    Marriage in the reformed tradition in Scotland with particular reference to the Scottish Synod of the URC

    Drawing on a foundation of the history of Reformed marriage traditions and theology, as it has bearing on the Scottish Synod of the United Reformed Church, this dissertation aims to analyse the perspectives of members and clergy of the Synod on marriage, and offer a pastoral theology with which to make progress in the ongoing, topical, conversation around human sexuality. In particular, the aim is to unpack “what we mean by marriage”, how we discern Godʼs will and presence in our human relationships, and how the Church may better serve its members, in a world whose values at times appear somewhat at odds with those of the Church. The dissertation engages with the questions of scripture and tradition, creation ordinance and Covenant, and the ways that love and intimacy offer the potential for reconciliation with self, other and God. I will argue that marital-type relationships exist beyond the confines of the institution, as it is currently formulated, and utilise Paul Tillichʼs existentialist approach to argue that the moralism of Law must be superseded by the morality of Grace, as we uphold a progressive Reformed approach to human relationships. I will propose that the United Reformed Church, in Scotland and denominationally, has a responsibility to continue to reform, congruent with its tradition, and respond faithfully to the fresh ways in which the Spirit is revealing the Word of God and calling of the Church, in ever developing cultural contexts, specifically with the case of marriage. In practice, the empirical research of this dissertation will show that this process of ongoing reform is already at work within the Synod, both in the changing dynamics of relationship models present in our midst, and the level of faithful conversation, reflection and discernment taking place, at all levels of the Synod. Therefore, this dissertation will propose that further work must be undertaken, in the area of Reformed pastoral theology and sexual ethics, building upon the findings presented here. I believe that it is the responsibility of the Church to formulate theology and practice that encourages its members towards finding and maintaining healthy and stable relationships, no matter their classification, that may offer glimpses of the New Kingdom, as they foster reconciliation with Self, Other and God. This will require a reanalysis of dogma, tradition and the world we live in today, and require a good deal of courage

    The VMC survey - XIV : First results on the look-back time star formation rate tomography of the Small Magellanic Cloud

    Date of Acceptance: 20/01/2015We analyse deep images from the VISTA survey of the Magellanic Clouds in the YJKs filters, covering 14 deg2 (10 tiles), split into 120 subregions, and comprising the main body and Wing of the Small Magellanic Cloud (SMC). We apply a colour-magnitude diagram reconstruction method that returns their best-fitting star formation rate SFR(t), age-metallicity relation (AMR), distance and mean reddening, together with 68 per cent confidence intervals. The distance data can be approximated by a plane tilted in the East-West direction with a mean inclination of 39°, although deviations of up to ±3 kpc suggest a distorted and warped disc. After assigning to every observed star a probability of belonging to a given age-metallicity interval, we build high-resolution population maps. These dramatically reveal the flocculent nature of the young star-forming regions and the nearly smooth features traced by older stellar generations. They document the formation of the SMC Wing at ages <0.2 Gyr and the peak of star formation in the SMC Bar at ~40 Myr. We clearly detect periods of enhanced star formation at 1.5 and 5 Gyr. The former is possibly related to a new feature found in the AMR, which suggests ingestion of metal-poor gas at ages slightly larger than 1 Gyr. The latter constitutes a major period of stellar mass formation. We confirm that the SFR(t) was moderately low at even older ages.Peer reviewe

    Linear modeling of possible mechanisms for parkinson tremor generation

    The power of Parkinson tremor is expressed in terms of possibly changed frequency response functions between relevant variables in the neuromuscular system. The derivation starts out from a linear loopless equivalent model of mechanisms for general tremor generation. Hypothetical changes in this model from the substrate of the disease are indicated, and possible ones are inferred from literature about experiments on patients. The result indicates that in these patients tremor appears to have been generated in loops, which did not include the brain area which in surgery usually is inactivated. For some patients in the literature, these loops could involve muscle length receptors, the static sensitivity of which may have been enlarged by pathological brain activity

    The VMC survey - XV : The Small Magellanic Cloud-Bridge connection history as traced by their star cluster populations

    Date of Acceptance: 19/03/2015We present results based on YJKs photometry of star clusters located in the outermost, eastern region of the Small Magellanic Cloud (SMC). We analysed a total of 51 catalogued clusters whose colour-magnitude diagrams (CMDs), having been cleaned from field-star contamination, were used to assess the clusters' reality and estimate ages of the genuine systems. Based on CMD analysis, 15 catalogued clusters were found to be possible non-genuine aggregates. We investigated the properties of 80 per cent of the catalogued clusters in this part of the SMC by enlarging our sample with previously obtained cluster ages, adopting a homogeneous scale for all. Their spatial distribution suggests that the oldest clusters, log(t/yr) ≥ 9.6, are in general located at greater distances to the galaxy's centre than their younger counterparts - 9.0 ≤ log(t/yr) ≤ 9.4 - while two excesses of clusters are seen at log(t/yr) ~9.2 and log(t yr-1) ˜ 9.7. We found a trail of younger clusters which follow the wing/bridge components. This long spatial sequence does not only harbour very young clusters, log(t yr-1) ~7.3, but it also hosts some of intermediate ages, log(t/yr) ~9.1. The derived cluster and field-star formation frequencies as a function of age are different. The most surprising feature is an observed excess of clusters with ages of log(t/yr) < 9.0, which could have been induced by interactions with the LMC.Peer reviewedFinal Accepted Versio

    Growth disrupting mutations in epigenetic regulatory molecules are associated with abnormalities of epigenetic aging.

    Germline mutations in fundamental epigenetic regulatory molecules including DNA methyltransferase 3 alpha (DNMT3A) are commonly associated with growth disorders, whereas somatic mutations are often associated with malignancy. We profiled genome-wide DNA methylation patterns in DNMT3A c.2312G > A; p.(Arg771Gln) carriers in a large Amish sibship with Tatton-Brown-Rahman syndrome (TBRS), their mosaic father, and 15 TBRS patients with distinct pathogenic de novo DNMT3A variants. This defined widespread DNA hypomethylation at specific genomic sites enriched at locations annotated as genes involved in morphogenesis, development, differentiation, and malignancy predisposition pathways. TBRS patients also displayed highly accelerated DNA methylation aging. These findings were most marked in a carrier of the AML-associated driver mutation p.Arg882Cys. Our studies additionally defined phenotype-related accelerated and decelerated epigenetic aging in two histone methyltransferase disorders: NSD1 Sotos syndrome overgrowth disorder and KMT2D Kabuki syndrome growth impairment. Together, our findings provide fundamental new insights into aberrant epigenetic mechanisms, the role of epigenetic machinery maintenance, and determinants of biological aging in these growth disorders

    Systematic profiling of DNMT3A variants reveals protein instability mediated by the DCAF8 E3 ubiquitin ligase adaptor

    Clonal hematopoiesis is a prevalent age-related condition associated with greatly increased risk of hematologic disease; mutations in DNA methyltransferase 3A (DNMT3A) are the most common driver of this state. DNMT3A variants occur across the gene with some particularly associated with malignancy, but the functional relevance and mechanisms of pathogenesis of the majority of mutations is unknown. Here, we systematically investigated the methyltransferase activity and protein stability of 253 disease-associated DNMT3A mutations, finding that 74% were loss-of-function mutations. Half of these variants exhibited reduced protein stability and, as a class, correlated with greater clonal expansion and AML development. We investigated the mechanisms underlying the instability using a CRISPR screen and uncovered regulated destruction of DNMT3A mediated by the DCAF8 E3 ubiquitin ligase adaptor. We establish a new paradigm to classify novel variants that has prognostic and potential therapeutic significance for patients with hematologic disease
