in London and New York's advertising and law professional service firm clusters.

    The value of regional economies for collective learning has been reported by numerous scholars. However often work has been criticised for lacking analytical clarity and failing to explore the architectures of collective learning and the role of the knowledge produced in making firms in a cluster economy successful. This paper engages with these problematics and investigates how collective learning is facilitated in the advertising and law professional service firm clusters in London and New York. It explores the role of professional associations and investigates how they mediate a collective learning process in each city. It argues that professional associations seed urban communities of practice that emerge outside of the formal activities of professional associations. In these communities individual with shared interests in advertising and law learn from one-another and are therefore able to adapt and evolve one-another approaches to common industry challenges. The paper suggests this is another form of the variation Marshall highlighted in relation to cluster-based collective learning. The paper also shows how the collective learning process is affected by the presence, absence and strength of an institutional thickness. It is therefore argued that a richer understanding of institutional affects is needed in relation to CL

    Histological insights into the pathogenesis of post‐Roux‐en‐Y hyperinsulinaemic hypoglycaemia

    Background β‐cell hyperplasia has been implicated in the aetiology of post Roux‐en‐Y gastric bypass hyperinsulinaemic hypoglycaemia, but the pathogenesis of this condition is still unclear. Case report We report a case of a 52‐year‐old man with post‐Roux‐en‐Y gastric bypass hyperinsulinaemic hypoglycaemia who underwent distal pancreatectomy to alleviate his symptoms. Pancreatic histopathology showed chronic pancreatitis with a corresponding loss of exocrine tissue and islet retention. Amyloid deposition was found in pancreatic islets. These features are more typically associated with Type 2 diabetes. Discussion This case highlights the potential multifactorial pathogenesis of symptomatic hypoglycaemia after Roux‐en‐Y gastric bypass. What's new? We present a case of a patient with post Roux‐en‐Y hypoglycaemia and pancreatic histology typically associated with diabetes.
