    Liquefaction source layer for sand blows induced by the 2016 megathrust earthquake (Mw 7.8) in Ecuador (Boca de Briceño)

    Numerous sand boils were generated in the alluvial plain at the mouth of the Rio Brice˜no valley (Ecuador) during the Mw 7.8 earthquake of April 2016. The area is characterized by a series of raised marine terraces formed as a consequence of the rapid tectonic coastal uplift during the Quaternary. Boreholes and geotechnical investigations were carried during post-earthquake surveys and for the purpose of mitigating the liquefaction effects. Five lithological units were identified at a site of embankment, which represented continental-marine and transitional sedimentation since the Last Glacial Maximum. A comprehensive study of texture and petrographic composition of sand boils has been performed and compared with sandy silts and silty sands of the buried sedimentary sequence in order to identify the source levels for liquefaction. The petrographic components, in particular the low content of bioclasts and carbonate fragments of the sand boils, allow to pinpoint a source layer made up of fine-grained silty sands located between 2 and 4.5 m depth (Unit 2) whereas the deeper marine sands, richer in bioclasts, were not involved. The results support the idea that earthquake-induced liquefaction phenomena are not restricted to clean sands and well-sorted deposits, but may affect sand layers with significant amount of nonplastic silt

    Turning a green alga red: engineering astaxanthin biosynthesis by intragenic pseudogene revival in Chlamydomonas reinhardtii.

    SummaryThe green alga Chlamydomonas reinhardtii does not synthesize high-value ketocarotenoids like canthaxanthin and astaxanthin, however, a β-carotene ketolase (CrBKT) can be found in its genome. CrBKT is poorly expressed, contains a long C-terminal extension not found in homologues and likely represents a pseudogene in this alga. Here, we used synthetic re-design of this gene to enable its constitutive overexpression from the nuclear genome of C. reinhardtii. Overexpression of the optimized CrBKT extended native carotenoid biosynthesis to generate ketocarotenoids in the algal host causing noticeable changes the green algal colour to a reddish-brown. We found that up to 50% of native carotenoids could be converted into astaxanthin and more than 70% into other ketocarotenoids by robust CrBKT overexpression. Modification of the carotenoid metabolism did not impair growth or biomass productivity of C. reinhardtii, even at high light intensities. Under different growth conditions, the best performing CrBKT overexpression strain was found to reach ketocarotenoid productivities up to 4.5 mg L-1 day-1. Astaxanthin productivity in engineered C. reinhardtii shown here is competitive with that reported for Haematococcus lacustris (formerly pluvialis) which is currently the main organism cultivated for industrial astaxanthin production. In addition, the extractability and bio-accessibility of these pigments was much higher in cell wall deficient C. reinhardtii than the resting cysts of H. lacustris. Engineered C. reinhardtii strains could thus be a promising alternative to natural astaxanthin producing algal strains and may open the possibility of other tailor-made pigments from this host

    Obsidian from the Site of Piano dei Cardoni, Ustica (Palermo, Italy): Preliminary Results on the First Occupation of the Island

    Abstract New investigations on Ustica (Palermo, Sicily) originated from the need to improve our knowledge of the island's archaeological and environmental heritage. Through field surveys, particular attention was paid to human occupation in the Neolithic phases and focused on the less investigated southern side of the island. The systematic survey of the area of Piano dei Cardoni in 2018 brought to light a new Middle/Late Neolithic site, already partially documented in the literature. The island was settled for the first time during these phases, as also testified from the area of Punta Spalmatore. The presence of Serra d'Alto, incised dark burnished, and Diana styles suggests that the site and the archaeological assemblage dates from the mid to late 5th millennium BC, as confirmed by AMS dating. In addition to pottery, obsidian artifacts were also recovered, and a preliminary study of these materials is presented here. Portable XRF analyses on a sample of 41 obsidian artifacts, representing a high percentage of the lithic assemblage compared to chert tools, show that the provenance of the raw material is Gabellotto Gorge (Lipari) and Balata dei Turchi (Pantelleria). These results provide new insight into broader regional debates about obsidian technology and its exchange during the Neolithic and open an important consideration for sites that are far from the raw material sources

    Mussels as a dietary source of omega-3 fatty acids

    Numerous United Kingdom and European Union expert panels recommend that the general adult population consumes ~250mg of EPA+DHA per day through the consumption of 1 portion of oily fish per week. Of particular importance are the long chain omega-3 fatty acids EPA (eicosapentaenoic acid) and DHA (docosahexaenoic acid), which are only found in appreciable amounts in marine organisms. Increasing oily fish consumption conflicts with sustaining fisheries and so alternative dietary sources of EPA+DHA must be explored. Mussels are high in omega-3 PUFAs and are a good source of essential amino acids. Therefore, the present study aimed to investigate the impact of introducing mussels as a protein source in the lunchtime meal 3 times per week for 2 weeks on omega-3 status in free-living participants. Following an initial 2-week monitoring period, 12 participants (8 male, 4 female) attended the nutrition laboratory 3 times per week for two weeks. Each participant received a personalised lunch constituting one-third of their typical daily calorie consumption with ~20% of the calories supplied as cooked mussels. A portion of cooked mussels from each feeding occasion was tested for total omega-3 content. The mean ± SD mussel EPA+DHA content was 518.9 ± 155.7mg/100g cooked weight meaning that each participant received on average 709.2 ± 252.6mg of EPA+DHA per meal or 304.0 ± 108.2mg of EPA+DHA per day. Blood spot analysis revealed a significant increase in the omega-3 index (week 1 = 4.27 ± 0.81; week 4 = 5.07 ± 1.00) and whole blood EPA content during the study (%EPA week 1 = 0.70 ± 0.0.35; %EPA week4 = 0.98 ± 0.35). Consuming mussels 3 times per week for two weeks as the protein source in a personalised lunchtime meal is sufficient to moderately improve the omega-3 index and whole blood DHA+EPA content in young healthy adults

    Functionalised peptide hydrogel for the delivery of cardiac progenitor cells.

    Heart failure (HF) remains one of the leading causes of death worldwide; most commonly developing after myocardial infarction (MI). Since adult cardiomyocytes characteristically do not proliferate, cells lost during MI are not replaced. As a result, the heart has a limited regenerative capacity. There is, therefore, a need to develop novel cell-based therapies to promote the regeneration of the heart after MI. The delivery and retention of cells at the injury site remains a significant challenge. In this context, we explored the potential of using an injectable, RGDSP-functionalised self-assembling peptide - FEFEFKFK - hydrogel as scaffold for the delivery and retention of rat cardiac progenitor cells (CPCs) into the heart. Our results show that culturing CPCs in vitro within the hydrogel for one-week promoted their spontaneous differentiation towards adult cardiac phenotypes. Injection of the hydrogel on its own, or loaded with CPCs, into the rat after injury resulted in a significant reduction in myocardial damage and left ventricular dilation

    Nuclear organisation and replication timing are coupled through RIF1-PP1 interaction

    Three-dimensional genome organisation and replication timing are known to be correlated, however, it remains unknown whether nuclear architecture overall plays an instructive role in the replication-timing programme and, if so, how. Here we demonstrate that RIF1 is a molecular hub that co-regulates both processes. Both nuclear organisation and replication timing depend upon the interaction between RIF1 and PP1. However, whereas nuclear architecture requires the full complement of RIF1 and its interaction with PP1, replication timing is not sensitive to RIF1 dosage. The role of RIF1 in replication timing also extends beyond its interaction with PP1. Availing of this separation-of-function approach, we have therefore identified in RIF1 dual function the molecular bases of the co-dependency of the replication-timing programme and nuclear architecture

    Type Ia Supernova Rate Measurements To Redshift 2.5 From CANDELS: Searching For Prompt Explosions In The Early Universe

    dThe Cosmic Assembly Near-infrared Deep Extragalactic Legacy Survey (CANDELS) was a multi-cycle treasury program on the Hubble Space Telescope (HST) that surveyed a total area of -0.25 deg2 with -900 HST orbits spread across five fields over three years. Within these survey images we discovered 65 supernovae (SNe) of all types, out to z 2.5. We classify -24 of these as Type Ia SNe (SNe Ia) based on host galaxy redshifts and SN photometry (supplemented by grism spectroscopy of six SNe). Here we present a measurement of the volumetric SN Ia rate as a function of redshift, reaching for the first time beyond z =- 2 and putting new constraints on SN Ia progenitor models. Our highest redshift bin includes detections of SNe that exploded when the universe was only -3 Gyr old and near the peak of the cosmic star formation history. This gives the CANDELS high redshift sample unique leverage for evaluating the fraction of SNe Ia that explode promptly after formation ( 40 Myr. However, mild tension is apparent between ground-based low-z surveys and space-based high-z surveys. In both CANDELS and the sister HST program CLASH (Cluster Lensing And Supernova Survey with Hubble), we find a low rate of SNe Ia at z > 1. This could be a hint that prompt progenitors are in fact relatively rare, accounting for only 20% of all SN Ia explosions-though further analysis and larger samples will be needed to examine that suggestion.     Results from the centers for disease control and prevention's predict the 2013-2014 Influenza Season Challenge

    Background: Early insights into the timing of the start, peak, and intensity of the influenza season could be useful in planning influenza prevention and control activities. To encourage development and innovation in influenza forecasting, the Centers for Disease Control and Prevention (CDC) organized a challenge to predict the 2013-14 Unites States influenza season. Methods: Challenge contestants were asked to forecast the start, peak, and intensity of the 2013-2014 influenza season at the national level and at any or all Health and Human Services (HHS) region level(s). The challenge ran from December 1, 2013-March 27, 2014; contestants were required to submit 9 biweekly forecasts at the national level to be eligible. The selection of the winner was based on expert evaluation of the methodology used to make the prediction and the accuracy of the prediction as judged against the U.S. Outpatient Influenza-like Illness Surveillance Network (ILINet). Results: Nine teams submitted 13 forecasts for all required milestones. The first forecast was due on December 2, 2013; 3/13 forecasts received correctly predicted the start of the influenza season within one week, 1/13 predicted the peak within 1 week, 3/13 predicted the peak ILINet percentage within 1 %, and 4/13 predicted the season duration within 1 week. For the prediction due on December 19, 2013, the number of forecasts that correctly forecasted the peak week increased to 2/13, the peak percentage to 6/13, and the duration of the season to 6/13. As the season progressed, the forecasts became more stable and were closer to the season milestones. Conclusion: Forecasting has become technically feasible, but further efforts are needed to improve forecast accuracy so that policy makers can reliably use these predictions. CDC and challenge contestants plan to build upon the methods developed during this contest to improve the accuracy of influenza forecasts. © 2016 The Author(s)
