New Insights into the Lactate Shuttle: Role of MCT4 in the Modulation of the Exercise Capacity.

Lactate produced by muscle during high-intensity activity is an important end product of glycolysis that supports whole body metabolism. The lactate shuttle model suggested that lactate produced by glycolytic muscle fibers is utilized by oxidative fibers. MCT4 is a proton coupled monocarboxylate transporter preferentially expressed in glycolytic muscle fibers and facilitates the lactate efflux. Here we investigated the exercise capacity of mice with disrupted lactate shuttle due to global deletion of MCT4 (MCT4−/−) or muscle-specific deletion of the accessory protein Basigin (iMSBsg−/−). Although MCT4−/− and iMSBsg−/− mice have normal muscle morphology and contractility, only MCT4−/− mice exhibit an exercise intolerant phenotype. In vivo measurements of compound muscle action potentials showed a decrement in the evoked response in the MCT4−/− mice. This was accompanied by a significant structural degeneration of the neuromuscular junctions (NMJs). El Servicio de Urgencias del C.H.G.U.V. cuenta con una plantilla de 84 Enfermeros/as. Una parte considerable de ellos es personal de nueva incorporación, de ahí que sea prioritaria la elaboración de protocolos de actuación. La administración de medicación intravenosa es sin duda una de las técnicas de mayor aplicación en los pacientes que acuden a Urgencias, que en nuestro Hospital oscila entre 550-600 atenciones diarias. Por ello nos planteamos, mediante la realización de reuniones de grupo, la elaboración del presente trabajo con el objetivo de elaborar un manual informativo sobre la medicación más habitual utilizada en nuestro Servicio de Urgencias; indicaciones, contraindicaciones, diluciones, cuidados y recomendaciones sobre la administración que nos ayude a visionar la practica en nuestro trabajo y la calidad del Servicio que prestamos a nuestros pacientes, y sirva de consulta sobre todo al personal de nueva incorporación.The First aid service of the C.H.G.U.V. Biochemical parameters of silver catfish (Rhamdia quelen) after transport with eugenol or essential oil of Lippia alba added to the water

The transport of live fish is a routine practice in aquaculture and constitutes a considerable source of stress to the animals. The addition of anesthetic to the water used for fish transport can prevent or mitigate the deleterious effects of transport stress. This study investigated the effects of the addition of eugenol (EUG) (1.5 or 3.0 mu L L-1) and essential oil of Lippia alba (EOL) (10 or 20 mu L L-1) on metabolic parameters (glycogen, lactate and total protein levels) in liver and muscle, acetylcholinesterase activity (AChE) in muscle and brain, and the levels of protein carbonyl (PC), thiobarbituric acid reactive substances (TBARS) and nonprotein thiol groups (NPSH) and activity of glutathione-S-transferase in the liver of silver catfish (Rhamdia quelen; Quoy and Gaimard, 1824) transported for four hours in plastic bags (loading density of 169.2 g L-1). The addition of various concentrations of EUG (1.5 or 3.0 mu L L-1) and EOL (10 or 20 mu L L-1) to the transport water is advisable for the transportation of silver catfish, since both concentrations of these substances increased the levels of NPSH antioxidant and decreased the TBARS levels in the liver. Spinal cord from body donors is suitable for multicolor immunofluorescence

Immunohistochemistry is a powerful tool for studying neuronal tissue from humans at the molecular level. Obtaining fresh neuronal tissue from human organ donors is difficult and sometimes impossible. In anatomical body donations, neuronal tissue is dedicated to research purposes and because of its easier availability, it may be an alternative source for research. In this study, we harvested spinal cord from a single organ donor 2 h (h) postmortem and spinal cord from body donors 24, 48, and 72 h postmortem and tested how long after death, valid multi-color immunofluorescence or horseradish peroxidase (HRP) immunohistochemistry is possible. We used general and specific neuronal markers and glial markers for immunolabeling experiments. Here we showed that it is possible to visualize molecularly different neuronal elements with high precision in the body donor spinal cord 24 h postmortem and the quality of the image data was comparable to those from the fresh organ donor spinal cord. High-contrast multicolor images of the 24-h spinal cords allowed accurate automated quantification of different neuronal elements in the same sample. Although there was antibody-specific signal reduction over postmortem intervals, the signal quality for most antibodies was acceptable at 48 h but no longer at 72 h postmortem. In conclusion, our study has defined a postmortem time window of more than 24 h during which valid immunohistochemical information can be obtained from the body donor spinal cord. Cultivo do girassol no Rio Grande do Sul: plantio janeiro a fevereiro.

A demanda por esta oleaginosa está aumentando no RS, de forma que os produtores estão expandindo a área e a época de plantio, com isso surge um interesse especial no período de plantio janeiro/fevereiro (safrinha). Visando atender esta demanda, foram realizados ensaios com quatro datas de semeadura e em seis locais, a fim de determinar a viabilidade técnica deste cultivo no período da safrinha 2007 no RS. Cartoons kill: casualties in animated recreational theater in an objective observational new study of kids' introduction to loss of life.

Objectives To assess the risk of on-screen death of important characters in children’s animated films versus dramatic films for adults. Design Kaplan-Meier survival analysis with Cox regression comparing time to first on-screen death. Setting Authors’ television screens, with and without popcorn. Participants Important characters in 45 top grossing children’s animated films and a comparison group of 90 top grossing dramatic films for adults. Main outcome measures Time to first on-screen death. Results Important characters in children’s animated films were at an increased risk of death compared with characters in dramatic films for adults (hazard ratio 2.52, 95% confidence interval 1.30 to 4.90). Risk of on-screen murder of important characters was higher in children’s animated films than in comparison films (2.78, 1.02 to 7.58). Conclusions Rather than being the innocuous form of entertainment they are assumed to be, children’s animated films are rife with on-screen death and murder
