    Spring Hydrology of Colorado Bend State Park, Central Texas

    Karst development in Ellenburger carbonates near Colorado Bend State Park in central Texas exhibits complex polygenetic origins, with porosity development dominated by an early hypogene phase that has subsequently been overprinted to varying degrees by epigene processes. Quarterly physicochemical and continuous thermal monitoring analyses of eight springs in the study area indicate that modern groundwater flow paths are highly variable. Springs exhibit patterns that range from shallow, distributed recharge into diffuse-flow dominated systems, to focused recharge into well-connected conduit systems, to deep-circulation systems that equilibrate with bedrock. All springs, except Sulphur Spring, exhibit physicochemical characteristics indicative of proximal epigenic groundwater flow through Ellenburger carbonates, while Sulphur Spring shows elevated temperature and dissolved-ion concentrations indicative of longer groundwater flow paths through deeper strata. The polygenetic nature of karst development in the Colorado Bend State Park has created an enhanced porosity structure which forms a complex modern groundwater flow network

    This qualitative content analysis study examined the framing of equity audits and the Texas Accountability Intervention System (TAIS) plans implemented by aspiring principal fellows to develop a support system for marginalized students. The purpose of this study was to demonstrate how a principal preparation program leverages equity-driven data to support the learning and engagement of all students, with an emphasis on supporting English Language Learners (ELL)s and Special Education (SPED) students.     Proposed method for predicting pair matching of skeletal elements allows too many false rejections

    Byrd proposes a method for predicting pair matches in commingled remains to reduce visual comparison. The method compares differences between left and right postcranial element measurements in commingled samples with differences in known pairs from a reference sample using a t-score approach. We duplicated his protocol using six elements from two samples of known paired elements (n=854 to 1063) and calculated the number of pairs correctly predicted. Time commitment was estimated by mathematically attempting matches with all left and right elements in these samples. Although the results show an 86% reduction in the number of potential pairs requiring visual matching, we do not recommend the method because (i) the normality assumption for use of a t-score approach is violated, (ii) no account is made for bilateral asymmetry, and (iii) the high rate of false rejections (up to 22%) undermines its ability to show true incompatibilities for potential pair matches

    Circulating Endothelial Microparticles in Diabetes Mellitus

    Background. Endothelial Microparticles (EMPs) are small vesicles shed from activated or apoptotic endothelial cells and involved in cellular cross-talk. Whether EMP immunophenotypes vary according to stimulus in Diabetes Mellitus (DM) is not known. We studied the cellular adhesion molecule (CAM) profile of circulating EMPs in patients with and without Diabetes Mellitus type 2, who were undergoing elective cardiac catheterization. Methods and Results. EMPs were analyzed by flow cytometry. The absolute median number of EMPs (EMPs/μL) specific for CD31, CD105, and CD106 was significantly increased in the DM population. The ratio of CD62E/CD31 EMP populations reflected an apoptotic process. Conclusion. Circulating CD31+, CD105+, and CD106+ EMPs were significantly elevated in patients with DM. EMPs were the only independent predictors of DM in our study cohort. In addition, the EMP immunophenotype reflected an apoptotic process. Circulating EMPs may provide new options for risk assessment

    Increasing compliance with wearing a medical device in children with autism

    Health professionals often recommend the use of medical devices to assess the health, monitor the well-being, or improve the quality of life of their patients. Children with autism may present challenges in these situations as their sensory peculiarities may increase refusals to wear such devices. To address this issue, we systematically replicated prior research by examining the effects of differential reinforcement of other behavior (DRO) to increase compliance with wearing a heart rate monitor in 2 children with autism. The intervention increased compliance to 100% for both participants when an edible reinforcer was delivered every 90 s. The results indicate that DRO does not require the implementation of extinction to increase compliance with wearing a medical device. More research is needed to examine whether the reinforcement schedule can be further thinned

    Spam1-associated transmission ratio distortion in mice: Elucidating the mechanism

    BACKGROUND: While transmission ratio distortion, TRD, (a deviation from Mendelian ratio) is extensive in humans and well-documented in mice, the underlying mechanisms are unknown. Our earlier studies on carriers of spontaneous mutations of mouse Sperm Adhesion Molecule 1 (Spam1) suggested that TRD results from biochemically different sperm, due to a lack of transcript sharing through the intercellular cytoplasmic bridges of spermatids. These bridges usually allow transcript sharing among genetically different spermatids which develop into biochemically and functionally equivalent sperm. OBJECTIVES: The goals of the study were to provide support for the lack of sharing (LOS) hypothesis, using transgene and null carriers of Spam1, and to determine the mechanism of Spam1-associated TRD. METHODS: Carriers of Spam1-Hyal5 BAC transgenes were mated with wild-type female mice and the progeny analyzed for TRD by PCR genotyping. Sperm from transgene and Spam1 null carriers were analyzed using flow cytometry and immunocytochemistry to detect quantities of Spam1 and/or Hyal5. Transgene-bearing sperm with Spam1 overexpression were detected by fluorescence in situ hybridization. In wild-type animals, EM studies of in situ transcript hybridization of testis sections and Northern analysis of biochemically fractionated testicular RNA were performed to localize Spam1 transcript. Finally, AU-rich motifs identified in the 3' UTR of Spam1 RNA were assayed by UV cross-linking to determine their ability to interact with testicular RNA binding proteins. RESULTS: The Tg8 line of transgene carriers had a significant (P < 0.001) TRD, due to reduced fertilizing ability of transgene-bearing sperm. These sperm retained large cytoplasmic droplets engorged with overexpressed Spam1 or Hyal5 protein. Caudal sperm from transgene carriers and caput sperm of null carriers showed a bimodal distribution of Spam1, indicating that the sperm in a male were biochemically different with respect to Spam1 quantities. Spam1 RNA was absent from the bridges, associated exclusively with the ER, and was shown to be anchored to the cytoskeleton. This compartmentalization of the transcript, mediated by cytoskeletal binding, occurs via protein interactions with 3' UTR AU-rich sequences that are likely involved in its stabilization. CONCLUSION: We provide strong support for the LOS hypothesis, and have elucidated the mechanism of Spam1-associated TRD
