    Photo-induced anticancer activity and singlet oxygen production of prodigiosenes

    © 2018 The Royal Society of Chemistry and Owner Societies. The photo-induced cytotoxicity of prodigiosenes is reported. One prodigiosene represents a synthetic analogue of the natural product prodigiosin, and two are conjugated to molecules that target the estrogen receptor (ER). A comparison of incubation and irradiation frameworks for the three prodigiosenes is reported, with activity against ER- and ER+ lines explored. Furthermore, the ability of the three prodigiosenes to photosensitise the production of singlet oxygen is demonstrated, shedding mechanistic light onto possible photodynamic therapeutic effects of this class of tripyrroles

    On the existence of deterministic chaos in the solar terrestrial environment

    Time series corresponding to F 10.7 solar flux, AE index and the ionospheric critical frequency f0F2 are analyzed, for periods of low solar activity, in order to find whether they show either deterministic behavior or stochastic noise. Both the correlation dimension and a lower bound of the Kolmogorov entropy are calculated. For the time series analyzed, deterministic chaos is found, and its implications are discussed.Se analizan series temporales correspondientes a: frecuencia 10.7 cm del flujo solar, índice AE y la frecuencia ionosférica crítica del sondeo vertical f0F2 para baja actividad solar, con el objeto de determinar si las variaciones en ellas presentan comportamiento determinístico o corresponden a ruido estocástico Se calculan la dimensión de correlación y el límite inferior de la entropía de Kolmogorov.     Science and Technology Issues in the 115th Congress

    Science and technology (S&T) have a pervasive influence over a wide range of issues confronting the nation. Public and private research and development spur scientific and technological advancement. Such advances can drive economic growth, help address national priorities, and improve health and quality of life. The constantly changing nature and ubiquity of science and technology frequently create public policy issues of congressional interest. The federal government supports scientific and technological advancement directly by funding and performing research and development and indirectly by creating and maintaining policies that encourage private sector efforts. Additionally, the federal government establishes and enforces regulatory frameworks governing many aspects of S&T activities. This report briefly outlines an array of science and technology policy issues that may come before the 115th Congress. Given the rapid pace of S&T advancement and its importance in many diverse public policy issues, S&T-related issues not discussed in this report may come before the 115th Congress. The selected issues are grouped into 9 categories: - Overarching S&T Policy Issues, - Agriculture, - Biomedical Research and Development, - Defense, - Energy, - Environment and Natural Resources, - Homeland Security, - Information Technology, - Physical and Material Sciences, and - Space. Each of these categories includes concise analysis of multiple policy issues. The material presented in this report should be viewed as illustrative rather than comprehensive. Each section identifies CRS reports, when available, and the appropriate CRS experts to contact for further information and analysis

    Coordinated Sumoylation and Ubiquitination Modulate EGF Induced EGR1 Expression and Stability

    Abstract: Background: Human early growth response-1 (EGR1) is a member of the zing-finger family of transcription factors induced by a range of molecular and environmental stimuli including epidermal growth factor (EGF). In a recently published paper we demonstrated that integrin/EGFR cross-talk was required for Egr1 expression through activation of the Erk1/2 and PI3K/Akt/Forkhead pathways. EGR1 activity and stability can be influenced by many different post-translational modifications such as acetylation, phosphorylation, ubiquitination and the recently discovered sumoylation. The aim of this work was to assess the influence of sumoylation on EGF induced Egr1 expression and/or stability. Methods: We modulated the expression of proteins involved in the sumoylation process in ECV304 cells by transient transfection and evaluated Egr1 expression in response to EGF treatment at mRNA and protein levels. Results: We demonstrated that in ECV304 cells Egr1 was transiently induced upon EGF treatment and a fraction of the endogenous protein was sumoylated. Moreover, SUMO-1/Ubc9 over-expression stabilized EGF induced ERK1/2 phosphorylation and increased Egr1 gene transcription. Conversely, in SUMO-1/Ubc9 transfected cells, EGR1 protein levels were strongly reduced. Data obtained from protein expression and ubiquitination analysis, in the presence of the proteasome inhibitor MG132, suggested that upon EGF stimuli EGR1 sumoylation enhanced its turnover, increasing ubiquitination and proteasome mediated degradation. Conclusions: Here we demonstrate that SUMO-1 modification improving EGR1 ubiquitination is involved in the modulation of its stability upon EGF mediated induction

    Instantaneous and time-averaged flow fields of multiple vortices in the tip region of a ducted propulsor

    The instantaneous and time-averaged flow fields in the tip region of a ducted marine propulsor are examined. In this flow, a primary tip-leakage vortex interacts with a secondary, co-rotating trailing edge vortex and other co- and counter-rotating vorticity found in the blade wake. Planar particle imaging velocimetry (PIV) is used to examine the flow in a plane approximately perpendicular to the mean axis of the primary vortex. An identification procedure is used to characterize multiple regions of compact vorticity in the flow fields as series of Gaussian vortices. Significant differences are found between the vortex properties from the time-averaged flow fields and the average vortex properties identified in the instantaneous flow fields. Variability in the vortical flow field results from spatial wandering of the vortices, correlated fluctuations of the vortex strength and core size, and both correlated and uncorrelated fluctuations in the relative positions of the vortices. This variability leads to pseudo-turbulent velocity fluctuations. Corrections for some of this variability are performed on the instantaneous flow fields. The resulting processed flow fields reveal a significant increase in flow variability in a region relatively far downstream of the blade trailing edge, a phenomenon that is masked through the process of simple averaging. This increased flow variability is also accompanied by the inception of discrete vortex cavitation bubbles, which is an unexpected result, since the mean flow pressures in the region of inception are much higher than the vapor pressure of the liquid.     Frequency of oral mucositis and microbiological analysis in children with acute lymphoblastic leukemia treated with 0.12% chlorhexidine gluconate

    Tendo em vista o potencial de morbidade das complicações orais em pacientes com leucemia, este estudo avaliou as alterações clínicas e microbiológicas que ocorrem na mucosa bucal de crianças com leucemia linfoblástica aguda (LLA), submetidas à quimioterapia antineoplásica e administração profilática do gluconato de clorexidina 0,12%. A amostra foi constituída de 17 crianças de 2 a 12 anos, as quais foram submetidas a exame clínico da mucosa oral para a detecção de lesões bucais. Além disso, foi coletado material biológico das mucosas labial e jugal para análises microbiológicas. A mucosite oral foi observada em apenas 5 (29,4%) pacientes. A análise microbiológica revelou a presença de um número reduzido de microorganismos potencialmente patogênicos, como estafilococos coagulase-negativos (47%), Candida albicans (35,3%), Klebsiella pneumoniae (5,9%), Escherichia coli enteropatogênica (5,9%) e Stenotrophomonas maltophilia (5,9%). Pacientes com mucosite oral apresentaram uma maior freqüência de estafilococos coagulase-negativos (80%) quando comparados aos pacientes que exibiam mucosa oral normal (33,3%). Em conclusão, os resultados do presente estudo sugerem que o uso profilático do gluconato de clorexidina 0,12% reduz a freqüência de mucosite oral e de patógenos orais em crianças com LLA. Além disso, os presentes achados sugerem uma possível relação entre estafilococos coagulase-negativos e o desenvolvimento de mucosite oral. _________________________________________________________________________________________ ABSTRACT: In view of the morbidity potential of oral complications in patients with leukemia, this study evaluated the clinical and microbiological alterations that occur in the oral mucosa of children with acute lymphoblastic leukemia (ALL) undergoing antineoplastic chemotherapy and prophylactic administration of 0.12% chlorhexidine gluconate. The sample consisted of 17 children aged 2 to 12 years that underwent clinical examination of the oral mucosa for the detection of oral lesions. In addition, biological material was collected from labial and buccal mucosa for microbiological analysis. Oral mucositis was observed in only 5 (29.4%) patients. Microbiological analysis revealed a reduced number of potentially pathogenic microorganisms, such as coagulase-negative staphylococci (47%), Candida albicans (35.3%), Klebsiella pneumoniae (5.9%), enteropathogenic Escherichia coli (5.9%), and Stenotrophomonas maltophilia (5.9%). Patients with oral mucositis showed a higher frequency of coagulase-negative staphylococci (80%) when compared with patients with normal oral mucosa (33.3%). In conclusion, the results of the present study suggest that the prophylactic use of 0.12% chlorhexidine gluconate reduces the frequency of oral mucositis and oral pathogens in children with ALL. In addition, the present findings suggest a possible relationship between coagulase-negative staphylococci and the development of oral mucositis
