    Evaluation of Ellagic acid on the activities of oral bacteria with the use of adenosine triphosphate (ATP) bioluminescence assay

    Ellagic acid, a natural herb extract from Galla Chinensis in traditional Chinese medicine, shows antimicrobial activity to certain bacteria. The present study evaluated the effect of Ellagic acid on the growth of oral bacteria as well as their generation of water-insoluble glucan and adhesion to salivacoated hydroxyapatite (S-HA) beads. Streptococcus mutans ATCC 25175, Streptococcus sanguis ATCC 10556, Streptococcus salivarius ATCC 25975, Actinomyces naeslundii ATCC 12104, Actinomyces viscosus ATCC 15987, Lactobacillus rhamnosus ATCC 53103, Porphyromonas gingivalis ATCC 33277 and Bacteroides forsythus ATCC 43037 were the bacterial cell lines used in this study. Antibacterial activity of Ellagic acid was determined by using adenosine triphosphate (ATP) bioluminescence assayat various concentrations from 0.125 to 8 mg/ml. Anthrone method was used to evaluate the level of water-insoluble glucan generated by oral bacteria. The numbers of 3H-thymidine labeled bacteria attached to S-HA was counted by scintillation counting method. Sprague Dawley rats were orally fed with 0.5mg/mL ellagic acid for 28 days and their behaviours and excretions were monitored. Ellagic acid reduced bacterial metabolic rates and inhibited the growth of the tested bacterial strains. The waterinsolubleglucan generated by S. mutans and its adhesion to S-HA were reduced. Ellagic acid demonstrated no toxicity in animals fed for 28 days. Ellagic acid might be a promising compound for the development of antimicrobial agents against oral pathogens in human, thereby reducing theincidence of dental caries

    Application of a Nano-antimicrobial film to prevent ventilator-associated pneumonia: A pilot study

    Ventilator-associated pneumonia (VAP) is one of the most common hospital-associated infections and has accounted for approximately 15% of all hospital-associated infections. In 76% of the VAP cases, the same bacteria colonize the oral cavity and lungs. Oral care interventions may play a role in the prevention of VAP, yet more than half of the hospitals do not have specific policies for the oral care of intubated patients. Oral cavity interlinks with respiratory tracts and digestive tracts. After surgery has been performed in these areas, aerobic and anaerobic bacteria frequently induce operative wound infections in teeth, gingiva and supporting tissues of the teeth and tonsils. This study investigates the effects of a nanotechnology antimicrobial spray (JUC) on the incidence of VAP. 320 patients diagnosed with VAP were randomly divided into treatment and control groups. After using chlorhexidine mouthrinse, the treatment group used a nanotechnology antimicrobial spray to the nose and mouth. The control group was given normal saline. The incidence rate of VAP was significantly lower in the treatment (8.38%) than control group (54.24%) (p<0.01). A physical antimicrobial film is formed on the surface of oral and nasal mucosa after using the JUC spray which effectively reduces the microbial colonization in the sprayed areas, thus reducing and delaying the incidence of VAP. © 2011 Academic Journals.published_or_final_versio

    The Entropy for General Extremal Black Holes

    We use the Kerr/CFT correspondence to calculate the entropy for all known extremal stationary and axisymmetric black holes. This is done with the help of two ansatzs that are general enough to cover all such known solutions. Considering only the contribution from the Einstein-Hilbert action to the central charge(s), we find that the entropy obtained by using Cardy's formula exactly matches with the Bekenstein-Hawking entropy.Comment: Minor corrections, section 5 refined, references added

    A coupled optical-thermal-electrical model to predict the performance of hybrid PV/T-CCPC roof-top systems

    A crossed compound parabolic concentrator (CCPC) is applied into a photovoltaic/thermal (PV/T) hybrid solar collector, i.e. concentrating PV/T (CPV/T) collector, to develop new hybrid roof-top CPV/T systems. However, to optimise the system configuration and operational parameters as well as to predict their performances, a coupled optical, thermal and electrical model is essential. We establish this model by integrating a number of submodels sourced from literature as well as from our recent work on incidence-dependent optical efficiency, six-parameter electrical model and scaling law for outdoor conditions. With the model, electrical performance and cell temperature are predicted on specific days for the roof-top systems installed in Glasgow, Penryn and Jaen. Results obtained by the proposed model reasonably agree with monitored data and it is also clarified that the systems operate under off-optimal operating condition. Long-term electric performance of the CPV/T systems is estimated as well. In addition, effects of transient terms in heat transfer and diffuse solar irradiance on electric energy are identified and discussed

    Adaptive design methods in clinical trials – a review

    In recent years, the use of adaptive design methods in clinical research and development based on accrued data has become very popular due to its flexibility and efficiency. Based on adaptations applied, adaptive designs can be classified into three categories: prospective, concurrent (ad hoc), and retrospective adaptive designs. An adaptive design allows modifications made to trial and/or statistical procedures of ongoing clinical trials. However, it is a concern that the actual patient population after the adaptations could deviate from the originally target patient population and consequently the overall type I error (to erroneously claim efficacy for an infective drug) rate may not be controlled. In addition, major adaptations of trial and/or statistical procedures of on-going trials may result in a totally different trial that is unable to address the scientific/medical questions the trial intends to answer. In this article, several commonly considered adaptive designs in clinical trials are reviewed. Impacts of ad hoc adaptations (protocol amendments), challenges in by design (prospective) adaptations, and obstacles of retrospective adaptations are described. Strategies for the use of adaptive design in clinical development of rare diseases are discussed. Some examples concerning the development of Velcade intended for multiple myeloma and non-Hodgkin's lymphoma are given. Practical issues that are commonly encountered when implementing adaptive design methods in clinical trials are also discussed

    Malignancy risk analysis in patients with inadequate fine needle aspiration cytology (FNAC) of the thyroid

    Background Thyroid fine needle aspiration cytology (FNAC) is the standard diagnostic modality for thyroid nodules. However, it has limitations among which is the incidence of non-diagnostic results (Thy1). Management of cases with repeatedly non-diagnostic FNAC ranges from simple observation to surgical intervention. We aim to evaluate the incidence of malignancy in non-diagnostic FNAC, and the success rate of repeated FNAC. We also aim to evaluate risk factors for malignancy in patients with non-diagnostic FNAC. Materials and Methods Retrospective analyses of consecutive cases with thyroid non diagnostic FNAC results were included. Results Out of total 1657 thyroid FNAC done during the study period, there were 264 (15.9%) non-diagnostic FNAC on the first attempt. On repeating those, the rate of a non-diagnostic result on second FNAC was 61.8% and on third FNAC was 47.2%. The overall malignancy rate in Thy1 FNAC was 4.5% (42% papillary, 42% follicular and 8% anaplastic), and the yield of malignancy decreased considerably with successive non-diagnostic FNAC. Ultrasound guidance by an experienced head neck radiologist produced the lowest non-diagnostic rate (38%) on repetition compared to US guidance by a generalist radiologist (65%) and by non US guidance (90%). Conclusions There is a low risk of malignancy in patients with a non-diagnostic FNAC result, commensurate to the risk of any nodule. The yield of malignancy decreased considerably with successive non-diagnostic FNAC

    Pathogenic Mechanism of the FIG4 Mutation Responsible for Charcot-Marie-Tooth Disease CMT4J

    CMT4J is a severe form of Charcot-Marie-Tooth neuropathy caused by mutation of the phosphoinositide phosphatase FIG4/SAC3. Affected individuals are compound heterozygotes carrying the missense allele FIG4-I41T in combination with a null allele. Analysis using the yeast two-hybrid system demonstrated that the I41T mutation impairs interaction of FIG4 with the scaffold protein VAC14. The critical role of this interaction was confirmed by the demonstration of loss of FIG4 protein in VAC14 null mice. We developed a mouse model of CMT4J by expressing a Fig4-I41T cDNA transgene on the Fig4 null background. Expression of the mutant transcript at a level 5× higher than endogenous Fig4 completely rescued lethality, whereas 2× expression gave only partial rescue, providing a model of the human disease. The level of FIG4-I41T protein in transgenic tissues is only 2% of that predicted by the transcript level, as a consequence of the protein instability caused by impaired interaction of the mutant protein with VAC14. Analysis of patient fibroblasts demonstrated a comparably low level of mutant I41T protein. The abundance of FIG4-I41T protein in cultured cells is increased by treatment with the proteasome inhibitor MG-132. The data demonstrate that FIG4-I41T is a hypomorphic allele encoding a protein that is unstable in vivo. Expression of FIG4-I41T protein at 10% of normal level is sufficient for long-term survival, suggesting that patients with CMT4J could be treated by increased production or stabilization of the mutant protein. The transgenic model will be useful for testing in vivo interventions to increase the abundance of the mutant protein

    The Reform of Employee Compensation in China’s Industrial Enterprises

    Although employee compensation reform in Chinese industrial sector has been discussed in the literature, the real changes in compensation system and pay practices have received insufficient attention and warrant further examination. This paper briefly reviews the pre- and post-reform compensation system, and reports the results of a survey of pay practices in the four major types of industrial enterprises in China. The research findings indicate that the type of enterprise ownership has little influence on general compensation practices, adoption of profit-sharing plans, and subsidy and allowance packages. In general, pay is linked more to individual performance and has become an important incentive to Chinese employees. However, differences are found across the enterprise types with regard to performance-related pay. Current pay practices are positively correlated to overall effectiveness of the enterprise
