    Salmonella Pathogenesis and Processing of Secreted Effectors by Caspase-3

    The enteric pathogen Salmonella enterica serovar Typhimurium causes food poisoning resulting in gastroenteritis. The S. Typhimurium effector Salmonella invasion protein A (SipA) promotes gastroenteritis by functional motifs that trigger either mechanisms of inflammation or bacterial entry. During infection of intestinal epithelial cells, SipA was found to be responsible for the early activation of caspase-3, an enzyme that is required for SipA cleavage at a specific recognition motif that divided the protein into its two functional domains and activated SipA in a manner necessary for pathogenicity. Other caspase-3 cleavage sites identified in S. Typhimurium appeared to be restricted to secreted effector proteins, which indicates that this may be a general strategy used by this pathogen for processing of its secreted effectors

    Optimizing end-labeled free-solution electrophoresis by increasing the hydrodynamic friction of the drag-tag

    We study the electrophoretic separation of polyelectrolytes of varying lengths by means of end-labeled free-solution electrophoresis (ELFSE). A coarse-grained molecular dynamics simulation model, using full electrostatic interactions and a mesoscopic Lattice Boltzmann fluid to account for hydrodynamic interactions, is used to characterize the drag coefficients of different label types: linear and branched polymeric labels, as well as transiently bound micelles. It is specifically shown that the label's drag coefficient is determined by its hydrodynamic size, and that the drag per label monomer is largest for linear labels. However, the addition of side chains to a linear label offers the possibility to increase the hydrodynamic size, and therefore the label efficiency, without having to increase the linear length of the label, thereby simplifying synthesis. The third class of labels investigated, transiently bound micelles, seems very promising for the usage in ELFSE, as they provide a significant higher hydrodynamic drag than the other label types. The results are compared to theoretical predictions, and we investigate how the efficiency of the ELFSE method can be improved by using smartly designed drag-tags.Comment: 32 pages, 11 figures, submitted to Macromolecule

    An investigation into the effects of solvent content on the image quality and stability of ink jet digital prints under varied storage conditions

    Increasing numbers of galleries, museums and archives are including ink jet printed materials into their collections, and therefore displays. There is evidence that the instability of these prints is such that images can suffer deterioration in print quality or in extreme cases, a loss of information over an extended period of time. This is shorter than the period typically required for perceptible deterioration to occur in many other paper-based artworks. The image stability of prints is affected by a number of factors some of which have already been studied. However the role played by the ink solvent in the loss of image quality has yet to be explored. This paper will outline research being undertaken to investigate the effects of solvent content which may increase/promote the loss in image quality of the hard copy prints when stored or displayed under a range of temperature and humidity conditions

    Is the sigma-1 receptor a potential pharmacological target for cardiac pathologies? A systematic review.

    Sigma-1 receptors are ligand-regulated chaperone proteins, involved in several cellular mechanisms. The aim of this systematic review was to examine the effects that the sigma-1 receptor has on the cardiovascular system. The interaction targets and proposed mechanisms of action of sigma-1 receptors were explored, with the aim of determining if the sigma-1 receptor is a potential pharmacological target for cardiac pathologies. This systematic review was conducted according to the PRISMA guidelines and these were used to critically appraise eligible studies. Pubmed and Scopus were systematically searched for articles investigating sigma-1 receptors in the cardiovascular system. Papers identified by the search terms were then subject to analysis against pre-determined inclusion criteria. 23 manuscripts met the inclusion criteria and were included in this review. The experimental platforms, experimental techniques utilised and the results of the studies were summarised. The sigma-1 receptor is found to be implicated in cardioprotection, via various mechanisms including stimulating the Akt-eNOS pathway, and reduction of Ca2 + leakage into the cytosol via modulating certain calcium channels. Sigma-1 receptors are also found to modulate other cardiac ion channels including different subtypes of potassium and sodium channels and have been shown to modulate intracardiac neuron excitability. The sigma-1 receptor is a potential therapeutic target for treatment of cardiac pathologies, particularly cardiac hypertrophy. We therefore suggest investigating the cardioprotective mechanisms of sigma-1 receptor function, alongside proposed potential ligands that can stimulate these functions

    An improved criterion for new particle formation in diverse atmospheric environments

    A dimensionless theory for new particle formation (NPF) was developed, using an aerosol population balance model incorporating recent developments in nucleation rates and measured particle growth rates. Based on this theoretical analysis, it was shown that a dimensionless parameter <i>L</i><sub>Γ</sub>, characterizing the ratio of the particle scavenging loss rate to the particle growth rate, exclusively determined whether or not NPF would occur on a particular day. This parameter determines the probability that a nucleated particle will grow to a detectable size before being lost by coagulation with the pre-existing aerosol. Cluster-cluster coagulation was shown to contribute negligibly to this survival probability under conditions pertinent to the atmosphere. Data acquired during intensive measurement campaigns in Tecamac (MILAGRO), Atlanta (ANARChE), Boulder, and Hyytiälä (QUEST II, QUEST IV, and EUCAARI) were used to test the validity of <i>L</i><sub>Γ</sub> as an NPF criterion. Measurements included aerosol size distributions down to 3 nm and gas-phase sulfuric acid concentrations. The model was applied to seventy-seven NPF events and nineteen non-events (characterized by growth of pre-existing aerosol without NPF) measured in diverse environments with broad ranges in sulfuric acid concentrations, ultrafine number concentrations, aerosol surface areas, and particle growth rates (nearly two orders of magnitude). Across this diverse data set, a nominal value of <i>L</i><sub>Γ</sub>=0.7 was found to determine the boundary for the occurrence of NPF, with NPF occurring when <i>L</i><sub>Γ</sub><0.7 and being suppressed when <i>L</i><sub>Γ</sub>>0.7. Moreover, nearly 45% of measured <i>L</i><sub>Γ</sub> values associated with NPF fell in the relatively narrow range of 0.1<<i>L</i><sub>Γ</sub><0.3

    Exploration Mission Particulate Matter Filtration Technology Performance Testing in a Simulated Spacecraft Cabin Ventilation System

    Human deep space exploration missions will require advances in long-life, low maintenance airborne particulate matter filtration technology. As one of the National Aeronautics and Space Administrations (NASA) developments in this area, a prototype of a new regenerable, multi-stage particulate matter filtration technology was tested in an International Space Station (ISS) module simulation facility. As previously reported, the key features of the filter system include inertial and media filtration with regeneration and in-place media replacement techniques. The testing facility can simulate aspects of the cabin environment aboard the ISS and contains flight-like cabin ventilation system components. The filtration technology test article was installed at the inlet of the central ventilation system duct and instrumented to provide performance data under nominal flow conditions. In-place regeneration operations were also evaluated. The real-time data included pressure drop across the filter stages, process air flow rate, ambient pressure, humidity and temperature. In addition, two video cameras positioned at the filtration technology test articles inlet and outlet were used to capture the mechanical performance of the filter media indexing operation under varying air flow rates. Recent test results are presented and future design recommendations are discussed

    High order amplitude equation for steps on creep curve

    We consider a model proposed by one of the authors for a type of plastic instability found in creep experiments which reproduces a number of experimentally observed features. The model consists of three coupled non-linear differential equations describing the evolution of three types of dislocations. The transition to the instability has been shown to be via Hopf bifurcation leading to limit cycle solutions with respect to physically relevant drive parameters. Here we use reductive perturbative method to extract an amplitude equation of up to seventh order to obtain an approximate analytic expression for the order parameter. The analysis also enables us to obtain the bifurcation (phase) diagram of the instability. We find that while supercritical bifurcation dominates the major part of the instability region, subcritical bifurcation gradually takes over at one end of the region. These results are compared with the known experimental results. Approximate analytic expressions for the limit cycles for different types of bifurcations are shown to agree with their corresponding numerical solutions of the equations describing the model. The analysis also shows that high order nonlinearities are important in the problem. This approach further allows us to map the theoretical parameters to the experimentally observed macroscopic quantities.Comment: LaTex file and eps figures; Communicated to Phys. Rev.
