    Insights into high temperature pretreatment on cellulase processing of bamboo

    Bamboo processing was performed with commercial cellulase. The properties of cellulase and the effect of high temperature pretreatment on cellulase hydrolysis of bamboo were investigated. Results indicated that cellulase hydrolysis performed fast and dramatically within 30 minutes, and then gradually reached its balance. It was found that pretreatment played an active role in cellulase processing, which enhanced the saccharification of bamboo and benefited high-molecular-weight lignin degradation and removal. Additionally, a better performance of bamboo processing was achieved under the cellulase concentration of 15IU in total reaction system of 100 ml at 50°C, pH 4.8, together with the high temperature pretreatment of 120°C for 15 minutes

    Inhibitory effects of adenovirus mediated tandem expression of RhoA and RhoC shRNAs in HCT116 cells

    <p>Abstract</p> <p>Background</p> <p>RhoA and RhoC are deregulated by over expression in many human tumors, including colorectal cancer. Some reports show that they play a pivotal role in the carcinogenesis, tumor development and infiltration metastasis. In this study, for the first time we constructed recombinant adenovirus to investigate the inhibitory effects of RhoA and RhoC shRNAs in tandem expression on the cell proliferation and invasion of colorectal cancer HCT116 cells.</p> <p>Methods</p> <p>The recombinant adenovirus carrying RhoA and RhoC shRNAs in tandem expression was transfected into HCT116. The mRNA transcription and protein expressions of RhoA and RhoC were examined by RT-FQPCR and Western blot respectively. Cellular proliferation inhibitory activity was determined by methyl thiazolyl tetrazolium (MTT) assay and invasive and migrating potential was detected through in vitro Matrigel coated invasion and migration assay.</p> <p>Results</p> <p>Both mRNA and proteins Levels of RhoA and RhoC were significantly reduced in HCT116 cells transfected with Ad-A1+A2+C1+C2 than those in Ad-HK group and control one. The relative RhoA and RhoC mRNA transcriptions were decreased to 40% and 36% (P < 0.05), while proteins expression reducing 42% and 35%, respectively (P < 0.05). Growth curves analysis showed that alive cell number in the Ad-A1+A2+C1+C2 group was lower than others in the third to sixth day and transwell chamber analysis showed that migration/invasion activity was significantly suppressed in Ad-A1+A2+C1+C2 group.</p> <p>Conclusion</p> <p>Our results indicate recombinant adenovirus carrying RhoA and RhoC shRNAs in tandem expression may inhibit the growth and invasion of HCT116 cells. Application of such vector to inhibit one or more genes may be a new method to cancer therapy.</p

    Abnormal magnetoresistance behavior is found in superconducting Nb films perforated with rectangular arrays of antidots (holes). Generally magnetoresistance were always found to increase with increasing magnetic field. Here we observed a reversal of this behavior for particular in low temperature or current density. This phenomenon is due to a strong 'caging effect' which interstitial vortices are strongly trapped among pinned multivortices.Comment: 4 pages, 2 figure

    Uplift, Climate and Biotic Changes at the Eocene-Oligocene Transition in Southeast Tibet

    The uplift history of southeastern Tibet is crucial to understanding processes driving the tectonic evolution of the Tibetan Plateau and surrounding areas. Underpinning existing palaeoaltimetric studies has been regional mapping based in large part on biostratigraphy that assumes a Neogene modernisation of the highly diverse, but threatened, Asian biota. Here, with new radiometric dating and newly-collected plant fossil archives, we quantify the surface height of part of Tibet’s southeastern margin of Tibet in the latest Eocene (~34 Ma) to be ~3 km and rising, possibly attaining its present elevation (3.9 km) in the early Oligocene. We also find that the Eocene-Oligocene transition in southeastern Tibet witnessed leaf size diminution and a floral composition change from sub-tropical/warm temperate to cool temperate, likely reflective of both uplift and secular climate change, and that by the latest Eocene floral modernization on Tibet had already taken place implying modernization was deeply-rooted in the Paleogene
