    Learning from support workers: can a dramatherapy group offer a community provision to support changes in care for people with learning disabilities and mental health difficulties?

    1.1 Background The UK Government's Transforming Care Agenda for people with learning disabilities has struggled to meet its goals of reducing inpatient beds and building communityā€based support. This article reports on the experiences of support staff who attended dramatherapy groups developed to assist transitions from an inpatient hospital and to prevent reā€admissions through postā€discharge support. The groups provide ongoing support and a place where relationships can be developed between supporter and those supported. 1.2 Materials and Methods A focus group with a purposive sample of paid support staff. The data was synthesised using a thematic framework approach. 1.3 Results Themes include: (a) new way of supporting and (b) hospital connection. The groups helped improve social interaction, friendship building, communication and selfā€confidence. Additional benefits include the pooling of support and a connection with professionals that enables difficulties to be caught early. 1.4 Conclusions Support workers valued these dramatherapy groups, recognising how the intervention enabled people with learning disabilities to develop relationships and provide easy access to mental health professionals. Support staff also found benefits for themselves which included shared support and an increased understanding and insight into the people they support

    Just One? Solo Dining, Gender and Temporal Belonging in Public Spaces

    In recent years, various lifestyle websites have offered tips on eating out alone as well as lists of the best restaurants for solo dining in major cities of the world. Utilising the theoretical concepts of participation units, territories of the self (Goffman 1972[1971]) and belonging (Author B2011, 2013), this paper explores the challenges that spatio-temporal conventions pose for women solo diners in particular. Through the lens of solo dining, we explore being alone and belonging in shared public spaces, and the gendered nature of aloneness and respectability. The papercontributes to existing theory by examining the influence that time has on a woman solo dinerā€™s ā€˜singleā€™ participation unit, her ability to lay claim to public space and her relationship with the surrounding social environment. The paper concludes by exploring what the new trend of solo dining can offer and the consequences this has for how sociologists conceptualise sociality inpublic spaces

    Lysophosphatidic Acid-Induced Transcriptional Profile Represents Serous Epithelial Ovarian Carcinoma and Worsened Prognosis

    BACKGROUND:Lysophosphatidic acid (LPA) governs a number of physiologic and pathophysiological processes. Malignant ascites fluid is rich in LPA, and LPA receptors are aberrantly expressed by ovarian cancer cells, implicating LPA in the initiation and progression of ovarian cancer. However, there is an absence of systematic data critically analyzing the transcriptional changes induced by LPA in ovarian cancer. METHODOLOGY AND PRINCIPAL FINDINGS:In this study, gene expression profiling was used to examine LPA-mediated transcription by exogenously adding LPA to human epithelial ovarian cancer cells for 24 h to mimic long-term stimulation in the tumor microenvironment. The resultant transcriptional profile comprised a 39-gene signature that closely correlated to serous epithelial ovarian carcinoma. Hierarchical clustering of ovarian cancer patient specimens demonstrated that the signature is associated with worsened prognosis. Patients with LPA-signature-positive ovarian tumors have reduced disease-specific and progression-free survival times. They have a higher frequency of stage IIIc serous carcinoma and a greater proportion is deceased. Among the 39-gene signature, a group of seven genes associated with cell adhesion recapitulated the results. Out of those seven, claudin-1, an adhesion molecule and phenotypic epithelial marker, is the only independent biomarker of serous epithelial ovarian carcinoma. Knockdown of claudin-1 expression in ovarian cancer cells reduces LPA-mediated cellular adhesion, enhances suspended cells and reduces LPA-mediated migration. CONCLUSIONS:The data suggest that transcriptional events mediated by LPA in the tumor microenvironment influence tumor progression through modulation of cell adhesion molecules like claudin-1 and, for the first time, report an LPA-mediated expression signature in ovarian cancer that predicts a worse prognosis

    Rattling Europeā€™s ordoliberal ā€˜iron cageā€™ : the contestation of austerity in Southern Europe

    This article explains the popular revolt against austerity in Southern Europe as the outcome of profound politico-economic changes that are shaped by the transformation of the European Unionā€™s (EUā€™s) macro-economic governance. It comprises three parts. The first part demonstrates how ordoliberalism ā€“ the Germanic variant of (neo)liberal economic thinking ā€“ was embedded in the EUā€™s new macro-economic governance, in processes that constitutionalise austerity and remove democratic controls over the economy. The second part examines the impact of austerity-driven reforms on welfare and employment in the aftermath of the sovereign debt crisis. These reforms undermined the social reproduction of Southern Europeā€™s familistic welfare model by destabilising three key pillars of social protection: employment security for householdsā€™ primary earners; small property ownership; and pension adequacy. The third part analyses the emergence of anti-austerity social politics in Southern Europe, both parliamentary and grassroots, and assesses their effectiveness in light of the collapse of public trust in both EU and domestic political institutions. The article concludes with our reflections on the fragility of EUā€™s integration process under the hegemony of ordoliberalism

    p53-dependent control of transactivation of the Pen2 promoter by presenilins

    The senile plaques found in the brains of patients with Alzheimer's disease are mainly due to the accumulation of amyloid Ī²-peptides (AĪ²) that are liberated by Ī³-secretase, a high molecular weight complex including presenilins, PEN-2, APH-1 and nicastrin. The depletion of each of these proteins disrupts the complex assembly into a functional protease. Here, we describe another level of regulation of this multimeric protease. The depletion of both presenilins drastically reduces Pen2 mRNA levels and its promoter transactivation. Furthermore, overexpression of presenilin-1 lowers Pen2 promoter transactivation, a phenotype abolished by a double mutation known to prevent presenilin-dependent Ī³-secretase activity. PEN-2 expression is decreased by depletion of Ī²-amyloid precursor protein (APP) and increased by the APP intracellular domain (AICD). We show that AICD and APP complement for Pen2 mRNA levels in APP/APLP1-2 knockout fibroblasts. Interestingly, overexpression of presenilin-2 greatly increases Pen2 promoter transactivation. The opposite effect triggered by both presenilins was reminiscent of our previous study, which showed that these two proteins elicit antagonistic effects on p53. Therefore, we examined the contribution of p53 on Pen2 transcription. Pen2 promoter transactivation, and Pen2 mRNA and protein levels were drastically reduced in p53ā€“/ā€“ fibroblasts. Furthermore, PEN-2 expression could be rescued by p53 complementation in p53- and APP-deficient cells. Interestingly, PEN-2 expression was also reduced in p53-deficient mouse brain. Overall, our study describes a p53-dependent regulation of PEN-2 expression by other members of the Ī³-secretase complex, namely presenilins

    Atypical PKCiota Contributes to Poor Prognosis Through Loss of Apical-basal Polarity and Cyclin E Overexpression in Ovarian Cancer

    We show that atypical PKCĪ¹, which plays a critical role in the establishment and maintenance of epithelial cell polarity, is genomically amplified and overexpressed in serous epithelial ovarian cancers. Furthermore, PKCĪ¹ protein is markedly increased or mislocalized in all serous ovarian cancers. An increased PKCĪ¹ DNA copy number is associated with decreased progression-free survival in serous epithelial ovarian cancers. In a Drosophila in vivo epithelial tissue model, overexpression of persistently active atypical PKC results in defects in apical-basal polarity, increased Cyclin E protein expression, and increased proliferation. Similar to the Drosophila model, increased PKCĪ¹ proteins levels are associated with increased Cyclin E protein expression and proliferation in ovarian cancers. In nonserous ovarian cancers, increased PKCĪ¹ protein levels, particularly in the presence of Cyclin E, are associated with markedly decreased overall survival. These results implicate PKCĪ¹ as a potential oncogene in ovarian cancer regulating epithelial cell polarity and proliferation and suggest that PKCĪ¹ is a novel target for therapy

    Development of SNP markers present in expressed genes of the plant-pathogen interaction: Theobroma cacao - Moniliophtora perniciosa

    We report the detection, validation and analysis of SNPs in the plant-pathogen interaction between cacao and Moniliophthora perniciosa ESTs using resequencing. This analysis in 73 EST sequences allowed the identification of 185 SNPs, 57% of them corresponding to transversion, 29% to transition and 14% to indels. The ESTs containing SNPs were classified into 14 main functional categories. After validation, 91 SNPs were confirmed, categorized and the parameters of nucleotide diversity and haplotype were calculated. Haplotype-based gene diversity and polymorphic information content (PIC) ranged from 0.559 to 0.56 and 0.115 to 0.12; respectively. Also, it was the advantage when considering haplotypes structure for each locus in place of single SNPs. Most of the gene fragments had a major haplotype combined to a series of low frequency haplotypes. Thus, the re-sequencing approach proved to be a valuable resource to identify useful SNPs for wide genetic applications. Furthermore, the cacao genome sequence availability allow a positional selection of DNA fragments to be re-sequenced enhancing the usefulness of the discovered SNPs. These results indicate the potential use of SNPs markers to identify allelic status of cacao resistance genes through marker-assisted selection to support the development of promising genotypes with high resistance to witch's broom disease.
