    G protein-coupled receptors (GPCRs) regulate many animal behaviors. GPCR signaling is mediated by agonist-promoted interactions of GPCRs with heterotrimeric G proteins, GPCR kinases (GRKs), and arrestins. To further elucidate the role of GRKs in regulating GPCR-mediated behaviors, we utilized the genetic model system Caenorhabditis elegans Our studies demonstrate that grk-2 loss-of-function strains are egg laying-defective and contain low levels of serotonin (5-HT) and high levels of the 5-HT metabolite 5-hydroxyindole acetic acid (5-HIAA). The egg laying defect could be rescued by the expression of wild type but not by catalytically inactive grk-2 or by the selective expression of grk-2 in hermaphrodite-specific neurons. The addition of 5-HT or inhibition of 5-HT metabolism also rescued the egg laying defect. Furthermore, we demonstrate that AMX-2 is the primary monoamine oxidase that metabolizes 5-HT in C. elegans, and we also found that grk-2 loss-of-function strains have abnormally high levels of AMX-2 compared with wild-type nematodes. Interestingly, GRK-2 was also found to interact with and promote the phosphorylation of AMX-2. Additional studies reveal that 5-HIAA functions to inhibit egg laying in a manner dependent on the 5-HT receptor SER-1 and the G protein GOA-1. These results demonstrate that GRK-2 modulates 5-HT metabolism by regulating AMX-2 function and that 5-HIAA may function in the SER-1 signaling pathway

    Assimilating GCOM-W1 AMSR2 and TRMM TMI Radiance Data in GEOS Analysis and Reanalysis

    The Tropical Rainfall Measurement Mission (TRMM) Microwave Imager (TMI) observed the Earth in lower latitudes between 1997 - 2015. Its conical-scan radiometer has nine channels and measured microwave radiances between 10 and 89 GHz. These data provide information on atmospheric temperature, humidity, clouds, precipitation, as well as sea surface temperature. Radiance data from other microwave radiometers such as Special Sensor Microwave Imager (SSM/I) and Special Sensor Microwave Imager Sounder (SSMIS) onboard various Defense Meteorological Satellite Program (DMSP) satellites are assimilated in clear-sky conditions in the Modern-Era Retrospective analysis for Research and Applications (MERRA) and its version 2 (MERRA-2) data sets at the Global Modeling and Assimilation Office (GMAO) at NASA Goddard Space Flight Center. The GMAO's Hybrid 4D-EnVar-based Atmospheric Data Assimilation System (ADAS) is enhanced with an all-sky microwave radiance data assimilation capability in the real-time GEOS-Forward Processing (FP) system. Currently, the FP system assimilates Global Precipitation Measurement (GPM) microwave imager (GMI) radiance data utilizing this all-sky capability, and is being extended to use more all-sky data from other microwave radiometers. In this presentation, we will focus on impacts of all-sky TMI radiance data on GEOS analyses of atmospheric moisture, precipitation and other fields, and discuss their applications for future GEOS reanalyses

    Translocator protein in late stage Alzheimer\u27s disease and Dementia with Lewy bodies brains

    OBJECTIVE: Increased translocator protein (TSPO), previously known as the peripheral benzodiazepine receptor (PBR), in glial cells of the brain has been used as a neuroinflammation marker in the early and middle stages of neurodegenerative diseases, such as Alzheimer\u27s disease (AD) and Dementia with Lewy Bodies (DLB). In this study, we investigated the changes in TSPO density with respect to late stage AD and DLB. METHODS: TSPO density was measured in multiple regions of postmortem human brains in 20 different cases: seven late stage AD cases (Braak amyloid average: C; Braak tangle average: VI; Aged 74-88, mean: 83 ± 5 years), five DLB cases (Braak amyloid average: C; Braak tangle average: V; Aged 79-91, mean: 84 ± 4 years), and eight age-matched normal control cases (3 males, 5 females: aged 77-92 years; mean: 87 ± 6 years). Measurements were taken by quantitative autoradiography using [ RESULTS: No significant changes were found in TSPO density of the frontal cortex, striatum, thalamus, or red nucleus of the AD and DLB brains. A significant reduction in TSPO density was found in the substantia nigra (SN) of the AD and DLB brains compared to that of age-matched healthy controls. INTERPRETATION: This distinct pattern of TSPO density change in late stage AD and DLB cases may imply the occurrence of microglia dystrophy in late stage neurodegeneration. Furthermore, TSPO may not only be a microglia activation marker in early stage AD and DLB, but TSPO may also be used to monitor microglia dysfunction in the late stage of these diseases

    An Archival Chandra and XMM-Newton Survey of Type 2 Quasars

    In order to investigate obscuration in high-luminosity type 2 active galactic nuclei (AGNs), we analyzed Chandra and XMM-Newton archival observations for 71 type 2 quasars detected at 0.05 100 eV in the rest frame) and we detect this line in the other sources through a joint fit (spectral stacking). The correlation between the Fe K alpha and [O III] fluxes and the inverse correlation of the equivalent width of the Fe K line with the ratio of hard X-ray and [O III] fluxes is consistent with previous results for lower luminosity Seyfert 2 galaxies. We conclude that obscuration is the cause of the weak hard X-ray emission rather than intrinsically low X-ray luminosities. We find that about half of the population of optically selected type 2 quasars are likely to be Compton thick. We also find no evidence that the amount of X-ray obscuration depends on the AGN luminosity (over a range of more than three orders of magnitude in luminosity)
