    Optimal scales to observe habitat dynamics: A coral reef example

    A new technique to estimate the characteristic length scales (CLSs) of real ecological systems provides an objective means to identify the optimal scale(s) of observation to best detect underlying dynamical trends. Application of the technique to natural systems has focused on identifying appropriate scales to measure the dynamics of species as descriptors of community and ecosystem dynamics. However, ecosystem monitoring is often based not on assessing single species, but on species assemblages, functional groups, or habitat types. We asked whether the concept of CLSs based on dynamic interactions among species could be extended to examine interactions among habitat types and thus to identify optimal scales for observing habitat dynamics. A time series of three spatial maps of benthic habitats on a Caribbean coral reef was constructed from aerial photographs, Compact Airborne Spectrographic Imager (CASI) images, and IKONOS satellite images, providing the short time sequence required for this technique. We estimated the CLS based on the dynamics of three distinct habitat types: dense stands of seagrass, sparse stands of seagrass, and Montastrea patch reefs. Despite notable differences in the areal extent of and relative change in these habitats over the 21-year observation period, analyses based on each habitat type indicated a similar CLS of similar to 300 m. We interpret the consistency of CLSs among habitats to indicate that the dynamics of the three habitat types are linked. The results are encouraging, and they indicate that CLS techniques can be used to identify the appropriate scale at which to monitor ecosystem trends on the basis of the dynamics of only one of a disparate suite of habitat types

    STOCHASTIC MODEL ANALYSIS OF THE IMPACT OF MEDIA CAMPAIGN ON TRANSMISSION OF COVID – 19 EPIDEMIC.

    The COVID - 19 pandemic is currently causing authorities and public health officials more concern. The goal of the project is to convert a deterministic model for COVID-19 transmissions to a stochastic model, and then analyze the results to see how media-driven awareness campaigns have an impact on the disease's spread. The dynamic COVID-19 model was converted to a stochastic model, which was then examined. The model includes the following categories: Susceptible (S), Exposed (E), Infected class (I),  Isolated class ( ), Aware class  and Recovered class (R), as well as the Cumulative density of awareness programs by media denoted by   . With the help of MATLAB, the converted model is then numerically solved using the Eula Maruyama approach, allowing the existence and uniqueness of the model to be examined. The implementation of awareness programs has been found to have a significant positive impact on the spread of COVID-19. As the rate of implementation of these programs rises, the population that is exposed to the virus and those who are infected with it declines, and it has been hypothesized that this will eventually cause COVID-19 to become extinct. According to the report, putting awareness campaigns into place can help stop the COVID-19 epidemic from spreading

    Identification of bacterial pathogens in dry bovine udder and their antimicrobial susceptibility

    Mastitis is a multi-etiological production disease of dairy cows worldwide. The likelihood of developing clinical mastitis in subsequent calving is enhanced by intramammary infections with pathogens during the late dry and post-calving period. There is an increase in the occurrence of intramammary infections during early and late dry period. Thirty apparently healthy, pregnant cross-bred dairy cows were screened for the presence of contagious mastitis pathogens in the secretions taken after 48 h of cessation of milking. A total of 118 bacterial isolates were obtained from the secretions collected from 108 quarters, and identified using morphological and cultural characteristics. Among them, 50.8 per cent were coagulase negative Staphylococci (CNS), 16 per cent were coagulase positive Staphylococci (CPS) and 27.1 per cent were Streptococci isolates. Micrococci and E. coli constituted 2.5 per cent each and Klebsiella spp. formed 0.8 per cent of the total isolates obtained. The CNS isolates identified included 22 S. chromogens, 14 S. xylosus, 12 S. saprophyticus, six S. hominis and S. epidermidis each. Among the CPS isolated 11.8 per cent were S. aureus and 4.2 per cent were non-aureus coagulase positive Staphylococcus which included three S. hyicus and two S. pseudointermedius. Molecular confirmation of S. aureus, CNS isolates, St. agalactiae were done by targeting nuc, cns and 16SrRNA gene respectively. Identification of bacterial pathogens that are present during dry period can help in formulating an effective dry cow therapy. Antibiogram studies of the isolates obtained were done using in vitro antibiotic sensitivity testing method

    Gcn5-Related N-Acetyltransferases (GNATs) With a Catalytic Serine Residue Can Play Ping-Pong Too

    Enzymes in the Gcn5-related N-acetyltransferase (GNAT) superfamily are widespread and critically involved in multiple cellular processes ranging from antibiotic resistance to histone modification. While acetyl transfer is the most widely catalyzed reaction, recent studies have revealed that these enzymes are also capable of performing succinylation, condensation, decarboxylation, and methylcarbamoylation reactions. The canonical chemical mechanism attributed to GNATs is a general acid/base mechanism; however, mounting evidence has cast doubt on the applicability of this mechanism to all GNATs. This study shows that the Pseudomonas aeruginosa PA3944 enzyme uses a nucleophilic serine residue and a hybrid ping-pong mechanism for catalysis instead of a general acid/base mechanism. To simplify this enzyme’s kinetic characterization, we synthesized a polymyxin B substrate analog and performed molecular docking experiments. We performed site-directed mutagenesis of key active site residues (S148 and E102) and determined the structure of the E102A mutant. We found that the serine residue is essential for catalysis toward the synthetic substrate analog and polymyxin B, but the glutamate residue is more likely important for substrate recognition or stabilization. Our results challenge the current paradigm of GNAT mechanisms and show that this common enzyme scaffold utilizes different active site residues to accomplish a diversity of catalytic reactions

    Disruption of thalamic functional connectivity is a neural correlate of dexmedetomidine-induced unconsciousness

    Understanding the neural basis of consciousness is fundamental to neuroscience research. Disruptions in cortico-cortical connectivity have been suggested as a primary mechanism of unconsciousness. By using a novel combination of positron emission tomography and functional magnetic resonance imaging, we studied anesthesia-induced unconsciousness and recovery using the α2-agonist dexmedetomidine. During unconsciousness, cerebral metabolic rate of glucose and cerebral blood flow were preferentially decreased in the thalamus, the Default Mode Network (DMN), and the bilateral Frontoparietal Networks (FPNs). Cortico-cortical functional connectivity within the DMN and FPNs was preserved. However, DMN thalamo-cortical functional connectivity was disrupted. Recovery from this state was associated with sustained reduction in cerebral blood flow and restored DMN thalamo-cortical functional connectivity. We report that loss of thalamo-cortical functional connectivity is sufficient to produce unconsciousness. DOI: http://dx.doi.org/10.7554/eLife.04499.00

    Evidence for a Prepore Stage in the Action of Clostridium perfringens Epsilon Toxin

    Clostridium perfringens epsilon toxin (ETX) rapidly kills MDCK II cells at 37°C, but not 4°C. The current study shows that, in MDCK II cells, ETX binds and forms an oligomeric complex equally well at 37°C and 4°C but only forms a pore at 37°C. However, the complex formed in MDCK cells treated with ETX at 4°C has the potential to form an active pore, since shifting those cells to 37°C results in rapid cytotoxicity. Those results suggested that the block in pore formation at 4°C involves temperature-related trapping of ETX in a prepore intermediate on the MDCK II cell plasma membrane surface. Evidence supporting this hypothesis was obtained when the ETX complex in MDCK II cells was shown to be more susceptible to pronase degradation when formed at 4°C vs. 37°C; this result is consistent with ETX complex formed at 4°C remaining present in an exposed prepore on the membrane surface, while the ETX prepore complex formed at 37°C is unaccessible to pronase because it has inserted into the plasma membrane to form an active pore. In addition, the ETX complex rapidly dissociated from MDCK II cells at 4°C, but not 37°C; this result is consistent with the ETX complex being resistant to dissociation at 37°C because it has inserted into membranes, while the ETX prepore readily dissociates from cells at 4°C because it remains on the membrane surface. These results support the identification of a prepore stage in ETX action and suggest a revised model for ETX cytotoxicity, i) ETX binds to an unidentified receptor, ii) ETX oligomerizes into a prepore on the membrane surface, and iii) the prepore inserts into membranes, in a temperature-sensitive manner, to form an active pore

    Knowledge, Practice, and Attitudes of Physicians in Low- and Middle-Income Countries on Fertility and Pregnancy-Related Issues in Young Women With Breast Cancer

    PURPOSE Fertility and pregnancy-related issues are highly relevant for young (≤ 40 years) patients with breast cancer. Limited evidence exists on knowledge, practice, and attitudes of physicians from low- and middle-income countries (LMICs) regarding these issues. METHODS A 19-item questionnaire adapted from an international survey exploring issues about fertility preservation and pregnancy after breast cancer was sent by e-mail between November 2019 and January 2020 to physicians from LMICs involved in breast cancer care. Descriptive analyses were performed. RESULTS A total of 288 physicians from Asia, Africa, America, and Europe completed the survey. Median age was 38 years. Responders were mainly medical oncologists (44.4%) working in an academic setting (46.9%). Among responders, 40.2% and 53.8% reported having never consulted the available international guidelines on fertility preservation and pregnancy after breast cancer, respectively. 25.0%, 19.1%, and 24.3% of responders answered to be not at all knowledgeable about embryo, oocyte, or ovarian tissue cryopreservation, respectively; 29.2%, 23.6%, and 31.3% declared that embryo, oocyte, and ovarian tissue cryopreservation were not available in their countries, respectively. 57.6% of responders disagreed or were neutral on the statement that controlled ovarian stimulation can be considered safe in patients with breast cancer. 49.7% and 58.6% of responders agreed or were neutral on the statement that pregnancy in breast cancer survivors may increase the risk of recurrence overall or only in those with hormone receptor–positive disease, respectively. CONCLUSION This survey showed suboptimal knowledge, practice, and attitudes of physicians from LMICs on fertility preservation and pregnancy after treatment completion in young women with breast cancer.
