    Premise of the studyIdentifying clonal lineages in asexually reproducing plants using microsatellite markers is complicated by the possibility of nonidentical genotypes from the same clonal lineage due to somatic mutations, null alleles, and scoring errors. We developed and tested a clonal identification protocol that is robust to these issues for the asexually reproducing hexaploid tree species coast redwood (Sequoia sempervirens).MethodsMicrosatellite data from four previously published and two newly developed primers were scored using a modified protocol, and clones were identified using Bruvo genetic distances. The effectiveness of this clonal identification protocol was assessed using simulations and by genotyping a test set of paired samples of different tissue types from the same trees.ResultsData from simulations showed that our protocol allowed us to accurately identify clonal lineages. Multiple test samples from the same trees were identified correctly, although certain tissue type pairs had larger genetic distances on average.DiscussionThe methods described in this paper will allow for the accurate identification of coast redwood clones, facilitating future studies of the reproductive ecology of this species. The techniques used in this paper can be applied to studies of other clonal organisms as well

    Penaeus vannamei Boone, 1931 commonly known as the white legged shrimp was observed along with Penaeus indicus in the landings at Kalamukku fish landing centre Ernakulam,on 15.11.20 18. The catch was from a single day trawler whose depth of operation was 15 m. Distributed along the Pacific coast of Mexico, central and south America up to Peru, the shrimp was introduced in India on a pilot scale for culture in 2003 and later in 2009 large scale production began. However, it was not recorded in the wild capture fisheries for shrimps. In the landings at Kalamukku, the females measured 109 to 146 mm in total length and weighed 8.9 to 26.6 g. The males measured 100 to 145 mm in total length and 7.5 to 24.4 g in weight. Among the sample analysed 51% were P. vannamei and 49% were P. indicus based on numbers. Its appearance in the landings is most likely due to the recent floods in Kerala during August, 2018

    Objective: Aim of the present research work was to develop a sensitive, rapid and accurate, stability-indicating RP-UPLC method for the simultaneous estimation of tezacaftor and ivacaftor in formulations. Methods: The chromatographic separation of the mixture of tezacaftor and ivacaftor was attained in isocratic method utilizing a mobile phase of 0.1 % orthophosphoric acid and acetonitrile in the proportion of 50:50%v/v utilizing a HSS C18 column which has dimensions of 100×2.1 mm, 1.7 m particle size and the flow rate of 0.3 ml/min. The detection system was monitored at 292 nm wavelength maximum with 1.5 ml injection volume. The present method was validated as per the guidelines given by the ICH for specificity, accuracy, sensitivity, linearity and precision. Results: The retaining time for tezacaftor and ivacaftor were achieved at 1.071 min and 0.530 min, respectively. Tezacaftor, ivacaftor and their combined drug formulation were exposed to thermal, acidic, oxidative, photolytic, and alkaline conditions. The developed method was highly sensitive, rapid, precise and accurate than the earlier reported methods. The total run time was decreased to 2.0 min; hence, the technique was more precise and economical. Stability studies directed for the suitability of the technique for degradation studies of tezacaftor and ivacaftor. Conclusion: The projected method can be utilized for routine analysis in the quality control department in pharmaceutical trades
