    The stirling engine for vehicle propulsion

    The performance data of experimental Stirling engines are considered along with questions of exhaust-gas composition, engine noise, engine volume and weight, engine control, and the engine-starting process. The Stirling engine can use practically any liquid or gaseous fuel for its operation. It is found that technically a use of the Stirling engine in motor vehicles is feasible. Economic questions related to an introduction of the Stirling engine are discussed along with possible new developments which could improve the economic situation in favor of a use of Stirling engine

    A Test of the Particle Paradigm in N-Body Simulations

    We present results of tests of the evolution of small ``fluid elements'' in cosmological N--body simulations, to examine the validity of their treatment as particles. We find that even very small elements typically collapse along one axis while expanding along another, often to twice or more their initial comoving diameter.     The Future of Affirmative Action: The Legal Imperative Nationally and the Ohio Experience

    This presentation of the legal future of affirmative action will be divided into five sections. The introductory material serves as a general introduction to the issues. The second section will review the origins and evolution of affirmative action. This section will also attempt to provide a definition of this complex concept. The third will provide a detailed analysis of Regents of the University of California v. Bakke. Bakke represents the Supreme Court\u27s first attempt to resolve the legal complexities of affirmative action. The Court\u27s holdings in Bakke have shaped the debate for over 20 years. The fourth segment of this chapter will review current affirmative action case law. Close attention will be paid to emerging legal distinctions essential to an accurate application of affirmative action principles in the future. Within this section, close attention will be paid to current controversies such as California Proposition 209 (the state constitutional amendment which outlaws the use of affirmative action), Hopwood v. State of Texas (nonminority students who challenged a law school\u27s affirmative action admissions program), and Taxman v. Board of Education of the Township of Piscataway (a teacher\u27s challenge to a school board\u27s affirmative action plan of retaining minority teachers over nonminority teachers in regard to layoff decisions). The last section will explain the Ohio Experience with affirmative action

    A comparison of successful and failed protein interface designs highlights the challenges of designing buried hydrogen bonds

    The accurate design of new protein–protein interactions is a longstanding goal of computational protein design. However, most computationally designed interfaces fail to form experimentally. This investigation compares five previously described successful de novo interface designs with 158 failures. Both sets of proteins were designed with the molecular modeling program Rosetta. Designs were considered a success if a high-resolution crystal structure of the complex closely matched the design model and the equilibrium dissociation constant for binding was less than 10 μM. The successes and failures represent a wide variety of interface types and design goals including heterodimers, homodimers, peptide-protein interactions, one-sided designs (i.e., where only one of the proteins was mutated) and two-sided designs. The most striking feature of the successful designs is that they have fewer polar atoms at their interfaces than many of the failed designs. Designs that attempted to create extensive sets of interface-spanning hydrogen bonds resulted in no detectable binding. In contrast, polar atoms make up more than 40% of the interface area of many natural dimers, and native interfaces often contain extensive hydrogen bonding networks. These results suggest that Rosetta may not be accurately balancing hydrogen bonding and electrostatic energies against desolvation penalties and that design processes may not include sufficient sampling to identify side chains in preordered conformations that can fully satisfy the hydrogen bonding potential of the interface

    The Genesis mission collected solar wind and brought it back to Earth in order to provide precise knowledge of solar isotopic and elemental compositions. The ions in the solar wind were stopped in the collectors at depths on the order of 10 to a few hundred nanometers. This shallow implantation layer is critical for scientific analysis of the composition of the solar wind and must be preserved throughout sample handling, cleaning, processing, distribution, preparation and analysis. We are working interactively with the community of scientists analyzing Genesis samples, using our unique laboratory facilities -- and, where needed, our unique cleaning techniques -- to significantly enhance the science return from the Genesis mission. This work is motivated by the need to understand the submicron contamination on the collectors in the Genesis payload as recovered from the crash site in the Utah desert, and -- perhaps more importantly -- how to remove it. That is, we are evaluating the effectiveness of the wet-chemical "cleaning" steps used by various investigators, to enable them to design improved methods of stripping terrestrial contamination from surfaces while still leaving the solar-wind signal intact

    Influence of Towing Force Magnitude on the Kinematics of Supramaximal Sprinting

    The purpose of this study was to determine the influence of towing force magnitude on the kinematics of supramaximal sprinting. Ten high school and collegiate aged track and field athletes ran 60m maximal sprints under 5 different conditions: non-towed (NT), Tow A (2.0% body weight), Tow B (2.8%BW), Tow C (3.8%BW), and Tow D (4.7%BW). Three-dimensional kinematics of a 4-segment model of the right side of the body were collected starting at the 35m point of the trial. Significant differences were observed in stride length (SL) and horizontal velocity of the center of mass (VH) during Tow C and Tow D. For Tow D, a significant increase in the distance from the center of mass to the foot at touchdown (DH) was also observed. Contact time (CT) decreased significantly in all towing conditions, while stride rate (SR) increased slightly (\u3c 2.0%) under towed conditions. There were no significant changes in joint or segment angles at touchdown, with the exception of a significant decrease in the flexion/extension angle at the hip during the Tow D condition. We concluded that towing force magnitude does influence the kinematics of supramaximal running. Furthermore, we suggest that coaches and practitioners adjust towing force magnitude for each individual and avoid using towing forces in excess of 3.8%BW

Particles of Genesis wafers, brine from the Utah Testing Range and an organic film have deleterious effects on many of the high-resolution instruments that have been developed to analyze the implanted solar wind. We have conducted a correlative microscopic study of the efficacy of cleaning Genesis samples with megasonically activated ultrapure water and UV/ozone cleaning.     Mechanism of selective recruitment of RNA polymerases II and III to snRNA gene promoters

    RNA polymerase II (Pol II) small nuclear RNA (snRNA) promoters and type 3 Pol III promoters have highly similar structures; both contain an interchangeable enhancer and "proximal sequence element" (PSE), which recruits the SNAP complex (SNAPc). The main distinguishing feature is the presence, in the type 3 promoters only, of a TATA box, which determines Pol III specificity. To understand the mechanism by which the absence or presence of a TATA box results in specific Pol recruitment, we examined how SNAPc and general transcription factors required for Pol II or Pol III transcription of SNAPc-dependent genes (i.e., TATA-box-binding protein [TBP], TFIIB, and TFIIA for Pol II transcription and TBP and BRF2 for Pol III transcription) assemble to ensure specific Pol recruitment. TFIIB and BRF2 could each, in a mutually exclusive fashion, be recruited to SNAPc. In contrast, TBP-TFIIB and TBP-BRF2 complexes were not recruited unless a TATA box was present, which allowed selective and efficient recruitment of the TBP-BRF2 complex. Thus, TBP both prevented BRF2 recruitment to Pol II promoters and enhanced BRF2 recruitment to Pol III promoters. On Pol II promoters, TBP recruitment was separate from TFIIB recruitment and enhanced by TFIIA. Our results provide a model for specific Pol recruitment at SNAPc-dependent promoters
