    Bobolink (Dolichonyx oryzivorus) Declines Follow Bison (Bison bison) Reintroduction on Private Conservation Grasslands

    Simple Summary North American grassland birds evolved with American bison (Bison bison), until overhunting drove bison to near-extinction > 150 years ago. Bison have now been reintroduced to many areas that provide important nesting habitat for grassland birds, which are now among the most rapidly declining birds in North America. However, little is known about bison interactions with birds such as Bobolinks (Dolichonyx oryzivorus), obligate grassland nesting songbirds of conservation concern. Using data collected over an 18-year period, we assessed the effects of bison reintroduction, together with other land management and climate factors, on Bobolinks in a private conservation area comprising 24 km(2) of native grasslands in the North American Great Plains. In grasslands where bison were reintroduced, Bobolink abundance (adult numbers) declined by 62%, and productivity (juvenile numbers) declined by 84%. By contrast, Bobolink populations remained stable over the same time period in adjacent grasslands where bison were not reintroduced. Bobolink abundance and productivity increased in years following warmer and wetter winters, but nevertheless declined over time in grasslands where the bison population doubled. Where bison are reintroduced and confined in high densities, overgrazing, trampling, and related impacts may drive severe declines in Bobolinks and other grassland birds of conservation concern. Among the most rapidly declining birds in continental North America, grassland birds evolved with American bison (Bison bison) until bison nearly became extinct due to overhunting. Bison populations have subsequently rebounded due to reintroductions on conservation lands, but the impacts of bison on grassland nesting birds remain largely unknown. We investigated how bison reintroduction, together with other land management and climate factors, affected breeding populations of a grassland bird species of conservation concern, the Bobolink (Dolichonyx oryzivorus). We quantified population changes in Bobolinks over an 18-year period in conservation grasslands where bison were reintroduced, compared with adjacent grasslands grazed by cattle and where hay was harvested after the bird breeding season. Four years after bison reintroduction, the bison population in the study area had doubled, while Bobolink abundance declined 62% and productivity declined 84%. Our findings suggest that bison reintroduction as a conservation strategy may be counterproductive in grassland fragments where overgrazing, trampling, and other negative impacts drive declines in grassland breeding birds. Where bird conservation is an objective, small grassland reserves may therefore be inappropriate sites for bison reintroduction. To maximize conservation benefits to birds, land managers should prioritize protecting grassland birds from disturbance during the bird breeding season

    Bond Characteristics of ASTM A1035 Steel Reinforcing Bars

    The results of a coordinated research program on the bond characteristics of the high-strength steel reinforcing bars that conform to ASTM A1035 are presented. Concrete with nominal strengths of 5000 and 8000 psi (35 and 55 MPa) were used. Sixtynine large-scale beam-splice specimens were tested. Maximum bar stresses are compared with predictions obtained using the bond equations in the ACI 318-05 code provisions and those proposed by ACI Committee 408. Maximum stress levels of 120, 110, and 96 ksi (830, 760, and 660 MPa) were developed in No. 5, No. 8, and No. 11 (No. 16, No. 25, and No. 36) bars, respectively, not confined by transverse reinforcement. Providing confinement for No. 8 and No. 11 (No. 25 and No. 36) spliced bars using transverse reinforcement allowed stresses of up to 150 ksi (1035 MPa) to be developed. The ACI Committee 408 equation provides a reasonable estimate of the strength for both unconfined and confined splices using a strength reduction factor (f-factor) of 0.82 and design parameters (cover, spacing, and concrete strengths) comparable to those used in this test program. The design equations in ACI 318 are less conservative, with a large percentage of the developed/calculated strength ratios below 1.0, and should not be used for development and splice design with high-strength reinforcing steel in their present form

    Dopamine Neuron Stimulating Actions of a GDNF Propeptide

    BACKGROUND: Neurotrophic factors, such as glial cell line-derived neurotrophic factor (GDNF), have shown great promise for protection and restoration of damaged or dying dopamine neurons in animal models and in some Parkinson's disease (PD) clinical trials. However, the delivery of neurotrophic factors to the brain is difficult due to their large size and poor bio-distribution. In addition, developing more efficacious trophic factors is hampered by the difficulty of synthesis and structural modification. Small molecules with neurotrophic actions that are easy to synthesize and modify to improve bioavailability are needed. METHODS AND FINDINGS: Here we present the neurobiological actions of dopamine neuron stimulating peptide-11 (DNSP-11), an 11-mer peptide from the proGDNF domain. In vitro, DNSP-11 supports the survival of fetal mesencephalic neurons, increasing both the number of surviving cells and neuritic outgrowth. In MN9D cells, DNSP-11 protects against dopaminergic neurotoxin 6-hydroxydopamine (6-OHDA)-induced cell death, significantly decreasing TUNEL-positive cells and levels of caspase-3 activity. In vivo, a single injection of DNSP-11 into the normal adult rat substantia nigra is taken up rapidly into neurons and increases resting levels of dopamine and its metabolites for up to 28 days. Of particular note, DNSP-11 significantly improves apomorphine-induced rotational behavior, and increases dopamine and dopamine metabolite tissue levels in the substantia nigra in a rat model of PD. Unlike GDNF, DNSP-11 was found to block staurosporine- and gramicidin-induced cytotoxicity in nutrient-deprived dopaminergic B65 cells, and its neuroprotective effects included preventing the release of cytochrome c from mitochondria. CONCLUSIONS: Collectively, these data support that DNSP-11 exhibits potent neurotrophic actions analogous to GDNF, making it a viable candidate for a PD therapeutic. However, it likely signals through pathways that do not directly involve the GFRalpha1 receptor

    A 15-year consolidated overview of data in over 6000 patients from the Transthyretin Amyloidosis Outcomes Survey (THAOS)

    Background Transthyretin amyloidosis (ATTR amyloidosis) is a progressive, multisystemic, life-threatening disease resulting from the deposition of variant or wild-type (ATTRwt amyloidosis) transthyretin amyloid fibrils in various tissues and organs.Methods Established in 2007, the Transthyretin Amyloidosis Outcomes Survey (THAOS) is the largest ongoing, global, longitudinal, observational study of patients with ATTR amyloidosis, including both hereditary and wild-type disease, and asymptomatic carriers of pathogenic TTR mutations. This analysis describes the baseline characteristics of symptomatic patients and asymptomatic gene carriers enrolled in THAOS since its inception in 2007 (data cutoff: August 1, 2022), providing a consolidated overview of 15-year data from the THAOS registry.Results This analysis included 4428 symptomatic patients and 1707 asymptomatic gene carriers. The majority of symptomatic patients were male (70.8%) with a mean (standard deviation [SD]) age at symptom onset of 56.6 (17.9) years. Compared with the 14-year analysis, V30M remained the most prevalent genotype in Europe (62.2%), South America (78.6%), and Japan (74.2%) and ATTRwt remained most common in North America (56.2%). Relative to the 14-year analysis, there was an increase of mixed phenotype (from 16.6 to 24.5%) and a reduction of predominantly cardiac phenotype (from 40.7 to 31.9%). The proportion of patients with predominantly neurologic phenotype remained stable (from 40.1 to 38.7%). Asymptomatic gene carriers were 58.5% female with a mean age at enrollment of 41.9 years (SD 15.5).Conclusions This overview of > 6000 patients enrolled over 15 years in THAOS represents the largest registry analysis of ATTR amyloidosis to date and continues to emphasize the genotypic and phenotypic heterogeneity of the disease. Nearly a quarter of the symptomatic population within THAOS was mixed phenotype, underscoring the need for multidisciplinary management of ATTR amyloidosis.Trial registrationClinicalTrials.gov Identifier: NCT00628745

    Meeting Report: Aging Research and Drug Discovery

    Aging is the single largest risk factor for most chronic diseases, and thus possesses large socioeconomic interest to continuously aging societies. Consequently, the field of aging research is expanding alongside a growing focus from the industry and investors in aging research. This year's 8th Annual Aging Research and Drug Discovery ARDD) meeting was organized as a hybrid meeting from August 30th to September 3rd 2021 with more than 130 attendees participating on-site at the Ceremonial Hall at University of Copenhagen, Denmark, and 1800 engaging online. The conference comprised of presentations from 75 speakers focusing on new research in topics including mechanisms of aging and how these can be modulated as well as the use of AI and new standards of practices within aging research. This year, a longevity workshop was included to build stronger connections with the clinical community
