    Mini-Collagens in Hydra Nematocytes

    We have isolated and characterized four collagen-related c-DNA clones (N-COL 1, N-COL 2, N-COL 3, N-COL 4) that are highly expressed in developing nematocytes in hydra. All four c-DNAs as well as their corresponding transcripts are small in size (600-1,000 bp). The deduced amino acid sequences show that they contain a central region consisting of 14 to 16 Gly-X-Y triplets. This region is flanked amino-terminal by a stretch of 14-23 proline residues and carboxy-terminal by a stretch of 6-9 prolines. At the NH2- and COOH-termini are repeated patterns of cysteine residues that are highly conserved between the molecules. A model is proposed which consists of a central stable collagen triple helix of 12-14 nm length from which three 9-22 nm long polyproline II type helices emerge at both ends. Disulfide linkage between cysteine- rich segments in these helices could lead to the formation of oligomeric network structures.     In der oberen Kruste sind Störungszonen üblicherweise aus komplexen Netzwerken von diskreten Brüchen unterschiedlicher Genese zusammengesetzt. Sie stellen daher meist maßgebliche Zonen bevorzugter unterirdischer Wasserwegigkeiten dar. Außerdem sind sie somit wesentlich für die hydrogeologische Beurteilung von Gebirgskörpern, da massive Wassereinbrüche im Untertagebau sehr häufig an Störungszonen gebunden sind. Solche Störungszonen zeigen für gewöhnlich eine räumlich-zeitliche Entwicklung vom Ausgangsgestein zu vollständig ausgeprägten Störungszonen mit einer internen strukturellen Gliederung (Protolith — damage zone — fault core). Die hydraulische Bewertung von Störungen ist ein wesentlicher Eingangsparameter für eine numerisch hydraulische Modellierung von Gebirgskörpern. Hierfür ist es notwendig das hydraulische Verhalten und die Wirksamkeit der verschiedenen Bereiche der Störung zu beschreiben und für die modellhafte Umsetzung mit hydraulischen Kenngrößen zu belegen. Das Ziel der Arbeit ist eine Korrelation von strukturgeologischen Parametern mit hydraulischen Parametern (kf , T, . . . ). Dies kann als eine hydraulische Kalibration der einzelnen Störungsklassen in Verbindung mit Wasserdurchlässigkeit angesehen werden.     Evaluation of the double-tracer gas single-breath washout test in a pediatric field study.

    BACKGROUND The early-life origins of chronic pulmonary diseases are thought to arise in peripheral small airways. Predictors of ventilation inhomogeneity, a proxy of peripheral airway function, are understudied in schoolchildren. RESEARCH QUESTION Is the double-tracer gas single-breath washout (DTG-SBW) measurement feasible in a pediatric field study setting? What are the predictors of the DTG-SBW derived ventilation inhomogeneity estimate in unselected schoolchildren? STUDY DESIGN AND METHODS In this prospective cross-sectional field study, a mobile lung function-testing unit visited participating schools in Switzerland. We applied DTG-SBW, fraction of exhaled nitric oxide (FeNO), and spirometry measurements. The DTG-SBW is based on tidal inhalation of helium (He) and sulfur-hexafluoride (SF6) and the phase III slope (SIIIHe-SF6) is derived. We assessed feasibility, repeatability, and associations of SIIIHe-SF6 with the potential predictors anthropometrics, presence of wheeze (i.e. parental report of ≥ 1 episode of wheeze in the prior year), FeNO, forced expiratory volume in the first second (FEV1), and FEV1/forced vital capacity (FVC). RESULTS In 1782 children, 5223 DTG-SBW trials were obtained. The DTG-SBW was acceptable in 1449 (81.3%) children, coefficient of variation was 39.8%. SIIIHe-SF6 was independently but weakly positively associated with age and BMI. In 276 (21.2%) children, wheeze was reported. SIIIHe-SF6 was higher by 0.049 g.mol.L-1 in children with wheeze as compared to those without and remained associated with wheeze after adjusting for age and BMI in a multi-variable linear regression model. SIIIHe-SF6 was not associated with FeNO, FEV1, and FEV1/FVC. INTERPRETATION The DTG-SBW is feasible in a pediatric field study setting. On the population level, age, body composition and wheeze are independent predictors of peripheral airway function in unselected schoolchildren. The variation of the DTG-SBW possibly constrains its current applicability on the individual level

    LuftiBus in the school (LUIS): a population-based study on respiratory health in schoolchildren.

    Respiratory disease is common in children and strongly associated with lifestyle and environmental exposures. Thus, it is important to study the epidemiology locally. The LuftiBus in the School (LUIS) study was set up to assess the respiratory health of schoolchildren in the canton of Zurich, Switzerland. LUIS is a cross-sectional population-based study that was carried out 2013 to 2016. Children aged 6–17 years living in the canton of Zurich were eligible to participate. All schools in the canton were approached and the school head decided whether the school would participate and with which classes. Consenting parents answered a standardised questionnaire at home and assenting children completed a shorter questionnaire by interview at school. Trained technicians measured children’s lung function, including spirometry, double tracer gas single-breath washout (DTG-SBW) and fractional exhaled nitric oxide (FeNO). Address histories of participants were geocoded to be linked with area-based socioeconomic measures and environmental exposures such as spatiotemporal air pollution estimates for specific time periods and locations. A subgroup was seen again 12 months later using the same procedures to collect longitudinal data. The study included 3870 children at baseline and 655 at the 1-year follow-up. Median age was 12.7 years; 281 (8%) had wheezed in the past year. At baseline we collected 3457 (89%) parental and 3546 (92%) child questionnaires, and 3393 (88%) FeNO, 3446 (89%) spirometry, and 1795 (46%) DTG-SBW measurements. LUIS is a rich resource of health-related data, with information on lung function, environmental exposures and respiratory health on Swiss schoolchildren

    Application of the multi-level time-harmonic fast multipole BEM to 3-D visco-elastodynamics

    Engineering Analysis with Boundary elements (accepted, to appear)International audienceThis article extends previous work by the authors on the single- and multi-domain time-harmonic elastodynamic multi-level fast multipole BEM formulations to the case of weakly dissipative viscoelastic media. The underlying boundary integral equation and fast multipole formulations are formally identical to that of elastodynamics, except that the wavenumbers are complex-valued due to attenuation. Attention is focused on evaluating the multipole decomposition of the viscoelastodynamic fundamental solution. A damping-dependent modification of the selection rule for the multipole truncation parameter, required by the presence of complex wavenumbers, is proposed. It is empirically adjusted so as to maintain a constant accuracy over the damping range of interest in the approximation of the fundamental solution, and validated on numerical tests focusing on the evaluation of the latter. The proposed modification is then assessed on 3D single-region and multi-region visco-elastodynamic examples for which exact solutions are known. Finally, the multi-region formulation is applied to the problem of a wave propagating in a semi-infinite medium with a lossy semi-spherical inclusion (seismic wave in alluvial basin). These examples involve problem sizes of up to about 3 1053\,10^{5} boundary unknowns

    Molecular Cytogenetic Profiling Reveals Similarities and Differences Between Localized Nodal and Systemic Follicular Lymphomas

    Recently, we have developed novel highly promising gene expression (GE) classifiers discriminating localized nodal (LFL) from systemic follicular lymphoma (SFL) with prognostic impact. However, few data are available in LFL especially concerning hotspot genetic alterations that are associated with the pathogenesis and prognosis of SFL. A total of 144 LFL and 527 SFL, enrolled in prospective clinical trials of the German Low Grade Lymphoma Study Group, were analyzed by fluorescence in situ hybridization to detect deletions in chromosomes 1p, 6q, and 17p as well as BCL2 translocations to determine their impact on clinical outcome of LFL patients. The frequency of chromosomal deletions in 1p and 17p was comparable between LFL and SFL, while 6q deletions and BCL2 translocations more frequently occurred in SFL. A higher proportion of 1p deletions was seen in BCL2-translocation–positive LFL, compared with BCL2-translocation–negative LFL. Deletions in chromosomes 1p, 6q, and 17p predicted clinical outcome of patients with SFL in the entire cohort, while only deletions in chromosome 1p retained its negative prognostic impact in R-CHOP–treated SFL. In contrast, no deletions in one of the investigated genetic loci predicted clinical outcome in LFL. Likewise, the presence or absence of BCL2 translocations had no prognostic impact in LFL. Despite representing a genetic portfolio closely resembling SFL, LFL showed some differences in deletion frequencies. BCL2 translocation and 6q deletion frequency differs between LFL and SFL and might contribute to distinct genetic profiles in LFL and SFL

    Status of the BELLE II Pixel Detector

    The Belle II experiment at the super KEK B-factory (SuperKEKB) in Tsukuba, Japan, has been collecting e+e−e^+e^− collision data since March 2019. Operating at a record-breaking luminosity of up to 4.7×1034cm−2s−14.7×10^{34} cm^{−2}s^{−1}, data corresponding to 424fb−1424 fb^{−1} has since been recorded. The Belle II VerteX Detector (VXD) is central to the Belle II detector and its physics program and plays a crucial role in reconstructing precise primary and decay vertices. It consists of the outer 4-layer Silicon Vertex Detector (SVD) using double sided silicon strips and the inner two-layer PiXel Detector (PXD) based on the Depleted P-channel Field Effect Transistor (DePFET) technology. The PXD DePFET structure combines signal generation and amplification within pixels with a minimum pitch of (50×55)μm2(50×55) μm^2. A high gain and a high signal-to-noise ratio allow thinning the pixels to 75μm75 μm while retaining a high pixel hit efficiency of about 9999%. As a consequence, also the material budget of the full detector is kept low at ≈0.21≈0.21%XX0\frac{X}{X_0} per layer in the acceptance region. This also includes contributions from the control, Analog-to-Digital Converter (ADC), and data processing Application Specific Integrated Circuits (ASICs) as well as from cooling and support structures. This article will present the experience gained from four years of operating PXD; the first full scale detector employing the DePFET technology in High Energy Physics. Overall, the PXD has met the expectations. Operating in the intense SuperKEKB environment poses many challenges that will also be discussed. The current PXD system remains incomplete with only 20 out of 40 modules having been installed. A full replacement has been constructed and is currently in its final testing stage before it will be installed into Belle II during the ongoing long shutdown that will last throughout 2023
