    Multiple discrete soluble aggregates influence polyglutamine toxicity in a Huntington\u27s disease model system

    Huntington’s disease (HD) results from expansions of polyglutamine stretches (polyQ) in the huntingtin protein (Htt) that promote protein aggregation, neurodegeneration, and death. Since the diversity and sizes of the soluble Htt-polyQ aggregates that have been linked to cytotoxicity are unknown, we investigated soluble Htt-polyQ aggregates using analytical ultracentrifugation. Soon after induction in a yeast HD model system, non-toxic Htt-25Q and cytotoxic Htt-103Q both formed soluble aggregates 29S to 200S in size. Because current models indicate that Htt-25Q does not form soluble aggregates, reevaluation of previous studies may be necessary. Only Htt-103Q aggregation behavior changed, however, with time. At 6 hr mid-sized aggregates (33S to 84S) and large aggregates (greater than 100S) became present while at 24 hr primarily only mid-sized aggregates (20S to 80S) existed. Multiple factors that decreased cytotoxicity of Htt-103Q (changing the length of or sequences adjacent to the polyQ, altering ploidy or chaperone dosage, or deleting anti-aging factors) altered the Htt-103Q aggregation pattern in which the suite of mid-sized aggregates at 6 hr were most correlative with cytotoxicity. Hence, the amelioration of HD and other neurodegenerative diseases may require increased attention to and discrimination of the dynamic alterations in soluble aggregation processes

    An Ultra-Low-Power Oscillator with Temperature and Process Compensation for UHF RFID Transponder

    This paper presents a 1.28MHz ultra-low-power oscillator with temperature and process compensation. It is very suitable for clock generation circuits used in ultra-high-frequency (UHF) radio-frequency identification (RFID) transponders. Detailed analysis of the oscillator design, including process and temperature compensation techniques are discussed. The circuit is designed using TSMC 0.18μm standard CMOS process and simulated with Spectre. Simulation results show that, without post-fabrication calibration or off-chip components, less than ±3% frequency variation is obtained from –40 to 85°C in three different process corners. Monte Carlo simulations have also been performed, and demonstrate a 3σ deviation of about 6%. The power for the proposed circuitry is only 1.18µW at 27°C

    Robust Estimation of 3D Human Poses from a Single Image

    Human pose estimation is a key step to action recognition. We propose a method of estimating 3D human poses from a single image, which works in conjunction with an existing 2D pose/joint detector. 3D pose estimation is challenging because multiple 3D poses may correspond to the same 2D pose after projection due to the lack of depth information. Moreover, current 2D pose estimators are usually inaccurate which may cause errors in the 3D estimation. We address the challenges in three ways: (i) We represent a 3D pose as a linear combination of a sparse set of bases learned from 3D human skeletons. (ii) We enforce limb length constraints to eliminate anthropomorphically implausible skeletons. (iii) We estimate a 3D pose by minimizing the L1L_1-norm error between the projection of the 3D pose and the corresponding 2D detection. The L1L_1-norm loss term is robust to inaccurate 2D joint estimations. We use the alternating direction method (ADM) to solve the optimization problem efficiently. Our approach outperforms the state-of-the-arts on three benchmark datasets

    Statistical description of small quantum systems beyond weak-coupling limit

    An explicit expression is derived for the statistical description of small quantum systems, which are relatively-weakly and directly coupled to only small parts of their environments. The derived expression has a canonical form, but is given by a renormalized self-Hamiltonian of the studied system, which appropriately takes into account the influence of the system-environment interaction. In the case that the system has a narrow spectrum and the environment is sufficiently large, the modification to the self-Hamiltonian usually has a mean-field feature, given by an environmental average of the interaction Hamiltonian.     Two dimensional scaling of resistance in flux flow region in Tl2Ba2CaCu2O8Tl_2Ba_2CaCu_2O_8 thin films

    The resistance of Tl2Ba2CaCu2O8Tl_2Ba_2CaCu_2O_8 thin films has been measured when the angle between the applied fields and abab-plane of the film is changed continuously at various temperatures. Under various magnetic fields, the resistance can be well scaled in terms of the c-axis component of the applied fields at the same temperature in the whole angle range. Meanwhile, we show that the measurement of resistance in this way is a complementary method to determine the growth orientation of the anisotropic high-TcT_c superconductors.Comment: 11 pages, 8 figures.     Spin-dependent tunneling through a symmetric semiconductor barrier is studied including the k^3 Dresselhaus effect. The spin-dependent transmission of electron can be obtained analytically. By comparing with previous work(Phys. Rev. B 67. R201304 (2003) and Phys. Rev. Lett. 93. 056601 (2004)), it is shown that the spin polarization and interface current are changed significantly by including the off-diagonal elements in the current operator, and can be enhanced considerably by the Dresselhaus effect in the contact regions.Comment: 10 pages, 5 figures, to appear in PR

    A Coherent Ising Machine Based On Degenerate Optical Parametric Oscillators

    A degenerate optical parametric oscillator network is proposed to solve the NP-hard problem of finding a ground state of the Ising model. The underlying operating mechanism originates from the bistable output phase of each oscillator and the inherent preference of the network in selecting oscillation modes with the minimum photon decay rate. Computational experiments are performed on all instances reducible to the NP-hard MAX-CUT problems on cubic graphs of order up to 20.     A classification of 2D fermionic and bosonic topological orders

    The string-net approach by Levin and Wen, and the local unitary transformation approach by Chen, Gu, and Wen, provide ways to classify topological orders with gappable edge in 2D bosonic systems. The two approaches reveal that the mathematical framework for 2+1D bosonic topological order with gappable edge is closely related to unitary fusion category theory. In this paper, we generalize these systematic descriptions of topological orders to 2D fermion systems. We find a classification of 2+1D fermionic topological orders with gappable edge in terms of the following set of data (Nkij,Fkij,Fjkn,χδijm,αβ,di)(N^{ij}_k, F^{ij}_k, F^{ijm,\alpha\beta}_{jkn,\chi\delta},d_i), that satisfy a set of non-linear algebraic equations. The exactly soluble Hamiltonians can be constructed from the above data on any lattices to realize the corresponding topological orders. When Fkij=0F^{ij}_k=0, our result recovers the previous classification of 2+1D bosonic topological orders with gappable edge.Comment: 19 page 5 figures, RevTeX
