    Outdoor micro-climate: Air temperature measurements around an office building in Denmark during summer

    The outdoor micro-climate caused by the presence of buildings can significantly differ from that around weather stations located outside of urban areas. However, the latter is often used to design buildings and size building systems. This could lead to significant mistakes and performance gaps. To date, there is a certain lack of experimental studies assessing the microclimate around buildings, especially in Scandinavian countries. The current paper presents the preliminary results and analysis of a measurement campaign of the temperature gradient in the two-meter air layer around the envelope of a multi-storey office building in Denmark in the summertime. Depending on the orientation of the external building surface (South/North façade or rooftop), the distance from the latter and the weather conditions, the temperature in this two-meter air layer can vary significantly and differ from the air temperature measured at nearby open fields or recorded by the reference weather station. During sunny days, a temperature gradient of up to 3.4 °C and 13.6 °C was measured in the air layer around the South façade and the rooftop, respectively. These results could help to validate urban climate models and bridge the gap between building design and real-condition performance.     Bioorthogonal Non-canonical Amino Acid Tagging Combined With Flow Cytometry for Determination of Activity in Aquatic Microorganisms

    In this study, we have combined bioorthogonal non-canonical amino acid tagging (BONCAT) and flow cytometry (FCM) analysis, and we demonstrate the applicability of the method for marine prokaryotes. Enumeration of active marine bacteria was performed by combining the DNA stain SYBR Green with detection of protein production with BONCAT. After optimization of incubation condition and substrate concentration on monoculture of Escherichia coli, we applied and modified the method to natural marine samples. We found that between 10 and 30% of prokaryotes in natural communities were active. The method is replicable, fast, and allow high sample throughput when using FCM. We conclude that the combination of BONCAT and FCM is an alternative to current methods for quantifying active populations in aquatic environments.publishedVersio

    Flow Cytometric Analysis of Bacterial Protein Synthesis: Monitoring Vitality After Water Treatment

    Bacterial vitality after water disinfection treatment was investigated using bio-orthogonal non-canonical amino acid tagging (BONCAT) and flow cytometry (FCM). Protein synthesis activity and DNA integrity (BONCAT–SYBR Green) was monitored in Escherichia coli monocultures and in natural marine samples after UV irradiation (from 25 to 200 mJ/cm2) and heat treatment (from 15 to 45 min at 55°C). UV irradiation of E. coli caused DNA degradation followed by the decrease in protein synthesis within a period of 24 h. Heat treatment affected both DNA integrity and protein synthesis immediately, with an increased effect over time. Results from the BONCAT method were compared with results from well-known methods such as plate counts (focusing on growth) and LIVE/DEAD™ BacLight™ (focusing on membrane permeability). The methods differed somewhat with respect to vitality levels detected in bacteria after the treatments, but the results were complementary and revealed that cells maintained metabolic activity and membrane integrity despite loss of cell division. Similarly, analysis of protein synthesis in marine bacteria with BONCAT displayed residual activity despite inability to grow or reproduce. Background controls (time zero blanks) prepared using different fixatives (formaldehyde, isopropanol, and acetic acid) and several different bacterial strains revealed that the BONCAT protocol still resulted in labeled, i.e., apparently active, cells. The reason for this is unclear and needs further investigation to be understood. Our results show that BONCAT and FCM can detect, enumerate, and differentiate bacterial cells after physical water treatments such as UV irradiation and heating. The method is reliable to enumerate and explore vitality of single cells, and a great advantage with BONCAT is that all proteins synthesized within cells are analyzed, compared to assays targeting specific elements such as enzyme activity.publishedVersio

    On-site and laboratory evaluations of soundscape quality in recreational urban spaces

    Context: Regulations for quiet urban areas are typically based on sound level limits alone. However, the nonacoustic context may be crucial for subjective soundscape quality. Aims: This study aimed at comparing the role of sound level and nonacoustic context for subjective urban soundscape assessment in the presence of the full on-site context, the visual context only, and without context. Materials and Methods: Soundscape quality was evaluated for three recreational urban spaces by using four subjective attributes: loudness, acceptance, stressfulness, and comfort. The sound level was measured at each site and simultaneous sound recordings were obtained. Participants answered questionnaires either on site or during laboratory listening tests, in which the sound recordings were presented with or without each site’s visual context consisting of two pictures. They rated the four subjective attributes along with their preference toward eight sound sources. Results: The sound level was found to be a good predictor of all subjective parameters in the laboratory, but not on site. Although all attributes were significantly correlated in the laboratory setting, they did not necessarily covary on site. Moreover, the availability of the visual context in the listening experiment had no significant effect on the ratings. The participants were overall more positive toward natural sound sources on site. Conclusion: The full immersion in the on-site nonacoustic context may be important when evaluating overall soundscape quality in urban recreational areas.
