    This report seeks to assess the environmental impact of existing and potential fiscal instruments in Ireland. This is achieved by first conducting a simple assessment of potential environmental impacts of a large number of existing and potential fiscal instruments. This considers the incentives that a particular measure sets, the likely resulting behaviour and the consequent expected environmental impact. In total, 142 measures are considered. The environmental impacts considered cover the main domains of climate change, air quality, water quality and land; 246 impacts are identified, which implies that on average measures impact on more than one domain. The most widespread impact is on climate change emissions, with 98 measures having impacts. The least common impact is on water, with just 23 measures. Just over half the measures were assessed to have a likely positive impact

    THE EFFECT OF CARBON-PLATED RUNNING SHOES ON PERFORMANCE

    Differences in running economy (RE) in shoes with and without a carbon fiber plate were assessed. Nine participants who had previously run under 18-minutes in the 5km completed the study. Three shoe conditions were assessed at intensities slightly above and below the ventilatory threshold (VT), totaling six 5-minute RE bouts. In two otherwise identical Skechers running shoes, RE was unchanged with a carbon fiber plate running at intensities below (p = 0.355) and above VT (p = 0.715). The Nike Vaporfly 4% improved RE with respect to the Skechers Speed Elite carbon fiber shoe both below (p = 0.005) and above VT (p = 0.019). Changes in RE did not differ at intensities below and above VT (p = 0.893). The current interim findings suggest that a carbon fiber plate alone may not improve RE, but there may be a combined effect of midsole cushioning and a carbon fiber plate.Master of Art

    Inflammatory Leiomyosarcoma and Histiocyte-rich Rhabdomyoblastic Tumor : a clinicopathological, immunohistochemical and genetic study of 13 cases, with a proposal for reclassification as Inflammatory Rhabdomyoblastic Tumor

    Inflammatory leiomyosarcoma (ILMS), defined as a malignant neoplasm showing smooth muscle differentiation, a prominent inflammatory infiltrate, and near-haploidization , is a very rare soft tissue tumor with a generally favorable prognosis. The morphologic features of histiocyte-rich rhabdomyoblastic tumor (HRRMT) are similar to those of ILMS, although this lesion shows by definition a skeletal muscle phenotype. Recent gene expression profiling and immunohistochemical studies have also suggested that ILMS and HRRMT may be related. We studied the clinicopathologic, immunohistochemical and genetic features of four cases previously classified as ILMS and nine classified as HRRMT. Tumors from both groups tended to occur in the deep soft tissues of the extremities of young to middle-aged males and exhibited indolent behavior. Morphologically, all were well-circumscribed, often encapsulated, and showed a striking histiocyte-rich inflammatory infiltrate admixed with variably pleomorphic tumor cells showing spindled and epithelioid to rhabdoid morphology, eosinophilic cytoplasm, and prominent nucleoli, but few, if any, mitotic figures. Immunohistochemically, the tumor cells expressed desmin, alpha-smooth muscle actin, and the rhabdomyoblastic markers PAX7, MyoD1, and myogenin. H-caldesmon expression was absent in all cases, using the specific h-CD antibody. Karyotypic study (1 HRRMT) and genome-wide copy number analysis (7 HRRMT, OncoScan SNP assay), revealed near-haploidization in four cases, with subsequent genome doubling in one, an identical phenotype to that seen in ILMS. We propose reclassification of ILMS and HRRMT as inflammatory rhabdomyoblastic tumor , a name which accurately describes the salient morphologic and immunohistochemical features of this distinctive tumor, as well as its intermediate (rarely metastasizing) clinical behavior

    ORM Expression Alters Sphingolipid Homeostasis and Differentially Affects Ceramide Synthase Activity

    Sphingolipid synthesis is tightly regulated in eukaryotes. This regulation in plants ensures sufficient sphingolipids to support growth while limiting the accumulation of sphingolipid metabolites that induce programmed cell death. Serine palmitoyltransferase (SPT) catalyzes the first step in sphingolipid biosynthesis and is considered the primary sphingolipid homeostatic regulatory point. In this report, Arabidopsis (Arabidopsis thaliana) putative SPT regulatory proteins, orosomucoidlike proteins AtORM1 and AtORM2, were found to interact physically with Arabidopsis SPT and to suppress SPT activity when coexpressed with Arabidopsis SPT subunits long-chain base1 (LCB1) and LCB2 and the small subunit of SPT in a yeast (Saccharomyces cerevisiae) SPT-deficient mutant. Consistent with a role in SPT suppression, AtORM1 and AtORM2 overexpression lines displayed increased resistance to the programmed cell death-inducing mycotoxin fumonisin B1, with an accompanying reduced accumulation of LCBs and C16 fatty acid-containing ceramides relative to wild-type plants. Conversely, RNA interference (RNAi) suppression lines of AtORM1 and AtORM2 displayed increased sensitivity to fumonisin B1 and an accompanying strong increase in LCBs and C16 fatty acid-containing ceramides relative to wild-type plants. Overexpression lines also were found to have reduced activity of the class I ceramide synthase that uses C16 fatty acid acyl-coenzyme A and dihydroxy LCB substrates but increased activity of class II ceramide synthases that use very-long-chain fatty acyl-coenzyme A and trihydroxy LCB substrates. RNAi suppression lines, in contrast, displayed increased class I ceramide synthase activity but reduced class II ceramide synthase activity. These findings indicate that ORM mediation of SPT activity differentially regulates functionally distinct ceramide synthase activities as part of a broader sphingolipid homeostatic regulatory network

    A Framework for Automated Measurement of Responsible AI Harms in Generative AI Applications

    We present a framework for the automated measurement of responsible AI (RAI) metrics for large language models (LLMs) and associated products and services. Our framework for automatically measuring harms from LLMs builds on existing technical and sociotechnical expertise and leverages the capabilities of state-of-the-art LLMs, such as GPT-4. We use this framework to run through several case studies investigating how different LLMs may violate a range of RAI-related principles. The framework may be employed alongside domain-specific sociotechnical expertise to create measurements for new harm areas in the future.     An objective measure for the assessment and management of fluid shifts in acute major burns

    Background: Major burns are life threatening. Fluid resuscitation is required for survival to maintain intravascular volumes and prevent hypovolemic shock. Bioimpedance spectroscopy (BIS) has been recognised as a potential method of monitoring fluid shifts after burn and in other disease states. The aims of this study were to examine the reliability of BIS across different dressing conditions and electrode positions, establish the influence of Acticoat™ on BIS variable measures and determine the validity of whole-body BIS to assess net fluid shift in the presence of moderate to major burns. Methods: An observational longitudinal cohort study was conducted from December 2014 to February 2016. Patients with over 15% total body surface area (TBSA) burns and injury less than 48 h were enrolled in the study. BIS triplicate measures were collected in an open wound and with an ActicoatTM dressing (at 5 half hour intervals). Standard and alternate electrode placements were utilised for the reliability analysis and standard placement only for determining the validity of BIS in moderate to major burns. The ImpediMde SFB7 was used to collect wholebody and segmental BIS measures. Stata statistical software, release 14 was utilised to analyse all results. Descriptive analyses were performed and were reported using the means and standard deviations (SD). Results: BIS-repeated measures established BIS raw resistance (R), and predicted volume variables were reliable in any condition (intra-class correlation coefficient (ICC) 0.996–0.999, 95% confidence intervals (CI) 0.996–0.999) without a systematic difference. Acticoat™ dressings significantly influenced all BIS-predicted volumes (p ≤ 0.01) as determined by multilevel mixed effects (MLME) linear regression analysis. Validity of BIS was demonstrated by resistance variables significantly decreasing with increasing net ionic fluid shift and increased TBSA (severity of injury) and calculated fluid volumes increasing with increasing net fluid shift and TBSA. BIS resistance also decreased with time as oedema reduced. For clinical use, a calculator was developed to adjust BIS variables when an Acticoat™ dressing is in situ, thus facilitating BIS variable change estimates in real time, with dressings intact. Conclusion: BIS may be used clinically to monitor fluid volume change in major acute burns

    Oncogenic ERBB3 Mutations in Human Cancers

    SummaryThe human epidermal growth factor receptor (HER) family of tyrosine kinases is deregulated in multiple cancers either through amplification, overexpression, or mutation. ERBB3/HER3, the only member with an impaired kinase domain, although amplified or overexpressed in some cancers, has not been reported to carry oncogenic mutations. Here, we report the identification of ERBB3 somatic mutations in ∼11% of colon and gastric cancers. We found that the ERBB3 mutants transformed colonic and breast epithelial cells in a ligand-independent manner. However, the mutant ERBB3 oncogenic activity was dependent on kinase-active ERBB2. Furthermore, we found that anti-ERBB antibodies and small molecule inhibitors effectively blocked mutant ERBB3-mediated oncogenic signaling and disease progression in vivo
