    Penduduk Dusun Krajan tergolong sebagai masyarakat Tengger. Mereka memilikiaturan tersendiri mengenai tata letak hunian antar kerabat sebagai perwujudanadat istiadat dan budaya masyarakatnya. Selain kentalnya kebudayaan, dusuntersebut juga memiliki keindahan alam yang berdampak pada bertambahnyajumlah pendatang, yang diindikasikan akan membawa kebudayaan dan tradisiberbeda. Tujuan dari penelitian ini adalah untuk mengidentifikasi dan menganalisispengaruh sistem kekerabatan terhadap pola permukiman Dusun Krajan KabupatenLumajang. Metode identifikasi yang digunakan adalah deskriptif-kualitatif, denganmetode analisis family tree. Hasil menunjukan bahwa sistem kekerabatanmemberikan pengaruh terhadap transparasi antar bangunan hunian, orientasibangunan terhadap Gunung Bromo dan Pesanggrahan, hirarki dan tata letakhunian kakak tertua dalam kelompok bangunan pada permukiman Dusun Kraja

    Long non-coding RNAs as local regulators of pancreatic islet transcription factor genes

    The transcriptional programs of differentiated cells are tightly regulated by interactions between cell type-specific transcription factors and cis-regulatory elements. Long non-coding RNAs (lncRNAs) have emerged as additional regulators of gene transcription. Current evidence indicates that lncRNAs are a very heterogeneous group of molecules. For example, selected lncRNAs have been shown to regulate gene expression in cis or trans, although in most cases the precise underlying molecular mechanisms is unknown. Recent studies have uncovered a large number of lncRNAs that are selectively expressed in pancreatic islet cells, some of which were shown to regulate β cell transcriptional programs. A subset of such islet lncRNAs appears to control the expression of β cell-specific transcription factor (TF) genes by local cis-regulation. In this review, we discuss current knowledge of molecular mechanisms underlying cis-regulatory lncRNAs and discuss challenges involved in using genetic perturbations to define their function. We then discuss known examples of pancreatic islet lncRNAs that appear to exert cis-regulation of TF genes. We propose that cis-regulatory lncRNAs could represent a molecular target for modulation of diabetes-relevant genes

    Variaciones hematológicas y de presión sanguínea en perros después de una picadura de abejas

    RESUMEN Objetivo. El objetivo de este estudio fue determinar los cambios hematológicos y de presión arterial en perros después de una picadura cutánea de abejas. Material y métodos. Se utilizaron 10 perros adultos, a los cuales se les inoculó veneno de abeja mediante una picadura cutánea controlada. De cada animal se extrajo una muestra de sangre previa y los 15 y 300 minutos post inoculación. Se registró la presión arterial sistólica (PAS), diastólica (PAD), media (PAM) y de pulso (PP) desde 15 minutos previos hasta 45 minutos posteriores a la inoculación. Resultados. El recuento de eritrocitos, el hematocrito y la concentración de hemoglobina se redujeron a los 15 minutos post-inoculación (p0.05). Conclusiones. La picadura cutánea de abejas produce un descenso transitorio del recuento de eritrocitos, hematocrito y concentración de hemoglobina, junto con una disminución en el recuento de reticulocitos y del recuento relativo de eosinófilos, sin alterar mayormente la presión sanguínea

    Nkx2.2 and Arx genetically interact to regulate pancreatic endocrine cell development and endocrine hormone expression

    AbstractNkx2.2 and Arx are essential pancreatic transcription factors. Nkx2.2 is necessary for the appropriate specification of the islet alpha, beta, PP and epsilon cell lineages, whereas Arx is required to form the correct ratio of alpha, beta, delta and PP cells. To begin to understand the cooperative functions of Nkx2.2 and Arx in the development of endocrine cell lineages, we generated progenitor cell-specific deletions of Arx on the Nkx2.2 null background. The analysis of these mutants demonstrates that expansion of the ghrelin cell population in the Nkx2.2 null pancreas is not dependent on Arx; however, Arx is necessary for the upregulation of ghrelin mRNA levels in Nkx2.2 mutant epsilon cells. Alternatively, in the absence of Arx, delta cell numbers are increased and Nkx2.2 becomes essential for the repression of somatostatin gene expression. Interestingly, the dysregulation of ghrelin and somatostatin expression in the Nkx2.2/Arx compound mutant (Nkx2.2null;ArxΔpanc) results in the appearance of ghrelin+/somatostatin+ co-expressing cells. These compound mutants also revealed a genetic interaction between Nkx2.2 and Arx in the regulation of the PP cell lineage; the PP cell population is reduced when Nkx2.2 is deleted but is restored back to wildtype numbers in the Nkx2.2null;ArxΔpanc mutant. Moreover, conditional deletion of Arx in specific pancreatic cell populations established that the functions of Arx are necessary in the Neurog3+ endocrine progenitors. Together, these experiments identify novel genetic interactions between Nkx2.2 and Arx within the endocrine progenitor cells that ensure the correct specification and regulation of endocrine hormone-producing cells

    A review of the methods for the most common parameters determined in wine—namely, ethanol, sulfur dioxide, reducing sugars, polyphenols, organic acids, total and volatile acidity, iron, soluble solids, pH, and color—reported in the last 10 years is presented here. The definition of the given parameter, official and usual methods in wineries appear at the beginning of each section, followed by the methods reported in the last decade divided into discontinuous and continuous methods, the latter also are grouped in nonchromatographic and chromatographic methods because of the typical characteristics of each subgroup. A critical comparison between continuous and discontinuous methods for the given parameter ends each section. Tables summarizing the features of the methods and a conclusions section may help users to select the most appropriate method and also to know the state-of-the-art of analytical methods in this area

    Low BMI-1 expression is associated with an activated BMI-1-driven signature, vascular invasion, and hormone receptor loss in endometrial carcinoma

    We studied the expression of polycomb group (PcG) protein BMI-1 in a large population-based patient series of endometrial carcinomas in relation to clinical and molecular phenotype. Also, 57 fresh frozen endometrial carcinomas were studied for the relationship between BMI-1 protein expression, BMI-1 mRNA level, and activation of an 11-gene signature reported to represent a BMI-1-driven pathway. BMI-1 protein expression was significantly weaker in tumours with vascular invasion (P<0.0001), deep myometrial infiltration (P=0.004), and loss of oestrogen receptor (ER) (P<0.0001) and progesterone receptors (PR) (P=0.03). Low BMI-1 protein expression was highly associated with low BMI-1 mRNA expression (P=0.002), and similarly low BMI-1 mRNA expression correlated significantly with vascular invasion, ER and PR loss, and histologic grade 3. In contrast, activation of the reported 11-gene signature, supposed to represent a BMI-1-driven pathway, correlated with low mRNA expression of BMI-1 (P<0.001), hormone receptor loss, presence of vascular invasion, and poor prognosis. We conclude that BMI-1 protein and mRNA expression are significantly correlated and that BMI-1 expression is inversely associated with activation of the 11-gene signature. Loss of BMI-1 seems to be associated with an aggressive phenotype in endometrial carcinomas

    P-cadherin expression in breast cancer: a review

    P-cadherin is frequently over-expressed in high-grade invasive breast carcinomas and has been reported to be an enhancer of migration and invasion of breast cancer cells, being correlated with tumour aggressiveness. In addition, expression of P-cadherin is well established as an indicator of poor prognosis in human breast cancer, which has stimulated our interest in studying its role in this setting. This review describes the most important findings on P-cadherin expression and function in normal mammary tissue and breast cancer cells, emphasizing that further research is required to elucidate the role played by this protein in human mammary tumours
