    An analysis of difficulties faced in job duties by academic digital library professionals working in university libraries in Delhi (India)

    The present study aims is to analyze the difficulties faced in performing the job duties by academic digital library professionals working in university libraries of Delhi (India). The survey method was adopted in the present study. The study is based on a questionnaire containing 34 job duties performed in academic digital libraries, classified into four categories, i.e. IT system management, information organization, user services, and collection management. The respondents were 146 library professionals working in university libraries in Delhi (India). The study found that professionals rated the difficulty of job duties such as network management and operation; hardware installation & upgradation; software selection and license management; library website development; database management; troubleshooting; accessing open-source software; working on content management softwares; handling digital preservation practices; initiating and managing digitization projects; providing access to eResources; maintaining an e-learning system; creating an institutional repository and budget planning and allocation as \u27high.\u27 \u27Very high\u27 difficulty rated job duties were handling library back-end servers, system analysis, and design and working on e-resource management software. The study may help library schools review the curriculum according to the difficulties faced by academic library professionals in the job market. In addition, findings will be helpful to review the curriculum updates which should be incorporated in library schools of India

    The occurrence and geographic distribution of the genus Longidorus was investigated during a nematological survey of Bohemia and South Moravia between 2003 and 2006. Soil samples were collected from 139 different fruit orchards and vineyards. Three Longidorus species (L. elongatus, L. euonymus and L. leptocephalus) were recorded. L. euonymus constitutes a new record for the Czech nematofauna

    Regions within the mitochondrial gene encoding for the nicotinamide dehydrogenase subunit 4 (nad4) were characterized to evaluate the extent of genetic variation within and among Xiphinema diversicaudatum and Xiphinema simile populations. Four different sequence variants of nad4 were determined among eight populations of X. diversicaudatum and three variants among three populations of X. simile. Nucleotide variation was detected in 28 of 411 bp (2.43 to 4.87 %) in X. diversicaudatum and in three of 395 bp (0.25 to 0.76 %) in X. simile. This represents the first study based on the characterization of the nad4 gene for the analysis of population genetic of two Xiphinema species. © 2013 KNPV

    Using ribosomal (18S, ITS1, ITS2, D2-D3 expansion segments of 28S rDNA) and mitochondrial (partial cox1 and nad4) DNA markers in a study of several populations of Xiphinema americanum-group from Europe and Morocco, two cryptic species X. browni sp. n. (formerly reported as X. pachtaicum) and X. penevi sp. n. were revealed. The species are described, illustrated and their phylogenetic relationships discussed. The first species is most similar to X. parasimile and is a member of X. simile species complex. The phylogenetic reconstructions inferred from three molecular markers (18S, D2-D3 28S rDNA and cox1) showed that X. penevi sp. n. is part of X. pachtaicum-subgroup and is closely related to X. incertum, X. pachtaicum, X. parapachydermum, X. plesiopachtaicum, X. astaregiense and X. pachydermum. Also, a separate “X. simile-subgroup”, outside the X. pachtaicum-subgroup and so far consisting only of the parthenogenetic species X. simile, X. parasimile, X. browni sp. n. and probably X. vallense was formed. New primers for amplification and sequencing of part of the nad4 mitochondrial gene were designed and used

    Populations of Xiphinema brevicollum occurring in the Czech Republic were described morphologically and molecularly. Published species-specific primer set BL18 and BV3 was used to amplify three populations of X. brevicollum from the Czech Republic. These primers were tested against 9 species of Xiphinema and 11 species of Longidorus. Amplification was also observed for X. inaequale, X. italiae and X. lambertii. Three additional markers, mitochondrial cytochrome c oxidase subunit 1, ribosomal D2/D3 expansion segment of the 28S gene and 18S gene, were amplified and sequenced for X. brevicollum, X. inaequale and X. lambertii belonging to X. americanum-group. Comparison of cox1 sequences of X. brevicollum from the Czech Republic with X. taylori from the Slovak Republic (accession number AM086702) suggested that these populations represent the same species

    The plant-parasitic nematode Longidorus poessneckensis from the Czech Republic was morphologically and molecularly characterised. Molecular analyses were carried out using mitochondrial DNA (cytochrome c oxidase subunit 1-cox1) and ribosomal DNA (ITS2-second internal transcribed spacer, 18S gene and D2/D3 expansion segments of the 28S gene), which were amplified and sequenced. Phylogenetic relationship of L. poessneckensis with three morphologically closely related species, i.e. L. macrosoma, L. helveticus and L. uroshis, was inferred by using maximum likelihood and maximum parsimony methods, with a female of Xiphinema diversicaudatum and a bivulval female of X. vuittenezi as outgroups. All multiple alignments yielded similar basic trees supporting the uniqueness of L. poessneckensis and the validity of the four Longidorus species identified using morphological characters. Phylogenetic analyses revealed that L. poessneckensis is more closely related to L. macrosoma and L. helveticus than to L. uroshis. High inter-population diversity (19%) was observed across the cox1 gene between two populations of L. poessneckensis

    Genetic analyses using sequences of partial cytochrome c oxidase subunit 1 (cox1) gene of mitochondrial DNA were conducted to determine the extent of genetic variation within and among Xiphinema diversicaudatum, X. pachtaicum, X. simile and X. vuittenezi populations. Pairwise distance among the four species was 22.5 to 31.2%. Four different sequence variants of cox1 were determined among six populations of X. diversicaudatum and three variants among three populations of X. simile. Nucleotide variation was detected at 18 of 414 bp (1.9 to 2.7%) in X. diversicaudatum and 4 of 435 bp (0.2 to 0.9%) in X. simile. All changes were at silent sites. No nucleotide variation was detected within three populations of X. pachtaicum and within three populations of X. vuittenezi
