    Land remote sensing commercialization: A status report

    The current offer by the United States Department of Commerce to transfer the U.S. land remote sensing program to the private sector is described. A Request for Proposals (RFP) was issued, soliciting offers from U.S. firms to provide a commercial land remote sensing satellite system. Proposals must address a complete system including satellite, communications, and ground data processing systems. Offerors are encouraged to propose to take over the Government LANDSAT system which consists of LANDSAT 4 and LANDSAT D'. Also required in proposals are the market development procedures and plans to ensure that commercialization is feasible and the business will become self-supporting at the earliest possible time. As a matter of Federal Policy, the solicitation is designed to protect both national security and foreign policy considerations. In keeping with these concerns, an offeror must be a U.S. Firm. Requirements for data quality, quantity, distribution and delivery are met by current operational procedures. It is the Government's desire that the Offeror be prepared to develop and operate follow-on systems without Government subsidies. However, to facilitate rapid commercialization, an offeror may elect to include in his proposal mechanisms for short term government financial assistance

    Molecular dissection of telomere dysfunction and analysis of G-overhangs in Arabidopsis thaliana

    Telomeres comprise the physical ends of chromosomes. In the absence of telomerase, the enzyme responsible for replenishing telomeric DNA, telomeres progressively shorten due to the end replication problem. Eventually telomeres reach a length where they are recruited into end-to-end chromosome fusions. Through the use of novel PCR strategies, I followed the fate of telomeres in plants lacking telomerase as they progressed into dysfunction. I uncovered two distinct structural/functional length transitions. The first transition (~1 kb) marks the onset of telomere dysfunction, where telomeres are transiently uncapped and a subset of them engage in end-to-end fusions. The second transition (~300 bp) defines complete telomere dysfunction as telomeres below this length lack G-overhangs and the vast majority of the chromosome ends fuse. Thus, these two telomere lengths define architectural transitions that link structure and function. In addition, I uncovered a hierarchy of end-joining pathways that join dysfunctional telomeres in which the non-homologous end-joining (NHEJ) protein, KU predominates. In the absence of KU, telomeres are joined by a microhomologymediated end-joining pathway (MMEJ) that is dependent on Mre11. I also show that DNA ligase IV (LIG4) is the predominant enzyme that ligates dysfunctional telomeres as fusions are reduced in its absence. These studies highlight the importance of repairing DSBs and demonstrate that Arabidopsis possesses highly redundant means for processing dysfunctional telomeres. The G-overhang is an essential feature of the telomere that is required for proper telomere function. I employed methods to examine G-overhang status in various mutants known to contribute to telomere maintenance in Arabidopsis. My analysis revealed that the putative G-overhang binding proteins POT1a, POT1b and POT1c, make modest, but distinct contributions to the G-overhangs. Additionally, I uncovered a major role for the putative telomere capping protein, CIT1 in maintenance of the Goverhang. G-overhang signals obtained from cit1 mutants were grossly increased indicating that CIT1 is involved in either protecting the C-rich strand of the telomere from nuclease attack, or in controlling telomerase extension of the G-strand. Together, these data have provided new insight into factors that contribute to telomere integrity and have further developed Arabidopsis as a model for telomere biology

    This report summarizes attributes, costs, and returns for organic dairy farms in Maine that responded to the 2008 dairy cost of production survey. This survey and analysis was conducted by the University of Maine in cooperation with the Maine Milk Commission. This publication reports on data collected over the 2007 production year. Analysis and discussion of the data revolve around four categories. The first category averages all 30 organic farms to create a statewide group. The authors then broke these 30 farms into three size groups (small, medium, and large) based on the number of cows on each farm. There were nine small farms with an average of 30 cows on each farm, 10 medium farms with an average of 55 cows each, and 11 large farms with about an average of 100 cows each.     The integration of postmodern values and rhetorical analysis : a case study

    Both traditional preaching theory and the listening context of the hearers have undergone radical changes within the last thirty years. Contemporary preachers no longer can assume the authority inherent in their position or preaching methods, and postmodern listeners exhibit the desire for increased diversity and points of view in sermons. This thesis will address these challenges by advancing the notion that attention to rhetorical criticism in the exegesis of biblical texts sheds new light on the nature of preaching in terms of form and function. The resulting multi-vocal and non-hierarchical leadership orientation has application for postmodern audiences. The methodological structure of theological interpretation undergirding this thesis involves four tasks of the hermeneutical cycle adapted from Richard Osmer’s approach. This approach engages in the task of contextual interpretation that connects with both Christian tradition and Scripture, and furthermore leads to the construction of a pragmatic plan for future homiletics. Chapter 1 introduces the problem facing contemporary homileticians: the changed context of preacher and hearer. The chapter advocates that one way forward for preaching involves the use of rhetorical criticism as the exegetical basis for a values-based homiletic, and then finishes with an overview of the thesis chapters. Chapter 2 demonstrates the fourfold task of the hermeneutical cycle by establishing the provenance of the method, critiquing it and grounding the approach of the thesis in the contemporary postmodern setting. Chapter 3 engages in a contextual interpretation of historic shifts in the fields of rhetoric, biblical studies and homiletics, analyzing and evaluating these trends. The chapter concludes by constructing a pragmatic plan for future biblical studies, a rhetorical-critical-narratological methodology that will be applied to the text of Ezekiel. Chapter 4 demonstrates that a contextual interpretation, evaluation and analysis of the New Homiletic results in the formation of a values-based approach to preaching and leadership orientation that is appropriate to postmodernity. Chapter 5 builds upon a contextual interpretation of synchronic and diachronic methodologies and advances a complementary approach to exegesis. The chapter then applies the rhetorical-critical-narratological approach developed in Chapter 3 to the discourse of Ezekiel to establish its contextual and rhetorical situation. The chapter then engages in a close rhetorical-critical-narratological reading of the literary unit of Ezekiel 15. Chapter 6 engages in a contextual interpretation and evaluation of three Ezekiel commentaries and sermons from Ezekiel 15, locating them along the pendulum-like series of shifts identified within Chapter 3. Chapter 7 demonstrates the integration of biblical studies and homiletics with the production of a sample multiple point-of-view sermon based upon the exegesis of Ezekiel conducted in Chapter 5. The chapter critiques the sermon and provides an example of the rhetorical-critical method applied to a discursive genre from 1 Corinthians 4.18-5.13.     Telomere dynamics and fusion of critically shortened telomeres in plants lacking DNA ligase IV

    In the absence of the telomerase, telomeres undergo progressive shortening and are ultimately recruited into end-to-end chromosome fusions via the non-homologous end joining (NHEJ) double-strand break repair pathway. Previously, we showed that fusion of critically shortened telomeres in Arabidopsis proceeds with approximately the same efficiency in the presence or absence of KU70, a key component of NHEJ. Here we report that DNA ligase IV (LIG4) is also not essential for telomere joining. We observed only a modest decrease (3-fold) in the frequency of chromosome fusions in triple tert ku70 lig4 mutants versus tert ku70 or tert. Sequence analysis revealed that, relative to tert ku70, chromosome fusion junctions in tert ku70 lig4 mutants contained less microhomology and less telomeric DNA. These findings argue that the KU-LIG4 independent end-joining pathway is less efficient and mechanistically distinct from KU-independent NHEJ. Strikingly, in all the genetic backgrounds we tested, chromosome fusions are initiated when the shortest telomere in the population reaches ∼1 kb, implying that this size represents a critical threshold that heralds a detrimental structural transition. These data reveal the transitory nature of telomere stability, and the robust and flexible nature of DNA repair mechanisms elicited by telomere dysfunction

    Response of neural reward regions to food cues in autism spectrum disorders

    BACKGROUND: One hypothesis for the social deficits that characterize autism spectrum disorders (ASD) is diminished neural reward response to social interaction and attachment. Prior research using established monetary reward paradigms as a test of non-social reward to compare with social reward may involve confounds in the ability of individuals with ASD to utilize symbolic representation of money and the abstraction required to interpret monetary gains. Thus, a useful addition to our understanding of neural reward circuitry in ASD includes a characterization of the neural response to primary rewards. METHOD: We asked 17 children with ASD and 18 children without ASD to abstain from eating for at least four hours before an MRI scan in which they viewed images of high-calorie foods. We assessed the neural reward network for increases in the blood oxygenation level dependent (BOLD) signal in response to the food images RESULTS: We found very similar patterns of increased BOLD signal to these images in the two groups; both groups showed increased BOLD signal in the bilateral amygdala, as well as in the nucleus accumbens, orbitofrontal cortex, and insula. Direct group comparisons revealed that the ASD group showed a stronger response to food cues in bilateral insula along the anterior-posterior gradient and in the anterior cingulate cortex than the control group, whereas there were no neural reward regions that showed higher activation for controls than for ASD. CONCLUSION: These results suggest that neural response to primary rewards is not diminished but in fact shows an aberrant enhancement in children with ASD
