Selective nociceptor activation in volunteers by infrared diode laser
<p>Abstract</p> <p>Background</p> <p>Two main classes of peripheral sensory neurons contribute to thermal pain sensitivity: the unmyelinated C fibers and thinly myelinated Aδ fibers. These two fiber types may differentially underlie different clinical pain states and distinctions in the efficacy of analgesic treatments. Methods of differentially testing C and Aδ thermal pain are widely used in animal experimentation, but these methods are not optimal for human volunteer and patient use. Thus, this project aimed to provide psychophysical and electrophysiological evidence that whether different protocols of infrared diode laser stimulation, which allows for direct activation of nociceptive terminals deep in the skin, could differentially activate Aδ or C fiber thermonociceptors in volunteers.</p> <p>Results</p> <p>Short (60 ms), high intensity laser pulses (SP) evoked monomodal "pricking" pain which was not enhanced by topical capsaicin, whereas longer, lower power pulses (LP) evoked monomodal "burning" pain which was enhanced by topical capsaicin. SP also produced cortical evoked EEG potentials consistent with Aδ mediation, the amplitude of which was directly correlated with pain intensity but was not affected by topical capsaicin. LP also produced a distinct evoked potential pattern the amplitude of which was also correlated with pain intensity, which was enhanced by topical capsaicin, and the latency of which could be used to estimate the conduction velocity of the mediating nociceptive fibers.</p> <p>Conclusions</p> <p>Psychophysical and electrophysiological data were consistent with the ability of short high intensity infrared laser pulses to selectively produce Aδ mediated pain and of longer pulses to selectively produce C fiber mediated thermal pain. Thus, the use of these or similar protocols may be useful in developing and testing novel therapeutics based on the differential molecular mechanisms underlying activation of the two fiber types (e.g., TRPV1, TRPV2, etc). In addition, these protocol may be useful in determining the fiber mediation of different clinical pain types which may, in turn be useful in treatment choice.</p
The Effect of Map Boundary on Estimates of Landscape Resistance to Animal Movement
BACKGROUND: Artificial boundaries on a map occur when the map extent does not cover the entire area of study; edges on the map do not exist on the ground. These artificial boundaries might bias the results of animal dispersal models by creating artificial barriers to movement for model organisms where there are no barriers for real organisms. Here, we characterize the effects of artificial boundaries on calculations of landscape resistance to movement using circuit theory. We then propose and test a solution to artificially inflated resistance values whereby we place a buffer around the artificial boundary as a substitute for the true, but unknown, habitat. METHODOLOGY/PRINCIPAL FINDINGS: We randomly assigned landscape resistance values to map cells in the buffer in proportion to their occurrence in the known map area. We used circuit theory to estimate landscape resistance to organism movement and gene flow, and compared the output across several scenarios: a habitat-quality map with artificial boundaries and no buffer, a map with a buffer composed of randomized habitat quality data, and a map with a buffer composed of the true habitat quality data. We tested the sensitivity of the randomized buffer to the possibility that the composition of the real but unknown buffer is biased toward high or low quality. We found that artificial boundaries result in an overestimate of landscape resistance. CONCLUSIONS/SIGNIFICANCE: Artificial map boundaries overestimate resistance values. We recommend the use of a buffer composed of randomized habitat data as a solution to this problem. We found that resistance estimated using the randomized buffer did not differ from estimates using the real data, even when the composition of the real data was varied. Our results may be relevant to those interested in employing Circuitscape software in landscape connectivity and landscape genetics studies
Peripheral blood T-cell signatures from high-resolution immune phenotyping of γδ and αβ T-cells in younger and older subjects in the Berlin Aging Study II
Background Aging and latent infection with Cytomegalovirus (CMV) are thought
to be major factors driving the immune system towards immunosenescence,
primarily characterized by reduced amounts of naĂŻve T-cells and increased
memory T-cells, potentially associated with higher morbidity and mortality.
The composition of both major compartments, γδ as well as αβ T-cells, is
altered by age and CMV, but detailed knowledge of changes to the γδ subset is
currently limited. Results Here, we have surveyed a population of 73 younger
(23–35 years) and 144 older (62–85 years) individuals drawn from the Berlin
Aging Study II, investigating the distribution of detailed differentiation
phenotypes of both γδ and αβ T-cells. Correlation of frequencies and absolute
counts of the identified phenotypes with age and the presence of CMV revealed
a lower abundance of Vδ2-positive and a higher amount of Vδ1-positive cells.
We found higher frequencies of late-differentiated and lower frequencies of
early-differentiated cells in the Vδ1+ and Vδ1-Vδ2-, but not in the Vδ2+
populations in elderly CMV-seropositive individuals confirming the association
of these Vδ2-negative cells with CMV-immunosurveillance. We identified the
highest Vδ1:Vδ2 ratios in the CMV-seropositive elderly. The observed increased
CD4:CD8 ratios in the elderly were significantly lower in CMV-seropositive
individuals, who also possessed a lower naĂŻve and a larger late-differentiated
compartment of CD8+ αβ T-cells, reflecting the consensus in the literature.
Conclusions Our findings illustrate in detail the strong influence of CMV on
the abundance and differentiation pattern of γδ T-cells as well as αβ T-cells
in older and younger people. Mechanisms responsible for the phenotypic
alterations in the γδ T-cell compartment, associated both with the presence of
CMV and with age require further clarification

Protecting labor rights in preferential trade agreements: the role of trade unions, left governments, and skilled labor
This paper investigates variation in the design of labor provisions in preferential trade agreements (PTAs) by focusing on the power of trade unions, the role of government partisanship, and the relative strength of skilled labor. We expect strong trade unions and left-leaning governments to be associated with more, and more far-reaching labor provisions in PTAs. We also expect the strength of skilled workers relative to the strength of unskilled workers to negatively correlate with the depth of labor provisions in PTAs. In addition, the effect of trade unions should be conditional on both the presence of left government and democracy. We test these hypotheses relying on an original dataset of labor provisions included in 483 PTAs signed between 1990 and 2016. This dataset covers 140 different labor provisions that relate to six overarching dimensions. The quantitative analysis finds support for the expectations concerning the influence of trade unions and the role of a country’s skill profile
Analyses of least cost paths for determining effects of habitat types on landscape permeability: wolves in Poland
Determining ecological corridors is crucial for conservation efforts in fragmented habitats. Commonly employed least cost path (LCP) analysis relies on the underlying cost matrix. By using Ecological Niche Factor Analysis, we minimized the problems connected with subjective cost assessment or the use of presence/absence data. We used data on the wolf presence/absence in Poland to identify LCPs connecting patches of suitable wolf habitat, factors that influence patch occupancy, and compare LCPs between different genetic subpopulations. We found that a lower proportion of cities and roads surrounds the most densely populated patches. Least cost paths between areas where little dispersal takes place (i.e., leading to unpopulated patches or between different genetic subpopulations) ran through a higher proportion of roads and human settlements. They also crossed larger maximal distances over deforested areas. We propose that, apart from supplying the basis for direct conservation efforts, LCPs can be used to determine what factors might facilitate or hinder dispersal by comparing different subsets of LCPs. The methods employed can be widely applicable to gain more in-depth information on potential dispersal barriers for large carnivores
